E-mail: davetpat@ufrgs.br Bovine viral diarrhea virus (BVDV) is responsible for different syndromes that affect cattle worldwide causing important economic losses. This study analyzed the clinical, pathological, immunohistochemical and viral aspects of persistent infection by BVDV in five animals of a farm located in the county of Viamão, Rio Grande do Sul, southern Brazil. The clinical signs included growth impairment, nasal and ocular discharge and, in two animals, congenital cataract. The main gross lesions observed at the necropsy were enlargement of mesenteric lymph nodes and Peyer’s patches, and in one case, pododermatitis and crusted lesions on nasal planum and periocular region. Microscopic findings were characterized mostly by mononuclear infiltrate in the lamina propria, primarily in the small intestine and lymphoid depletion with histiocytic infiltrate in follicular centers of lymph nodes and Peyer’s patches. Viral antigens were more frequently demonstrated in epidermal keratinocytes, epithelium of hair follicles and dendritic cells of the dermis of the ears and skin, histiocytes and lymphocytes in lymph nodes, thyroid follicular cells, in the cytoplasm of neurons and to a lesser extent, in glial cells in the cerebral cortex and hippocampus. Viral isolation from blood samples and organs confirmed the presence of non-cytopathic BVDV. Moreover, viral RNA was detected by RT-PCR in serum samples. Phylogenetic analysis of a partial fragment of the5’ non-translated region of the viral genome allowed the classification of the sample as BVDV type 2b. The present study strengthens the need to investigate and to characterize BVD outbreaks and to describe its different clinic-pathological presentations.

O presente trabalho analisou as características clínicas, patológicas e imuno-histoquímicas e virais de cinco bovinos persistentemente infectados pelo BVDV de uma mesma propriedade, localizada no Município de Viamão, Rio Grande do Sul. Dentre os sinais clínicos verificados destacaram-se subdesenvolvimento, secreções nasais e oculares, além de catarata congênita unilateral em dois bovinos. As principais lesões observadas durante a necropsia consistiram de aumento dos linfonodos mesentéricos, evidenciação das placas de Peyer e pododermatite e lesões crostosas no plano nasal e na região periocular em um animal. Os achados microscópicos caracterizavam-se, principalmente, por infiltrado mononuclear na lâmina do intestino delgado e rarefação linfoide com infiltrado histiocitário nos centrofoliculares de linfonodos e nas placas de Peyer. Antígenos virais foram detectados por imuno-histoquímica principalmente em queratinócitos da epiderme, no epitélio de folículos pilosos e células mononucleares da derme de orelhas e pele; histiócitos e em linfócitos dos linfonodos; células foliculares da tireoide; no citoplasma de neurônios e, em menor escala, em células da micróglia no córtex cerebral e no hipocampo. O isolamento viral de amostras de sangue e órgãos dos animais confirmou a presença de BVDV não citopático. Também foi possível detectar a presença do genoma viral por RT-PCR no soro dos animais. A análise filogenética do fragmento parcial da região 5’ não traduzida do genoma viral permitiu a classificação da amostra viral como BVDV tipo 2b. O presente estudo reforça a necessidade de investigar e caracterizar surtos de BVD e descrever suas diferentes formas de apresentação.

Bento Gonçalves 9090, Porto Alegre, RS 91540-000, Brazil. E-mail: davetpat@ufrgs.br Bluetongue (BT) is a disease caused by bluetongue virus (BTV) and transmitted by vectors of the genus Culicoides. Serological studies have demonstrated the widespread presence of the virus in Brazil, however, clinical information of BT in South America are limited. This article describes clinical and pathological changes observed in sheep naturally infected by BTV in southern Brazil. In two outbreaks on different farms, 15 sheep showed clinical signs such as severe hyperthermia, apathy, swelling of the face and submandibular area, difficulty in swallowing with regurgitation, greenish mucopurulent nasal secretion, severe weight loss, and erosions in the oral mucosa. Necropsy findings in six sheep included subcutaneous edema of the face and ventral region of the chest, greenish nasal discharge, and dilated esophagus filled with abundant food contents, collapsed lungs with areas of anteroventral consolidation, and trachea and bronchi filled by foamy material mixed with food. In the heart and base of the pulmonary artery there were foci of hemorrhage. Histologically, the main changes were in cardiac and skeletal muscles and consisted of biphasic lesions characterized by hyaline and floccular degeneration/necrosis of myofibers associated with micro-mineralization and mononuclear cell infiltration. Pneumonia associated with the presence of organic matter and bacteria in the lumen of the bronchi was also observed. The diagnosis of BT was confirmed by detection of the viral genome by duplex RT-PCR in blood of affected animals, followed by the identification of BTV, serotype 12 by nucleotide sequencing.

Estudos sorológicos têm demonstrado a ampla presença do vírus no Brasil; entretanto, informações clínicas da LA na América do Sul são limitadas. Esse trabalho descreve alterações clínico-patológicas em ovinos acometidos pela LA no Sul do Brasil. Em dois surtos, em propriedades distintas, 15 ovinos apresentaram como principais sinais clínicos hipertermia, apatia, aumento de volume da face e região submandibular, dificuldade de deglutição com regurgitação, secreção nasal mucopurulenta esverdeada, alterações respiratórias, além de acentuada perda de peso e erosões na mucosa oral. Os achados de necropsia em seis ovinos afetados incluíram edema subcutâneo na face e região ventral do tórax, secreção nasal esverdeada, esôfago dilatado preenchido por grande quantidade de conteúdo alimentar, pulmões não colabados com áreas consolidadas anteroventrais, bem como luz da traquéia e brônquios preenchida por espuma misturada com conteúdo alimentar. No coração e base da artéria pulmonar, havia focos de hemorragia. Histologicamente, as principais alterações observadas ocorriam no tecido muscular cardíaco e esquelético, especialmente no esôfago e consistiam de lesões bifásicas caracterizadas por degeneração/necrose hialina e flocular de miofibras associadas com micro-calcificação e infiltrado inflamatório mononuclear. Pneumonia aspirativa associada à presença de material vegetal e bactérias na luz de brônquios também foi observada. O diagnóstico de LA foi confirmado pela detecção do genoma viral por duplex RT-PCR em amostras de sangue de animais afetados, seguido da identificação do VLA, sorotipo 12 por sequenciamento.

E-mail: davetpat@ufrgs.br Two cases of acute necrotizing myositis caused by Clostridium septicum in horses are described. Both horses presented swelling of the right pelvic limb extending to the ventral abdominal region. The cut surface of the affected area revealed blood-stained edema and gas bubbles. The skeletal muscles of the caudal region of the thigh of the affect limbs had dark red discolored areas of blood-stained edema and crepitation; the deep musculature was dry. The main histopathological findings were swelling, vacuolation and hyaline and floccular necrosis of skeletal myofibers; in between myofibers there were hemorrhage, edema and large amounts of bacilli. In both cases, C. septicum was isolated from the edema fluid of muscular lesions.

E-mail: davetpat@ufrgs.br Descrevem-se dois casos de miosite necrosante causada por Clostridium septicum em equinos. Os dois equinos apresentavam aumento de volume no membro pélvico direito e que se estendia para a região abdominal ventral. Ao corte, essa área era formada por edema sanguinolento e bolhas de gás. Os músculos esqueléticos da região caudal da coxa apresentavam áreas vermelho-escuras, crepitantes, com edema sanguinolento e, ao corte, as áreas mais profundas da musculatura tinham aspecto seco. As principais alterações histopatológicas observadas foram tumefação, vacuolização, necrose hialina e necrose flocular de fibras musculares esqueléticas. Entre as fibras, havia hemorragia, edema e grande quantidade de bacilos com tamanho de 3-6µm. Na coloração de Gram, os bacilos se apresentavam roxos (gram-positivos); quando impregnados pela prata (Warthin-Starry), se mostraram enegrecidos. Nos dois casos, C. septicum foi isolado, em ambiente anaeróbio, do líquido de edema das lesões musculares.

E-mail: davetpat@ufrgs.br Fulguração é a morte de animais pela ação de correntes elétricas atmosféricas ou raios durante tempestades. Quatro bovinos provenientes de dois rebanhos foram encontrados mortos nas proximidades de árvores de Eucalyptus sp. imediatamente após registros de tempestades no Estado do Rio Grande do Sul. Nos locais haviam árvores com linhas de queimadura recente no tronco, cascas parcialmente soltas, além de restos de galhos e folhas espalhados sob as copas das árvores. Não foram encontradas lesões macroscópicas e microscópicas significativas, nem indícios de enfermidades tóxicas ou infecciosas. Tais achados sugerem morte súbita por fulguração nos bovinos.

E-mail: davetpat@ufrgs.br Lightning strike may cause sporadic deaths of animals that have been exposed to high-voltage electric currents during thunderstorms. Four healthy adult cattle from two herds were found dead next to eucalypt trees immediately after storms in Rio Grande do Sul, Brazil. There were recent burns and loose barks on the trunks of the trees, apart numerous branches and leaves scattered on the ground under the canopy of the trees. No gross or microscopic lesions were observed. In addition, there was no evidence of any toxic or infectious disease. These findings suggest that lightning strike caused the death of those animals.

Most of them were from southern Brazil. Ten cases (3.25%) of congenital anomalies were seen. The most frequent congenital anomalies were artrogryposis, Amorphous globosus, and cleft palate (palatoschisis). Infectious causes were investigated, but only BVDV infection was detected by immunohistochemistry in one case, which was affected with porencephalia.

Microscopic lesions included non-suppurative myocarditis, myocardial necrosis and fibrosis, mineralization foci and intranuclear inclusion bodies in cardiomyocytes, and interstitial mononuclear pneumonia. Samples from 7 (5.78 per cent) of 121 aborted fetuses and stillborn piglets had lesions consistent with a viral cause and showed both positive anti-PCV2 immunostaining as well as PCV2-PCR. In samples from 3 (2.47 per cent) of these 7 fetuses, co-infection with PPV was confirmed by Nested-PCR. Both viruses were detected in fetuses at different stages of gestation.
