#11 - Occurrence of anti-Toxoplasma gondii antibodies in cats (Felis catus) in the municipality of Maceió, Alagoas

Abstract in English:

Toxoplasma gondii is an intracellular protozoan parasite responsible for toxoplasmosis, with felids serving as its definitive hosts. This neglected zoonotic disease affects both humans and animals worldwide. This study aimed to perform a serological survey and spatial analysis of the occurrence of anti-T. gondii IgG antibodies in cats from the municipality of Maceió, Alagoas. Overall, blood serum samples from 337 adult cats, collected from a private laboratory (137 samples) and the Zoonosis Surveillance Unit (200 samples), were tested using the indirect immunofluorescence test to detect IgG antibodies, with a cutoff dilution of 1:16. Samples with peripheral tachyzoite fluorescence were considered positive. Spatial distribution maps of Maceió’s neighborhoods and Health Districts were designed, and additional factors such as age, sex, and breed were analyzed. Serological analysis revealed that 33.53% (113/337) of the samples were seropositive, with 57.52% (65/113) originating from the private laboratory. The neighborhoods with the highest positivity rates were Ponta Verde, with 14.15% (16/113), and Benedito Bentes, with 11.50% (13/113). Health District I, a coastal and affluent area, had the highest positivity rate at 31.86% (36/113). Age analysis indicated that 60.20% (68/113) of seropositive cats were older than two years, with males comprising 55.75% of the positive cases. Most seroreactive cats were of undefined breed, with 91.15% (103/113). This study seems to be the first serological survey of T. gondii in cats from Maceió, and its findings are consistent with those from other states in northeastern Brazil. Furthermore, it highlights the need to acknowledge the relationship between the occurrence of infection and social vulnerability.

Toxoplasma gondii é um protozoário intracelular responsável pela toxoplasmose, tendo os felídeos como seus hospedeiros definitivos. Essa doença zoonótica negligenciada afeta tanto humanos quanto animais em todo o mundo. O objetivo deste estudo foi realizar um levantamento sorológico e uma análise espacial da ocorrência de anticorpos anti-T. gondii IgG em gatos do município de Maceió, Alagoas. No total, 337 amostras de soro sanguíneo de gatos adultos, coletadas de um laboratório privado (137 amostras) e da Unidade de Vigilância de Zoonoses (200 amostras), foram testadas utilizando o teste de imuno fluorescência indireta para detectar anticorpos IgG, com uma diluição de corte de 1:16. Amostras com fluorescência periférica de taquizoítos foram consideradas positivas. Mapas de distribuição espacial dos bairros e distritos de saúde de Maceió foram elaborados, e fatores adicionais, como idade, sexo e raça, foram analisados. A análise sorológica revelou que 33,53% (113/337) das amostras foram soropositivas, sendo 57,52% (65/113) provenientes de laboratório privado. Os bairros com as maiores taxas de positividade foram Ponta Verde (14,15%; 16/113) e Benedito Bentes (11,50%; 13/113). O Distrito de Saúde I, uma área litorânea e de alto poder aquisitivo, apresentou a maior taxa de positividade, com 31,86% (36/113). A análise etária indicou que 60,20% (68/113) dos gatos soropositivos tinham mais de dois anos de idade, sendo que os machos representaram 55,75% dos casos positivos. A maioria dos gatos sororreativos era de raça indefinida (91,15%; 103/113). Este parece ser o primeiro levantamento sorológico de T. gondii em gatos de Maceió, e seus achados são consistentes com os de outros estados do nordeste do Brasil. Além disso, destaca a necessidade de reconhecer a relação entre a ocorrência da infecção e a vulnerabilidade social.


#12 - Epidemiology of toxoplasmosis in felids and canids in Brazil: A brief One Health overview

Abstract in English:

Toxoplasmosis is a disease caused by the obligate intracellular protozoan Toxoplasma gondii, which infects animals and humans worldwide. Felids are definitive hosts that eliminate oocysts, contaminating the environment, food, and water. Among carnivores, cats are the most important hosts for the epidemiology of the disease since a single individual can shed millions of oocysts. Wild canids are considered sentinels and play an essential role in the epidemiology of toxoplasmosis due to tissue cysts in muscle cells, representing a source of infection for carnivores through predation, while environmental contamination is mainly due to oocyst shedding by felids. Free-ranging felids are more likely to be infected by T. gondii than felids living in captivity. The free-living wild canids have lower seropositivity compared to canids in captivity. This review article presents epidemiological data on toxoplasmosis in domestic and wild carnivores in Brazil, targeting professionals in clinical practice, veterinary pathology, diagnostics, and One Health. Therefore, we understand the importance of disseminating diagnoses, epidemiological investigations and animal health programs to control, prophylaxis and treat toxoplasmosis in domestic and wild carnivores.

A toxoplasmose é uma doença causada pelo protozoário intracelular obrigatório Toxoplasma gondii, responsável por infectar animais e seres humanos em todo o mundo. Os felinos são hospedeiros definitivos e eliminam oocistos que contaminam o ambiente, alimentos e água. Dentre os carnívoros, os felinos são os hospedeiros mais importantes para a epidemiologia da doença, visto que um único indivíduo é capaz de excretar milhões de oocistos. Canídeos selvagens são considerados sentinelas e desempenham um papel essencial na epidemiologia da toxoplasmose, pois os cistos teciduais presentes nas células musculares representam uma fonte de infecção para carnívoros por meio da predação, enquanto a contaminação ambiental ocorre principalmente pela eliminação de oocistos por felídeos. Os felinos de vida livre têm maior probabilidade de serem infectados por T. gondii do que os felinos mantidos sob cuidados humanos. Os canídeos silvestres de vida livre apresentam menor soropositividade em comparação aos canídeos em cativeiro. Este artigo de revisão apresenta dados sobre a epidemiologia da toxoplasmose em carnívoros domésticos e silvestres no Brasil focado para profissionais da área clínica, patologia veterinária, diagnóstico e Saúde Única. Portanto entendemos a importância de divulgação de métodos de diagnósticos, investigações epidemiológicas e programas de saúde animal para contribuir com o controle, profilaxia e tratamento da toxoplasmose em carnívoros domésticos e silvestres.


#13 - Clinical, pathological and immunohistochemical characterization of spontaneous neoplasms in pet rodents in Northeastern Brazil

In the last decade, there has been a significant increase in the demand for small rodents such as hamsters, guinea pigs, rats, and mice to be kept as pets. Consequently, the veterinary care provided to these animals has also increased. The aim of this study was to describe gross, histopathological and immunohistochemical findings of 26 spontaneous neoplasms diagnosed in 25 pet rodents in Northeastern Brazil. A retrospective study was carried out from 2014 to 2022 in two veterinary diagnostic laboratories to identify cases of tumoriform lesions in these species. Hamsters represented the most frequent species in this study (60%, 15/25), followed by rats (28%, 7/25), guinea pigs (8%, 2/25), and mice (4%, 1/25), with a mean age of 17.63 months. The anatomical regions of the face/head and thoracic region were the most affected. The most affected tissues were the skin/subcutaneous/mucosal (65%, 15/26) and mammary gland (23%, 6/26). Eighteen different types of neoplasms were diagnosed, and T-cell lymphomas and mammary adenocarcinomas were the most diagnosed tumors, each corresponding to 12% (3/26) of the cases. Leiomyosarcoma, myxosarcoma and mammary adenocarcinoma metastases were also noted. Immunohistochemistry was essential for the diagnosis of certain malignant mesenchymal and round-cell neoplasms. Pet rodent neoplasms in Northeastern Brazil are common, especially in hamsters, and immunohistochemistry can be a useful tool for the definitive diagnosis of these tumors.

#14 - Co-infection by tick-borne pathogens and Leishmania spp. in dogs with clinical signs suggestive of leishmaniasis from an endemic area in northeastern Brazil

Abstract in English:

The present study aimed to investigate the occurrence of Leishmania spp., hemotropic Mycoplasma spp., tick-borne pathogens (TBP), and co-infection in dogs with clinical signs suggestive of visceral leishmaniasis (VL). It also aimed to determine the factors associated with infection and to map the distribution of co-infected dogs in an endemic area in the Northeast region of Brazil. Blood samples from 168 dogs were evaluated for serological analysis to Leishmania spp., Anaplasma spp., Ehrlichia spp., Babesia spp., and molecular assays to Leishmania spp., Anaplasma platys, Ehrlichia canis, Babesia spp., and hemotropic Mycoplasma spp. In serological and molecular analysis, 29.8% and 5.9% of dogs were co-infected. In the regression analysis, seropositivity for Ehrlichia spp., Babesia spp., and Leishmania spp. was significantly associated with the presence of petechiae, young dogs, and weight loss. Serology revealed that co-exposure with Babesia spp. and Ehrlichia spp. was associated with fever and thrombocytopenia, and there was an association between seropositivity for Ehrlichia spp. and Babesia spp. in dogs seropositive for Leishmania spp. The presence of hemotropic Mycoplasma spp. DNA was associated with anorexia. Thus, dogs with clinical VL have co-infection with other pathogens, reinforcing the importance of this study for a better understanding of these co-infections in dogs from endemic areas.

O presente estudo objetivou investigar a prevalência de Leishmania spp., Mycoplasma spp. hemotrópico, patógenos transmitidos por carrapatos (PTC), e coinfecção em cães com sinais clínicos sugestivos de leishmaniose visceral (LV), determinar os fatores associados à infecção, e mapear a distribuição de cães coinfectados em uma área endêmica no Nordeste do Brasil. Amostras de sangue de 168 cães foram avaliadas por análises sorológicas para Leishmania spp., Anaplasma spp., Ehrlichia spp., Babesia spp., e ensaio molecular para Leishmania spp., Anaplasma platys, Ehrlichia canis, Babesia spp., e Mycoplasma hemotrópico. Pelas análises sorológicas e moleculares, 29,8% e 5,9% dos cães apresentaram coinfecção, respectivamente. Na análise de regressão, a soropositividade para Ehrlichia spp., Babesia spp., e Leishmania spp. foram significantemente associadas com a presença de petéquias, cães jovens, e perda de peso. O diagnóstico sorológico revelou que a coexposição à Babesia spp. e Ehrlichia spp. está associada com febre e trombocitopenia, havendo associação entre a soropositividade para Ehrlichia spp. e Babesia spp. em cães soropositivos para Leishmania spp. A presença de DNA de Mycoplasma foi associada à anorexia. Desta forma, cães com sinais de LV possuem coinfecção com outros patógenos, reforçando a importância deste estudo para um melhor entendimento dessas coinfecções em cães de áreas endêmicas.


#15 - Comparison of direct blood smear methods to detect piroplasms in wandering horses from Midwest Brazil

Abstract in English:

Equine piroplasmosis is an apicomplexan hemoprotozoan-caused disease that affects equids worldwide. Horses that survive piroplasmosis can become asymptomatic carriers for the rest of their lives. The present study aimed to determine the frequency of piroplasms and to compare three different blood smear tests (jugular, peripheral, and splenic blood) to detect piroplasms in asymptomatic wandering horses seized by the “Secretaria de Estado da Agricultura, Abastecimento e Desenvolvimento Rural do Distrito Federal” (SEAGRI-DF), Midwest region of Brazil. Of the 100 horses evaluated, 38 were diagnosed positive for piroplasm (38%), with the etiological agents found at the jugular blood smear in 11% (11/100) of horses, peripheral blood smear in 13% (13/100), and splenic blood smear in 38% (38/100). Piroplasm-positive horses showed anemia, neutrophilia, and lymphopenia, but the hematological changes did not differ statistically between positive and negative horses. In summary, the stray horses evaluated showed a high incidence of piroplasm (38%). All positive horses presented one of the etiologic agents in the splenic blood, but some did not show the parasite in the jugular blood smear (27/38, 71%) or the peripheral blood smear (25/38, 65.8%). Thus, the splenic blood was shown to be the best sample to determine the presence of piroplasm in wandering horses. As it is a low-cost and easy-to-perform test, it can be included in the routine diagnosis of equine piroplasmosis, helping to monitor the prevalence of piroplasms in places where molecular techniques are not accessible.

A piroplasmose é uma doença causada por hemoprotozoários apicomplexos que afeta equídeos em todo o mundo. Equinos que sobrevivem à piroplasmose podem se tornar portadores assintomáticos pelo resto de suas vidas. O presente estudo teve como objetivo determinar a frequência de piroplasmas em equinos errantes apreendidos pela Secretaria de Estado da Agricultura, Abastecimento e Desenvolvimento Rural do Distrito Federal (SEAGRI-DF), região Centro-Oeste do Brasil, e comparar três diferentes exames diagnósticos de esfregaços sanguíneos: sangue jugular, periférico e esplênico. Dos 100 equinos avaliados, 38 foram diagnosticados positivos para piroplasmídeos, sendo o agente etiológico encontrado no esfregaço de sangue jugular em 11% (11/100), no esfregaço de sangue periférico em 13% (13/100) e no esfregaço de sangue esplênico em 38% (38/100) dos equinos. Os cavalos positivos apresentaram anemia, neutrofilia e linfopenia, mas as alterações hematológicas não apresentaram diferença estatística entre cavalos positivos e negativos. Em resumo, os equinos errantes avaliados apresentaram alta incidência de piroplasmídeos (38%). Todos os cavalos positivos apresentaram um dos agentes etiológicos no esfregaço sangue esplênico, mas alguns não apresentaram o parasita no esfregaço de sangue jugular (27/38, 71%) ou no esfregaço de sangue periférico (25/38, 65.8%). Assim, o sangue esplênico mostrou-se a melhor amostra para determinar a presença de piroplasmídeos em equinos errantes. Por ser um exame de baixo custo e fácil execução, pode ser incluído na rotina diagnóstica da piroplasmose equina, auxiliando no monitoramento da doença em locais onde as técnicas moleculares não são acessíveis.


#16 - Viability of Toxoplasma gondii in cattle semen cryopreserved with different concentrations of cryoprotectant

Abstract in English:

Toxoplasma gondii can be eliminated in bovine semen. Cryopreserved semen is often used due to the fact that artificial insemination in dairy and beef cattle provides benefits in terms of production. However, little is known regarding the viability and infectivity of T. gondii tachyzoites in cryopreserved bovine semen. In the present study, cattle semen negative for T. gondii were contaminated with 1 x 106 tachyzoites (RH strain) and cryopreserved with and without different cryoprotectants, such as DMSO (concentrations of 2.5%, 5.0%, 7.5%, 8.0% and 10.0%) and glycerol (2.25%, 2.5%, 3.0%, 5.0%, 7.5% and 10.0%), followed by freezing in liquid nitrogen (-196°C). After 24 hours, the samples were thawed and inoculated in 10 mice per cryoprotectant concentration. The mice were evaluated for clinical signs of toxoplasmosis (rough coat, diarrhea, hypoactivity and sudden death) as well as serum titers of IgM and IgG and the presence of tachyzoites in the peritoneal lavage. The results revealed that T. gondii remained infective in all samples. Clinical signs of toxoplasmosis were observed in the mice beginning with the 6th day post-inoculation (DPI) and 100% lethality was found between the 7th and 9th DPI. Viable tachyzoites were recovered from peritoneal exudate of dead mice (except for the control group), with higher mean of tachyzoite counts in the intraperitoneal lavage for 5% DMSO (±3.32 x 106), 8% DMSO (±3.53 x 106), 3% glycerol (±4.75 x 106), 7.5% glycerol (±6.26 x 106) and the absence of cryoprotectant (±3.11 x 106). Seroconversion occurred in the treated groups, with titers of IgG from 1:16 to 1:128 and IgM from 1:16 to 1:512. T. gondii viability and infectivity were maintained in cattle semen during 24 hours of cryopreservation at -196°C with and without cryoprotectant. However, further studies are necessary to determine whether cryopreserved semen contributes to the spread of toxoplasmosis through artificial insemination.

Sabe-se que Toxoplasma gondii pode ser eliminado no sêmen bovino. A inseminação artificial em bovinos leiteiros e de corte proporcionou avanços e benefícios nas produções e para isso o sêmen criopreservado é frequentemente utilizado. No entanto, pouco se sabe sobre a viabilidade e infectividade dos taquizoítos de T. gondii em sêmen bovino criopreservado. Para isso o sêmen bovino, negativo para T. gondii, foi contaminado com 1x106 taquizoítos (cepa RH), criopreservados com ou sem diferentes crioprotetores como DMSO (2.5%, 5.0%, 7.5%, 8.0% e 10.0%) e Glicerol (2.25%, 2.5%, 3.0%, 5.0%, 7.5% e 10.0%) e congelados em nitrogênio líquido (-196°C). Após 24 horas, essas amostras foram descongeladas e inoculadas em 10 camundongos por diluente de concentração de crioprotetor. Os camundongos foram avaliados quanto a sinais clínicos de toxoplasmose (pele áspera, diarreia, hipoatividade e morte súbita), títulos séricos de IgM e IgG e presença de taquizoítos no lavado peritoneal. Os resultados mostraram que T. gondii se manteve infectante em todas as amostras, inclusive naquelas sem crioprotetor. Sinais clínicos de toxoplasmose foram observados nos camundongos a partir do 6º dia pós-inoculação (DPI) e 100% de letalidade foi verificada entre o 7º ao 9º DPI. Nos camundongos mortos, exceto no grupo controle, taquizoítos viáveis foram recuperados do exsudato peritoneal, com maior média de taquizoítos quantificados na lavagem intraperitoneal para DMSO a 5% (±3.32x106), 8% (±3.53x106) e glicerol 3% (±4.75x106), 7,5% (±6.26x106) e livre de crioprotetor (±3.11x106). A soroconversão ocorreu nos grupos tratados com títulos de IgG (1:16 a 1:128) e IgM (1:16 a 1:512). A viabilidade e infectividade do T. gondii no sêmen bovino durante as 24 horas de criopreservação a -196°C foram mantidas com ou sem crioprotetor. No entanto, mais estudos são necessários para verificar se o sêmen criopreservado contribui para a disseminação da toxoplasmose na inseminação artificial.


#17 - Genetic diversity of Mycoplasma hyopneumoniae in finishing pigs in Minas Gerais

Abstract in English:

Mycoplasma hyopneumoniae is one of the most challenging respiratory pathogens involved with swine pneumonia worldwide, responsible for a chronic infection with high morbidity, which predisposes secondary bacterial infections in growing and finishing pigs. Advances in diagnostic techniques allowed identification of genetic characteristics associated with high antigenic and proteomic variability among bacterial strains. This study aimed to evaluate the genetic diversity of M. hyopneumoniae strains in lungs with pneumonic lesions obtained from 52 pig farms located in Minas Gerais, one of the largest swine production states in Brazil. Genotyping was performed using multilocus variable number of tandem repeat (VNTR) analysis (MLVA), targeting two loci encoding P97 and P146 adhesins VNTR. The results showed that this agent is widely disseminated in pig farms and there is a high polymorphism of M. hyopneumoniae variants circulating in the state of Minas Gerais. Different M. hyopneumoniae genotypes are randomly distributed in several regions of the state, with no specific geographic population structure pattern. M. hyopneumoniae association with viral agents was sporadic (3.17% with Influenza A and 1.9% with PCV2).

Mycoplasma hyopneumoniae é um dos patógenos respiratórios mais desafiadores envolvidos com pneumonia suína em todo o mundo. É responsável por uma infecção crônica de alta morbidade, que predispõe a infecções bacterianas secundárias nas fases de crescimento e terminação. Avanços nas técnicas diagnósticas permitiram a identificação de características genéticas do agente, associadas à alta variabilidade antigênica e proteômica entre cepas. Este estudo teve como objetivo examinar a ocorrência e diversidade genética de cepas de M. hyopneumoniae em pulmões com lesões pneumônicas em 52 granjas de suínos no estado de Minas Gerais, um dos maiores estados produtores de suínos do Brasil. A genotipagem foi realizada utilizando a técnica de “multilocus variable number of tandem repeat analysis” (VTNR/MLVA), usando dois loci que codificam VNTR das adesinas P97 e P146. Os resultados mostraram que esse agente está amplamente disseminado em granjas de suínos e que existe alto polimorfismo das variantes de M. hyopneumoniae circulando no estado de Minas Gerais. Diferentes genótipos de M. hyopneumoniae estão distribuídos aleatoriamente em várias regiões do estado, sem um padrão de estrutura populacional e geográfica específico. Associação de M. hyopneumoniae e agentes virais foi esporádica (3,17% com Influenza A e 1,9% com PCV2).


#18 - Lesions of the oral cavity of dogs: 720 cases

Abstract in English:

Seven hundred twenty cases of oral cavity lesions in dogs were diagnosed from 2015 to 2020 in Distrito Federal, Brazil. Four hundred thirty-two (60%) lesions were neoplastic, and 288 (40%) were non-neoplastic. Of the neoplasms, 229 (53%) were considered malignant and 203 (47%) benign, affecting mainly males, of 43 different breeds, with an average age of 9.4 years, mainly the gum and lip. Melanoma was the most (96/432, 22.2%) commonly diagnosed neoplasm, followed by peripheral odontogenic fibroma (68/432, 15.7%), papilloma (63/432, 14.5%) and squamous cell carcinoma (SCC) (56/432, 12.9%). Non-neoplastic lesions also affected males predominantly, from 41 different breeds, with an average age of 8.9 years, mainly in the gums, lip, and tongue. These lesions were diagnosed as inflammatory (230/288, 79.8%), hyperplastic (47/288, 16.3%) and other lesions (11/288, 3.8%).

Foram estudados 720 casos de lesões na cavidade oral em cães diagnosticados entre 2015 e 2020, no Distrito Federal. Dessas alterações, 432 (60%) foram neoplásicas e 288 (40%) não neoplásicas. Dentre os neoplasmas, 229 (53%) foram consideradas malignos e 203 (47%) benignos, acometendo principalmente machos, de 43 raças diferentes, com idade média 9,4 anos, afetando principalmente gengiva e lábio. Melanoma foi o neoplasma mais comumente diagnosticado (96/432; 22,2%), seguido do fibroma odontogênico periférico (68/432; 15,7%), papiloma (63/432; 14,5%) e carcinoma de células escamosas (56/432; 12,9%). As lesões não neoplásicas também acometeram predominantemente machos, de 41 raças diferentes, com idade média 8,9 anos, afetando principalmente gengiva, lábio e língua. Essas lesões foram diagnosticadas como inflamatórias (230/288; 79,8%), hiperplásicas (47/288; 16,3%) e como outras alterações (11/288; 3,8%).


#19 - Cytological grading of canine mast cell tumors: correlation with histologic grading and survival time

Mast cell tumors are one of the most common neoplasia in dogs and cytopathology and/or histopathology examinations are used for diagnosis. Histologic grading is considered the gold standard test to predict the prognosis of this neoplasia. However, studies have been conducted using the cytological grading system to provide similar information in a faster, less invasive, and more accessible way. This study aimed to investigate cytological graduation and correlate it with histological grading and the survival time of dogs diagnosed with cutaneous mast cell tumors at the Veterinary Hospital of “Universidade Federal de Uberlândia” over five years. For that, cytological and histological slides from 72 animals were reviewed. The statistical methods used were the kappa test for agreement between grading systems, the Kaplan–Meier for survival time, Cox regression for comparison of cytological and histological grades and survival time. The cytological grading when compared to the two-tier histologic grading, high and low grades, had a moderate agreement (kappa 0.566). When the correlation between survival time and the cytological grade was evaluated, there was a higher death rate in the group with high-grade mast cell tumors compared to low grade, pointing to a correlation between survival time and cytological grade (p=0.009). In conclusion, the cytological grade is useful to treatment planning and providing prognostic information that precedes tumor removal, showing a good correlation with the two-tier histologic grading and with the survival time of the animals.

O mastocitoma é uma das neoplasias cutâneas mais comum nos cães e os exames citopatológicos e/ou histopatológicos são utilizados para diagnóstico. A graduação histológica é considerada padrão ouro para prever o prognóstico dessa neoplasia. Contudo, estudos têm sido realizados visando utilizar graduação citológica para fornecer informações semelhantes de maneira rápida, menos invasiva e mais acessível. Esse trabalho objetivou realizar graduação citológica e correlacionar com as graduações histológicas e com a sobrevida de cães diagnosticados com mastocitoma cutâneo no Hospital Veterinário da Universidade Federal de Uberlândia durante 5 anos. Para isso, lâminas de citologia e histologia de mastocitomas de 72 animais foram revisadas. Os métodos estatísticos utilizados foram teste kappa para concordância entre os sistemas de graduação, método Kaplan-Meier para tempo de sobrevida dos animais, e análise pela regressão de Cox para comparação do grau citológico e grau histológico e o tempo de sobrevida global. A graduação citológica quando comparada com a histológica de dois níveis, alto grau e baixo grau, obteve uma concordância moderada (kappa 0,566). Na avaliação da correlação entre sobrevida e grau citológico, houve maior taxa de óbito no grupo de cães com mastocitoma de alto grau, comparado aos de baixo grau, sendo observada correlação entre a sobrevida e o grau citológico (p=0,009). Esse estudo concluiu que o grau citológico é útil para o planejamento do tratamento e para fornecer informações prognósticas que antecedem a exérese do tumor, tendo boa correlação com a graduação histológica de dois níveis e com a sobrevida dos animais.


#20 - Genetic diversity of Anaplasma marginale in calves under natural transmission conditions in the Northeast region of Pará

Abstract in English:

The objective of the present study was to detect the genetic diversity of Anaplasma marginale strains in naturally infected calves from a rural property located in the northeastern region of the state of Pará, Eastern Amazon, which has a history of mortality due to anaplasmosis. Fourteen calves positive for A. marginale were selected using a semi-nested polymerase chain reaction for the target msp1α gene, with asymptomatic (n=3) and symptomatic (n=11) infections. After sequencing the samples, two genotypes were verified in the E and C regions and the structures in tandem repeats were determined. Nine different strains were found: eight related to the E genotype (α-β-β-Γ = one animal, asymptomatic; 16-F-17-F-F = two animals, symptomatic; α-β-F-F-F-F = one animal, asymptomatic; 31-62-62-61 = one animal, symptomatic; τ-10-3 = three animals, two symptomatic and one asymptomatic; α-β-β-β = one animal, symptomatic; τ-22 -13-18 = two animals, both symptomatic; β-β-β-BRA1-31 = two animals, both symptomatic), and one related to genotype C (23-24-25-31-27-27 = one animal, asymptomatic). Genotype E was predominant in 92.86% of the samples (13/14), followed by genotype C (7.14%). This study made it possible to detect the genetic diversity of A. marginale in calves from the selected dairy farm, in addition to identifying the BRA1 sequence in the animals of the present study, which was recently diagnosed in Minas Gerais, demonstrating the dispersion of A. marginale strains in herds from different Brazilian states. Genetic diversity of A. marginale was observed in both symptomatic and asymptomatic calves. There were no significant differences when clinical signs were compared to the genotype verified in the infected animals. The prevalence of pathogenicity was not observed.

O objetivo do presente trabalho foi detectar a diversidade genética de cepas de Anaplasma marginale em bezerros naturalmente infectados oriundos de uma propriedade rural localizada na região nordeste do estado do Pará, Amazônia Oriental, a qual apresentava histórico de mortalidade devido à anaplasmose. Foram selecionados 14 bezerros positivos para A. marginale pela técnica de semi-nested PCR (nPCR) para o alvo no gene msp1α, com infecção assintomática (n=3) e sintomáticos (n=11). Após o sequenciamento das amostras foram verificados dois genótipos nas regiões E e C, e determinadas as estruturas em tandem repeats. Nove diferentes estirpes foram encontradas, sendo oito relacionadas ao genótipo E (α-β-β-Γ = um animal, assintomático; 16-F-17-F-F = dois animais, sintomáticos; α-β-F-F-F-F = um animal, assintomático; 31-62-62-61 = um animal, sintomático; τ-10-3 = três animais, dois sintomáticos e um assintomático; α-β-β-β = um animal, sintomático; τ-22-13-18 = dois animais, sintomáticos; β-β-β-BRA1-31 = dois animais, sintomáticos) e uma relacionada ao genótipo C (23-24-25-31-27-27 = um animal, assintomático). O genótipo E foi predominante em 92,86% das amostras (13/14), seguido pelo genótipo C (7,14%). O estudo possibilitou a detecção da diversidade genética de A. marginale em bezerros dessa propriedade leiteira, além de identificar a sequência BRA1 nos animais do presente estudo, a qual foi diagnosticada recentemente em Minas Gerais, o que demonstra a dispersão das estirpes de A. marginale nos rebanhos de diferentes estados brasileiros. A diversidade genética de A. marginale foi observada tanto em bezerros sintomáticos quanto em assintomáticos e não houve diferença significativa quando se comparou os sinais clínicos ao genótipo verificado no animal infectado, não observando a prevalência de patogenicidade de estirpes.
