Magnetic resonance imaging (MRI) is the most sensitive method of diagnostic imaging to evaluate soft tissues, specially the brain, however it is expensive. The method is based on the nuclear magnetic resonance phenomenon that occurs when atomic nucleus with magnetic proprieties in the body are submitted to a strong magnetic field, and excited with radio frequency generating a radio frequency signal captured by a receptive antenna. The signal is processed by Fourier Transform for the image formation. This study had the objective to obtain 10 complete exams of heads in cadavers of normal dogs to MRI and to make an Atlas of head structures. The images were obtained with a magnetic resonance unit Gyroscan S15/HP Philips using a magnetic field of 1,5Tesla. The cadavers were positioned with the head into a human head coil and submitted to sagittal slices used to plan transverse and dorsal slices in T1, T2 and DP spin-echo sequences. In T1 we adjusted TR=400ms and TE=30ms, in T2 TR=2000ms and TE=80ms and in DP TR=2000ms and TE=30ms. The slice thickness was 4mm, the number of averages 2, the matrix 256x256, the factor 1,0 and the field of view 14cm. The duration of the complete exam of the head was 74,5minutes. The images obtained with the described sequences and with the human head coil was of good quality. In T1 fat was hyperintense and fluid was hypointense. In T2 fat was less hyperintense and fluid was hyperintense. The cortical bone and the air were hypointense in all sequences used because of the low proton density. The DP sequence showed the best contrast between white and gray matter when compared with T2 and T1 sequences. Distinction of cerebral sulcus and gyrus was possible because T2 showed the cerebrospinal fluid. The identification of bone structures that compound the region, muscles, main venous and arterial vessels and structures of the central nervous system, besides elements of the digestory and respiratory systems and structures of the eyes among others was possible through contrast obtained with MRI. In this study the MRI acquired in T1, DP and T2 were complementary for the anatomic study of the head and been able to demonstrate the structures of the canine head with rich anatomic details. The time used to do the complete exam of the head is compatible with the use in live animals since properly anesthetized and controlled. We had opened a way for the study of live animals and for the beginning of disease investigation, mainly that of neurologic origin because this technique is excellent for brain visualization.

A imagem por ressonância magnética (IRM) é o método de diagnóstico por imagem não invasivo mais sensível para avaliar as partes moles, particularmente o encéfalo, porém trata-se de uma técnica onerosa. O método fundamenta-se no fenômeno da ressonância magnética nuclear que ocorre quando núcleos atômicos com propriedades magnéticas presentes no corpo são submetidos a um campo magnético intenso, sendo posteriormente excitados por energia de radiofrequência e gerando, por sua vez, um sinal de onda de radiofrequência capaz de ser captado por uma antena receptora, passando por um processo matemático, chamado Transformada de Fourier, para posterior formação da imagem. Esse estudo objetivou realizar 10 exames completos da cabeça em cadáveres de cães normais à IRM e confeccionar um Atlas com as estruturas identificadas. As imagens foram adquiridas em um aparelho de ressonância magnética Gyroscan S15/HP Philips com campo magnético de 1,5Tesla. Os cadáveres foram posicionados com a cabeça no interior de uma bobina de cabeça humana e foram submetidos a cortes iniciais sagitais a partir de onde se planejou os cortes transversais e dorsais nas sequências de pulso spin-eco T1, T2 e DP. Em T1 utilizou-se TR=400ms e TE=30ms, T2 utilizou-se TR=2000ms e TE=80ms e na DP utilizou-se TR=2000ms e TE=30ms. A espessura do corte foi de 4mm, o número de médias foi igual a 2, a matriz foi de 256x256, o fator foi igual a 1,0 e o campo de visão foi de 14cm. A duração do exame completo da cabeça foi de 74,5minutos. As imagens obtidas com as sequências utilizadas e com a bobina de cabeça humana foram de boa qualidade. Em T1 a gordura tornou-se hiperintensa e o líquido hipointenso. Em T2 a gordura ficou menos hiperintensa e o líquido hiperintenso. A cortical óssea e o ar foram hipointensos em todas as sequências utilizadas devido a baixa densidade de prótons. A sequência DP mostrou o melhor contraste entre a substância branca e cinzenta quando comparada a T2 e a T1. T2 evidenciou o líquido cefalorraquidiano tornando possível a distinção dos sulcos e giros cerebrais. Através do exame de IRM foi possível, pelo contraste, identificar as estruturas ósseas componentes da arquitetura da região, músculos, grandes vasos venosos e arteriais e estruturas do sistema nervoso central, além de elementos do sistema digestório, respiratório e estruturas dos olhos entre outras. Nesse estudo as IRM adquiridas nas sequências T1, DP e T2 foram complementares para o estudo dos aspectos anatômicos da cabeça de cães demonstrando-os com riqueza de detalhes. O tempo requerido para o exame completo da cabeça é compátivel para uso em animais vivos desde que devidamente anestesiados e controlados. Os resultados obtidos por esse trabalho abrem caminho em nosso meio, para o estudo de animais vivos e para o início da investigação de doenças, principalmente as de origem neurológica, visto ser esta técnica excelente para a visibilização do encéfalo.

The systemic aspect of vascular damage induced by angiotensin II (ANG II) has been poorly explored in the literature. Considering the presence of ANG II and its specific receptor AT1, in several organs, all tissues might be potentially affected by its effects. The aims of this study were: To evaluate the early histological changes in the heart, liver and kidneys, produced by ANG II infusion, to evaluate the protective effect of losartan. Wistar rats were distributed into three groups: control (no treatment), treated with ANG II, and treated with ANG II + losartan. ANG II was continuously infused over 72 hours by subcutaneous osmotic pumps. Histological sections of the myocardium, kidneys and liver were stained and observed for the presence of necrosis. There were ANG II-induced perivascular inflammation and necrosis of the arteriolar wall in the myocardium, kidney, and liver by, which were partially prevented by losartan. There was no significant correlation between heart and kidney damage. Tissue lesion severity was lower than that of vascular lesions, without statistical difference between groups. ANG II causes vascular injury in the heart, kidneys and liver, indicating a systemic vasculotoxic effect; the mechanisms of damage/protection vary depending on the target organ; perivascular lesions may occur even when anti-hypertensive doses of losartan are used.

O aspecto sistêmico da lesão vascular induzida pela angiotensina II (ANG II) tem sido pouco explorada na literatura. Considerando a presença de ANG II e de seu receptor AT1 em diversos órgãos, todos os tecidos poderiam ser potencialmente afetados por esses efeitos. Os objetivos deste estudo foram: avaliar as alterações histológicas iniciais no coração, fígado e rins, produzidas pela infusão de ANG II, e avaliar o efeito protetor do losartan. Ratos Wistar foram divididos em três grupos: controle (sem tratamento), tratados com ANG II, e tratados com ANG II + losartan. A ANG II foi infundida continuamente por 72 horas por meio de mini-bombas osmóticas. Foram realizados cortes histológicos de miocárdio, rim e fígado para coloração e observação para a presença de necrose. Observou-se a presença de inflamação perivascular e necrose de parede arteriolar em miocárdio, rins e fígado, que foram parcialmente prevenidas pelo losartan. Não houve correlação significante entre as lesões observadas no coração e nos rins. A severidade da lesão tissular foi menor quando comparada às lesões vasculares, sem diferença estatística entre os grupos. A ANG II causa injúria vascular no coração, rins e fígado, sugerindo um efeito vasculotóxico sistêmico; os mecanismos de lesão/proteção variam dependendo do órgão afetado; as lesões perivasculares podem ocorrer mesmo quando doses anti-hipertensivas de losartan forem utilizadas.

Radiography and ultrasonography were evaluated as tools for diagnosis of the rupture of cranial cruciate ligament (CrCL) in dogs. Twenty-five dogs were submitted to radiographic and ultrasonographic examinations and their results were compared with those obtained by artrotomy (gold standard). Radiography detected the rupture in 84% (21/25) of the cases, but 16% (4/25) were false-negative. Ultrasonography identified accurately 76% (19/25) of the cases and gave a probable diagnosis for the remaining 24% (6/25) what means that this technique presented 100% of positive results. It was possible to conclude that radiography and ultrasonography are valuable tools for the diagnosis of CrCL rupture in dogs.

Radiografia e ultrassonografia foram avaliadas como técnicas no diagnóstico por imagem na ruptura do ligamento cruzado cranial (LCCr) em cães. Vinte e cinco cães foram submetidos à radiografia e ultrassonografia e seus resultados foram comparados aos obtidos por artrotomia (teste padrão ouro). O exame radiográfico diagnosticou corretamente a lesão em 84% (21/25) dos casos, mas 16% (4/25) apresentaram resultado falso-negativo. O exame ultrassonográfico foi capaz de diagnosticar acertadamente 76% (19/25) dos casos, e sugeriu a ruptura do LCCr nos 24% (6/25) restantes, apresentando 100% de resultados positivos. Concluiu-se que a radiografia e a ultrassonografia são ferramentas valiosas para diagnosticar casos de ruptura do LCCr em cães.

Currently, wild animals have been received special attention for domestic breeding. In Wild Animal Medicine, ultrasonographic exams could be considered as a tool to diagnose and prevent disease. Ultrasonographic examination of twenty snakes (Boa constrictor) was performed in order to describe the morphological and ultrasonography appearance of their coelomic structures. Examination by ultrasonography revealed the parechymal liver ranging from slightly hypoechogenic to hyperechogenic, with echogenic margins and homogeneous echotexture in total scanning of this organ. The kidney was accessible for examination by sagittal ultrasound, showing an ellipsoid shape, a hyperechogenic capsule with a thin and regular wall. Ultrasonographic scanning of ovarian follicle showed an ovoid shape, as well as a thin, regular and slightly hyperechogenic edge. No evident structures from male reproductive system were identified by ultrasound examination because of their similar echogenic appearance to the tissues around and the “body fat” that lies in this region. Examination by ultrasound of the coelomic structures was a fast and repeatable technique, which provides useful data to recognize the morphology, sintopy and ultrasonographic appearance of organs such as liver, kidneys and vitellogenic follicles.

Twenty-five BVDV strains, detected in serum from persistently infected cattle from Peru (n=15) and Chile (n=10) were genetically characterized. The phylogenetic analysis based on the 5’ UTR showed that all 25 strains belonged to genotype 1. Twenty-three of the strains could further be subdivided into subtype 1b, and two out of ten Chilean strains into subtype 1a. In conclusion, in total 23 out of 25 strains analyzed were of genotype 1, subtype 1b. This is the predominant BVDV subtype in many countries all over the world, including USA. The close homology with previously described strains reflects the influence of livestock trade on the diversity of BVDV circulating within and between countries and continents. Peru and Chile have imported large numbers of cattle from USA and Europe, mostly with insufficient or lacking health documentation.

Clinical, serological and parasitemia exams were realized. Parasite investigation by bioassay and PCR was realized on semen and fragments of skeletal musculature, lymph nodes, brain, retina, spleen, liver, lung, testicle, epididymis and seminal vesicle. Blood and semen samples were collected on days -2, -1, 1, 3, 5, 7, 14 and weekly thereafter, up to postinfection day (PID) 84. The inoculated steers (GI and GII) presented hyperthermia from PID 3 to 16. Antibodies against T. gondii were detected through the indirect fluorescence antibody test (IFAT) on PID 5 (1:16) in both inoculated groups (oocysts and tachyzoites), reaching peaks of 1:4096 on PID 7. Parasitemia outbursts occurred in all infected bovines, principally from PID 7 to 28, independent of the strain and inoculate used. Bioassays revealed the presence of parasites in semen samples of animals infected with oocysts (GI) and tachyzoites (GII) on several experimental days between PID 7 and 84. Tissue parasitism by T. gondii was diagnosed by bioassay and the PCR technique in several organ and tissue fragments. These findings suggest the possibility of sexual transmission of T. gondii in the bovine species.

Porcine circovirus 2 (PCV2) is generally associated with the porcine circovirosis syndrome, which is considered an important disease of swine and has potentially serious economic impact on the swine industry worldwide. This article describes the construction of a recombinant plasmid expressing the PCV2 structural protein and the evaluation of cellular and humoral immune responses produced by this recombinant vaccine in BALB/c mice. The vaccine candidate was obtained and analyzed in vivo, in an effort to determine the ability to induce a specific immune response in mice. DNA was extracted from a Brazilian PCV2 isolate and the gene coding for Cap protein was amplified by PCR and inserted into an expression plasmid. Groups of BALB/c mice were inoculated intra-muscularly and intradermally in a 15-day interval, with 100 µg and 50 µg of the vaccine construct, respectively. Another group was inoculated intramuscularly with 100 µg of empty plasmid, corresponding to the control group. Seroconversion and cellular response in BALB/c mice were compared and used for vaccine evaluation. Seroconversion was analyzed by ELISA. After a series of 3 immunizations the spleen cells of the immunized animals were used to perform lymphocyte proliferation assays. Seroconversion to PCV2 was detected by ELISA in the animals inoculated with the vaccine construct when compared with control groups. Lymphocyte proliferation assays showed a stronger cell proliferation in the inoculated animals compared with the control group. Thus, the vaccine candidate construct demonstrated to be able to induce both humoral and cellular responses in inoculated mice.

Porcine circovirus 2 (PCV2) is generally associated with the porcine circovirosis syndrome, which is considered an important disease of swine and has potentially serious economic impact on the swine industry worldwide. This article describes the construction of a recombinant plasmid expressing the PCV2 structural protein and the evaluation of cellular and humoral immune responses produced by this recombinant vaccine in BALB/c mice. The vaccine candidate was obtained and analyzed in vivo, in an effort to determine the ability to induce a specific immune response in mice. DNA was extracted from a Brazilian PCV2 isolate and the gene coding for Cap protein was amplified by PCR and inserted into an expression plasmid. Groups of BALB/c mice were inoculated intra-muscularly and intradermally in a 15-day interval, with 100 µg and 50 µg of the vaccine construct, respectively. Another group was inoculated intramuscularly with 100 µg of empty plasmid, corresponding to the control group. Seroconversion and cellular response in BALB/c mice were compared and used for vaccine evaluation. Seroconversion was analyzed by ELISA. After a series of 3 immunizations the spleen cells of the immunized animals were used to perform lymphocyte proliferation assays. Seroconversion to PCV2 was detected by ELISA in the animals inoculated with the vaccine construct when compared with control groups. Lymphocyte proliferation assays showed a stronger cell proliferation in the inoculated animals compared with the control group. Thus, the vaccine candidate construct demonstrated to be able to induce both humoral and cellular responses in inoculated mice.

Both healthy eyes of 10 six-year-old male and female mongrel dogs were studied. With a contact specular microscope the corneal endothelium was examined. Endothelial cells were analyzed in the central and peripheral cornea. Morphological analysis with regard to polymegathism and pleomorphism was performed. Three images of each region with at least 100 cells were obtained. The analysis showed that polygonal cells formed a mosaic-like pattern uniform in size and shape. The predominant number of cells was hexagonal. The polymegathism index was 0.22. The study demonstrates that the morphology of the normal corneal endothelial cells of dogs is similar to that found in the human cornea.

The Brown Capuchin, Cebus apella, has a wide distribution in the northern and southern Brazilian Amazon region and in the Cerrado (savanna). These monkeys are usually submitted to predatory chase, increasing the need for preservation of this wild animal species. An ultrasonographic examination of 10 Brown Capuchins was made in order to describe the normal ultrasonographic anatomy of their abdominal cavity. The urinary bladder revealed its wall thickness with an average of 0.2cm, the topographic situation of which allowed close relation with the wall of uterus and descendent colon. Using caudal abdominal scan, images of aorta, caudal vena cava and right iliac vein were obtained. Liver was accessible for examination by sagittal and cross-section ultrasound, allowing visualization of gallbladder and hepatic vessels. Renal scan allowed accuracy to evidence the echogenicity differences between pelvis, renal sinus, as well as the cortical-medullary relationship. The mean length of the kidneys was 6.24±0.31cm, and no significant differences were observed between left and right kidney length (Student’s t-test and ANOVA). The renal volume obtained was 2.37±0.18cm3. Correlation Coefficients of Pearson between right and left renal length and between right and left renal volume were r = 0.74 and 0.51. Mean thickness for cortical and medullar regions was 0.75±0.11 and 0.39±0.06cm, respectively. Correlation Coefficient of corticomedullar relation between right and left renal was r = 0.19. Examination by ultrasound was considered an efficient, non-invasive, fast and repeatable technique which provides useful data for clinicians and surgeons engaged in wild animal medicine.

Risk areas of canine visceral leishmaniasis in the city of Camaçari, Bahia, Brazil, were investigated. A total of 278 dogs from 141 homes pertaining to 20 investigated risk areas was serologically screened (ELISA). The general seroprevalence was 21.7% (56/258) after exclusion of 20 dogs used at the beginning of the survey to limit the study area. The respective results of the univariated and multivariated analysis of factors related to infection of dogs by Leishmania chagasi, to vector distribu-tion pattern in the area and to the methodology used to localize the canine focuses are discussed.
