ABSTRACT.- Verocai G.G., Fernandes J.I., Correia T.R., Melo R.M.P.S., Alves P.A.M., Scott F.B. & Grisi L. 2009. Inefficacy of albendazole sulphoxide and ivermectin for the treatment of bovine parasitic otitis caused by rhabditiform nematodes. Pesquisa Veterinária Brasileira 29(11):910-912. Departamento de Parasitologia Animal, Instituto de Veterinária, Universidade Federal Rural do Rio de Janeiro, Seropédica, RJ 23890-000, Brazil. E-mail: gverocai@gmail.com The purpose of this study was to evaluate the efficacy of orally administered albendazole sulphoxide and pour-on ivermectin for the treatment of bovine parasitic otitis caused by rhabditiform nematodes. Eighteen Gyr cows presenting clinical otitis were divided in three groups with six animals each. The first one did not receive any treatment (control group). The second one was treated with 0.5% pour-on ivermectin, 500µg/kg of body weight, and the third group was treated with oral 6% albendazole sulphoxide, at 6.0mg/kg. Both ear canals of each animal were reexamined on days 7 and 21 post treatment. The animals in the control group remained infected throughout the days of observation. Ivermectin treatment did not show effectiveness on days 7 or 21 post treatment. The albendazole sulphoxide treatment had an efficacy of 16.7 and 25% on days 7 and 21, respectively. Further studies are required to assess an effective treatment for this parasitic disease, especially via alternative administration routes, because of its significant impact on Bos taurus indicus cattle breeding in Tropical and Subtropical Regions.

#112 - Shedding and transmission of bovine viral diarrhea virus by persistently infected calves, 29(9):736-742

Abstract in English:

ABSTRACT.- Arenhart S., Bauermann F.V., Oliveira S.A.M., Weiblen R. & Flores E.F. 2009. [Shedding and transmission of bovine viral diarrhea virus by persistently infected calves.] E-mail: eduardofurtadoflores@gmail.com Persistently infected (PI) calves born to cows infected with non-cytopathic bovine virus diarrhea virus (BVDV) represent the main reservoir of the virus in nature. We herein report an investigation on the patterns of virus shedding and transmission by five PI calves produced experimentally through inoculation of pregnant cows with Brazilian BVDV isolates. Five calves that survived intrauterine infection were born healthy, lacking neutralizing antibodies to BVDV and harboring virus in the blood. After weaning - and following the disappearance of colostral antibodies - PI calves were monitored for virus in serum and body secretions (ocular, oral, nasal and genital) at weekly intervals for up to 150 days. For each animal, the virus titers in serum showed minor variations throughout the collections (with one exception that presented an increase late in infection), yet the titers varied widely among animals (from 102 to 106TCID50/mL). Virus shedding in secretions was detected steadily during all the observation period with minor titer variations for each particular animal. The highest titers were generally detected in nasal and ocular secretions (titers 104 to 106TCID50mL) whereas genital and oral secretions usually contained low amount of virus (102 to 103TCID50mL). To evaluate the kinetics of virus transmission by these animals, one PI was introduced on a group of 10 seronegative calves maintained with a high animal density simulating the conditions of an intensive management. All 10 contact calves seroconverted to BVDV by day 30. Another PI calf was introduced into a 48-head herd kept under a low animal density, extensive grass management. Among these animals, 8/48 (16.6%) seroconverted by day 10, 26/48 (54.1%) by day 40 and 37/48 (77%) were seropositive at day 100, when the monitoring was discontinued. These results show that continuous viremia and virus shedding in high titers in secretions by PI animals assure an efficient and rapid virus transmission to contact animals, being the kinetics of transmission much faster under intensive conditions.

RESUMO.- Arenhart S., Bauermann F.V., Oliveira S.A.M., Weiblen R. & Flores E.F. 2009. [Shedding and transmission of bovine viral diarrhea virus by persistently infected calves.]

E-mail: cocaarduino@yahoo.com.br In the investigation 100 heifers were used, divided into 5 groups of 20 animals each. The four experimental groups were vaccinated using distinct commercial polyvalent bacterines: A, B, C and D, and a group was the control. Samples were collected at days 0, 3, 7, 14, 21, 28, 35, 42, 42, 56, 63, 70, 77, 84, 91, 120, 150 and 180 from the first injection of the vaccine. The selection of the animals for the experimental groups was done based on a serological screening with 6 antigens of Leptospira sp. constituted by non-reagent animals. The vaccine titers were monitored using the microscopic agglutination test (MAT) for Canicola, Grippotyphosa, Hardjo, Icterohaemorrhagiae, Pomona and Wolffi serovars. All vaccines used were capable to product agglutinins for the Hardjo and Wolffi serovars observed at 3 days after vaccination, remaining until the 150th day; those serovars induced the highest titres of agglutinins. Vaccine D, in spite of not containing the Wolffi serovar, induced the production of agglutinins to this serovar. Agglutinins to the Canicola serovar were only observed in the animals vaccinated with the D bacterine. Vaccine D induced the highest average titers of antibodies to all tested serovars.

ABSTRACT.- Medeiros E.S., Santos M.V., Pinheiro Jr J.W., Faria E.B., Wanderley G.G., Teles J.A.A. & Mota R.A. 2009. [In vitro evaluation of the efficacy of commercial disinfectants used in pre and post-dipping against Staphylococcus spp. isolated from bovine mastitis.] E-mail: rinaldo.mota@hotmail.com The objective of this investigation was to evaluate the in vitro sensibility of Staphylococcus spp. to several commercially available disinfectants used for pre and post dipping. A total of 60 isolates of Staphylococcus spp., identified as Staphylococcus aureus (50) and Positive coagulase Staphylococcus (10) were obtained from the mammary glands of dairy cows with subclinical mastitis in the regions of Metropolitan Recife, the Agreste and the Zona da Mata of the State of Pernambuco, Brazil. As active ingredients were used a chlorine base (25%), iodine (0.6%), chlorhexidine (2.0%), quaternary ammonium (4.0%), and lactic acid (2.0%) at four specific intervals (15", 30", 60", and 300"). One hundred percent of S. aureus was found to be sensitive to iodine, 93.3% to chlorhexidine, 80% to ammonia, 35.6% to lactic acid, and 97.8% were resistant to chlorine at a 60-minute interval. With respect to the Positive coagulase Staphylococcus (SCP), 100.0% of the isolates were sensitive to iodine, 81.8% to quaternary ammonium, 99.9% to lactic acid, 72.7% to chlorhexidine, and 100% was resistant to chlorine at an interval of 60 minutes. It can be concluded that the highest disinfectant activity in vitro was found to be with iodine and chlorhexidine for S. aureus, and with iodine and lactic acid for SCP. A further conclusion was that it is important to undertake a periodic evaluation of the disinfectants used on the dairy properties in the regions studied, given the variety of sensibilities and resistance to disinfectants used, which may prejudice the control of bovine mastitis caused by Staphylococcus spp.

Abstract in English:

ABSTRACT.- Arenhart S., Silva L.F., Henzel A., Ferreira R., Weiblen R. & Flores E.F. 2008. [Fetal protection against bovine viral diarrhea virus (BVDV) in pregnant cows previously immunized with an experimental attenuated vaccine.] E-mail: flores@ccr.ufsm.br This paper reports the antibody response and fetal protection in pregnant cows conferred by an experimental vaccine containing two attenuated strains of bovine viral diarrhea virus (BVDV-1 and BVDV-2). Cows (n=19) were vaccinated twice, with a 34-days-interval, with the experimental vaccine and together with non-vaccinated controls (n=18), were mated and challenged between days 60 and 90 of gestation by intranasal inoculation of four heterologous BVDV-1 and BVDV-2 isolates. The antibody response was evaluated by serum-neutralization tests performed at different intervals after vaccination (days 34, 78 and 138 post-vaccination [pv]). Fetal protection was monitored by ultrassonographic and clinical examination of the dams and fetuses during the rest of gestation; and through virological and serological examination of pre-colostral blood obtained from aborted and/or recently born fetuses/calves. At the day of challenge (day 138 pv), all vaccinated cows had neutralizing antibodies in high titers against BVDV-1 (1,280- ³10,240), and with one exception (titer 20), presented moderate to high titers to BVDV-2 (80-1,280). At the end of the monitoring, only three out of 18 control cows (16.6%) delivered healthy, virus-free calves. Fifteen non-vaccinated cows (83.3%) presented signs of fetal infection and/or had reproductive losses. Seven of these cows (38.8%) delivered virus-positive calves; five were healthy and survived (27.7%); two were premature or weak and lasted three and 15 days, respectively. The other eight cows (44.4%) aborted between day 30 post-challenge and the parturition; or delivered premature or stillbirth calves. In contrast, 17 out of 19 (89.4%) vaccinated cows delivery virus-free, healthy calves. One vaccinated cow aborted around day 130 post-challenge, yet this fetus could not be examined for the presence of virus. Another cow delivered a virus-positive calf (5.2%). In summary, the experimental vaccine induced adequate antibody titers in most animals and the immunological response induced by vaccination was able to prevent fetal infection and reproductive losses upon challenge with a pool of heterologous BVDV isolates. Hence, this experimental vaccine may be an attractive alternative for the prevention of reproductive losses associated with BVDV infection.

E-mail: fverechia@dracena.unesp.br The specific region of maternal fetal interface needs to be clarified and corresponds to the “arcade zone” of sheep and goat placentomes. In small ruminants that area is also characterized by macroscopic blood extravasation (hemophagous areas). This occurs possibly because the iron is transferred to the embryo by trophoblastic erytrophagocytosis in these hemophagous placental areas. In order to investigate the hemophagous placental areas in cattle, placentomes of 34 pregnant Bos indicus cows (2-3, 4-6, 7-8 and 9 months of gestation) were analyzed. They were fixed by perfusion with 10% formaldehyde aqueous solution and 4% paraformoldehyde in PBS, pH 7.4, 0.1M, processed and stained for light microscopy and histochemistry. The methodology used allowed to observe placental haematomes between the uterine and trophoblastic epithelium since 3 months of pregnancy. Erythrocytes had been found in the trophoblastic cells, elucidating the erytrophagocytosis. Through Perl’s histochemical reaction it was possible to prove the existence of ferric iron in the trophoblastic cells. The PAS reaction was positive staining mucoid substance in the epithelial cells, especially in the binucleate cells of the fetal epithelium. Based on histology and histochemistry, it can be inferred that the hemophagous areas are important sites for iron transfer in the bovine placenta.

ABSTRACT.- Mathias L.A., Meirelles R.B. & Buchala F.G. 2007. [Stability of Brucella abortus whole cell antigen for use in the serological diagnosis of bovine brucellosis by the complement fixation test.] E-mail: lmathias@fcav.unesp.br The complement fixation test is used worldwide in the confirmatory diagnosis of bovine brucellosis. For this technique the antigen is the same as the one used in the tube agglutination test. However, literature is poor in information about the stability of the whole cell Brucella antigen for use in the complement fixation test to establish a time of validity of the antigen. Hence the aim of this investigation was to evaluate the stability of this antigen under refrigeration for use in the complement fixation test. Fourteen batches of antigen prepared with Brucella abortus strain 1119/3, produced from 9 months to 23 years and 11 months before, were analysed. One hundred and sixty-seven cattle sera with varying titres of antibodies to Brucella were tested through the warm complement fixation microtechnique with five 50% haemolytic units of complement. Sera with at least 25% of complement fixation in dilution 1:4 were considered positive. The results with 13 of the antigen batches were compared with the results obtained with the batch produced 9 months before by the McNemar c2 test and kappa statistic. The oldest antigen batch gave a higher proportion of sera titres which were exactly the same observed with the 9-month-batch (90.4%), and the antigen produced 4 years and 3 months before the test gave de lowest proportion of sera with the same titre of the 9-month-antigen (73.7%). The comparison of the results after being classified as positive and negative showed that the highest proportion of agreed results was observed with the antigen produced 21 years and 4 months before (98.8%, kappa 0.98). The antigen with the lowest proportion of agreed results was the one produced 3 years and 2 months before (91.6%, kappa 0.84). The results of the study show that most sera gave very similar results with all antigen batches evaluated, and that there was no relationship between the period of antigen production and the difference in test results.

E-mail: lmathias@fcav.unesp.br The complement fixation test is used worldwide in the confirmatory diagnosis of bovine brucellosis.

Abstract in English:

ABSTRACT.- Monteiro L.A.R.C., Pellegrin A.O., Ishikawa M.M. & Osório A.L.A.R. 2006. [Epidemiological investigation of bovine brucellosis in an extract of Mato Grosso do Sul, Brazil.] E-mail: leticiacmonteiro@yahoo.com.br The study aimed to estimate the prevalence of bovine brucellosis in 22 counties which make up the region Extract 1 of the State of Mato Grosso do Sul, Brazil, in order to identify risk factors associated with the infection. The sample region encompasses an area of 70,214.1 km² and represents 19.7% of the State. The region studied has about 5.7 million head of cattle, corresponding to 23% of the total of 24.9 million cattle in the State of Mato Grosso do Sul. On 210 farms, between December 2003 and March 2004, 2,376 blood samples were collected from cows, aged 24 months or older, for serial diagnostic tests. Screening through the buffered acidified antigen test was confirmed by the 2-mercaptoetanol test. On the occasion of sample collection a questionnaire with information related to identification, kind of cattle and management practices was filled out. In individual animals the real prevalence was estimated at 5.6%, and in the cattle herds at 37.3%. The variables, which presented association through odds ratio (OR), univariate analysis and 95% confidence interval (CI) with serum positivity for brucellosis, were: the exploration of beef cattle (OR = 2.82, 95% CI = 1.49-5.34), Zebu breed (OR = 2.62, 95% CI = 1.40-4.88) and abortion (OR = 1.83, 95% CI = 1.01-3.33). The results shown here demonstrate, despite the prevalence of brucellosis in the extract of Mato Grosso do Sul studied, that the control of the disease may depend on adoption of a program focusing upon the exploration of beef cattle, the Zebu breed and the occurrence of abortion.

[Tick fever in southern Brazil: a retrospective study of 1978-2005.] Tristeza parasitária bovina na região sul do Rio Grande do Sul: estudo retrospectivo de 1978-2005. Pesquisa Veterinária Brasileira 26(4):236-242. Laboratório Regional de Diagnóstico, Faculdade de Veterinária, Universidade Federal de Pelotas, Campus Universitário s/n, Pelotas, RS 96010-900, Brazil. E-mail: alschild@terra.com.br A retrospective study of tick fever was made, which occurred from 1978-2005 in southern Rio Grande do Sul in the influence area of the Regional Diagnostic Laboratory of the Federal University of Pelotas. From 4,884 cattle specimens, sent by practitioners or which were from necropsies performed at the Diagnostic Laboratory, 231 (4.7%) were diagnosed as tick fever. Data from 221 of those outbreaks were analyzed. Ninety one (41.1%) outbreaks were caused by Babesia bovis, 11 (4.9%) by Babesia bigemina, and 65 (29.41%) by Anaplasma marginale. In other 33 (14.93%) outbreaks of babesiosis there is no information if the disease was caused by B. bovis or B. bigemina, and 21 (9.5%) outbreaks were caused by mixed infection of A. marginale and B. bovis or B. bigemina. Mean morbidity, mortality, and letality rates in 149 outbreaks were 11.17%, 6.81%, and 70.04%, respectively. Most outbreaks occurred during summer (January-March) and autumn (April-June), mainly in 1 to 3-year-old cattle. Clinical signs were depression, weakness, fallen ears, fever, and weight loss. Low packed cell volume values were always found. Hemoglobinury was observed in babesiosis. Neurological signs characterized by gait alterations, muscular tremors, aggressiveness and falling down with tonic and clonic convulsions were observed in babesiosis by B. bovis. The main gross lesions were anemia, jaundice, splenomegaly, hepatomegaly, yellow liver and cardiac hemorrhages. Hemoglobinury was observed in babesiosis, and congestion of the cerebral cortex in babesiosis by B. bovis. It is concluded that B. bovis is the main agent causing thick fever in southern Rio Grande do Sul. In that region with a cattle population of 2,630,000 heads the annual losses due to tick fever can be estimated in 6,220 cattle or US$ 1,623,000.00. Preventive measures to diminish tick fever losses in the region are necessary.

Departamento de Medicina Veterinária, Universidade para o Desenvolvimento do Estado e Região do Pantanal (Uniderp), Rua Alexandre Herculano 1400, Parque dos Poderes, Campo Grande, MS 79037-280, Brazil. E-mail: gustavoj@mail.uniderp.br The study evaluated the use of a reduced dose of the Brucella abortus strain 19 vaccine, in an adult herd negative for the disease, by serological diagnostic techniques, advocated by the Brazilian Program for Animal Brucellosis and Tuberculosis Control and Eradication, and by an indirect ELISA. The complement fixation test detecteed 46.77% positives, the rose bengal test 67.74%, the mercaptoethanol with standard agglutination test 87.09% and the ELISA ID 100%. The reduced dose influenced the serological diagnosis. None of the techniques reached a suitable specificity for use in the herd under those conditions, up to 3 months after vaccination.
