E-mail: marinabrito@usp.br Duchenne muscular dystrophy (DMD) is a neuromuscular disorder characterized by a continuous muscle necrosis and degeneration with eventual fibrosis and fatty tissue infiltration. Progressive increase in muscle interstitial fibrosis prevents the movement of myogenic cells, which is necessary for myotube formation. Canine model is the best phenocopies of the disease in humans when comparing with others animal models with dystrophy. Anti-fibrotic treatment of DMD patients, targeting the cytokine mediators, TGF-beta, and the treatment with antiinflammatories, may limit muscle degeneration and contribute for the improvement of the course of the illness. This work aimed to verify the possible adverse effects in renal physiology by means of evaluation sanguineous biochemist and arterial pressure, in order to verifying the viability of Losartan (a TGF-beta inhibiter) in affected dogs by muscle dystrophy. It was used four adults dogs, two of each gender. A dose of 50mg of Losartan was orally given once a day. The clinical exams, the kidney function, arterial blood pressure and potassium level did not show any adverse effect through the experimental period. Losartan utilization showed to be a safe therapy for the antifibrotic treatment in adults dogs, not affecting neither the kidney function nor the arterial blood pressure.

The main gross lesions were enlargement of the Peyer’s patches in the small intestine and hyperemia in the intestinal mucosa and serosa. Microscopically, the small intestine showed necrotizing enteritis in 77% (74/96) of the dogs examined. However, in 17.7% of the histological evaluation in the small intestine were damaged due to autolytic changes making it difficult to obtain an appropriate interpretation. The immunohistochemistry test was performed in tissues of small intestine, mesenteric lymph nodes, thymus, spleen, tonsils, tongue, and bone marrow in all the 96 selected cases. Parvovirus antigen was detected in 91.6% (88/96) of the dogs necropsied. The best result of the IHC test was seen in samples of small intestine which was positive in 77% (74/96) of the cases. The statistical analysis (Fisher test) showed a weak association between intestinal autolysis and positive result of the IHC test. The chance of the autolysed intestine showing a positive result in the immunohistochemistry test was 0.33 less (OR=0.33, 95% CI:0.10-1.17) when compared with small intestine not autolysed.

Viral antigen was detected by an immunohistochemical procedure using a mouse monoclonal anti-canine distemper antibody. The footpads were the more constantly (67.4% of the cases) immunolabeled tissue, followed by stomach with 62.7%. Immunohistochemistry was demonstrated to be a useful tool for the study of viral antigen distribution in distemper affected dogs as well as it indicated which is the best tissue to be examined in order to confirm a suspected case of canine.

E-mail: raquelsartor@yahoo.com.br A technique to restore acetabular anatomy by deepening the acetabular cavity and reconstructing the femoral head ligament and the joint capsule was tested on nine large breed dogs with severe hip dysplasia and acute subdislocation or dislocation. The technique consisted of two phases. First, all dogs were submitted to bilateral pectinotomy. In a second surgical intervention on the same dogs the acetabulum was approached and deepened, and the femoral head ligament and the joint capsule were reconstructed. In general, within 30 days of the surgery dogs could stand on the operated member to walk. Except for two dogs, all the others recovered pelvic member locomotive ability within 60-90 days after surgery. It is concluded that acetabuloplasty is a good alternative for treatment of severe canine hip dysplasia.

In contrast, overexpression of maspin is correlated with poor prognosis in other types of cancer. Little is known about expression, regulation and function of maspin in canine mammary tumors. It was demonstrated in this study, a loss of maspin expression in malignant canine mammary cells compared with a pool of normal canine mammary tissue, analyzed by quantitative real-time PCR; weak maspin expression in malignant canine mammary tumors were observed by immunohistochemistry. It was also demonstrated that a correlation with nuclear maspin expression and a good prognosis. It is suggested that maspin could be used as a prognostic marker in canine mammary neoplasia.

The labelled thymine was uptaken by the three cases of VCTT. 99mTc-Thymine is a promising imaging technique for non-invasive veterinarian evaluation of tumoral dissemination degree decurrent from the VCTT cases.

The expression of these markers was related to the clinical-pathological characteristics of 73 surgically removed mammary tumors in female dogs by immunohistochemistry. There was no statistical correlation between these markers and death by neoplasm, survival time and disease-free interval. However, the loss of e-cadherin expression and marked Ki-67 expression (p=0.016) were considered statistically significant for the diagnosis (p=0.032). When evaluated as independent factors, there was evidence of the relationship between the loss of e-cadherin expression and high PCNA expression with changes in the body status (divided into obese, normal and cachectic) of female dogs (p=0.030); there was also evidence of the relationship between pseudopregnancy and e-cadherin alone (p=0.021) and for ulceration and PCNA alone (p=0.035). The significant correlation between the markers expression and these well known prognostic factors used individually or in combination suggests their prognostic value in canine mammary tumors.

#128 - Hepatite infecciosa canina: 62 casos, p.325-332

Pesquisa Veterinária Brasileira 27(8):325-332. Departamento de Patologia, Universidade Federal de Santa Maria, 97105-900 Santa Maria, RS, Brazil. E-mail: claudioslbarros@uol.com.br Necropsy reports from 5,361 dogs necropsied over a 43-year period (1964-2006) were reviewed in search for cases of infectious canine hepatitis (ICH). Sixty two (1.2%) cases of the disease were found. Most of the 62 affected dogs (91.9%,) were 2-year-old or less. Clinical signs were recorded in the necropsy files of 45 ICH affected dogs and included anorexia (55.6%), apathy (35.6%), diarrhea (35.6%) (often with blood [43,8%]), neurological signs (33.3%), vomiting (26.7%), petechiae and echymosis in the mucous membranes and/or skin (24.4%), hypothermia (20.0%), abdominal pain (15.6%), icterus (13.3%), enlargement and congestion of the tonsils (11.1%), fever (11.1%) and ascites (6.7%). The clinical courses lasted from few hours to 15 days. The most frequent necropsy findings included hepatic changes (87.1%), edematous, congested and hemorrhagic lymph nodes (51.6%), bloodstained fluid, clear fluid or whole blood in the abdominal cavity (35.5%), and petechial or paint-brush hemorrhages over the pleural (27.4%) and gastrointestinal (24.2%) serosal surfaces. In 12.9% of the cases there was a granularity to the intestinal serosa. Hemorrhages in the leptomeninges and in the substance of the brain were observed in 9.7% of the cases. Hepatic gross changes included moderately enlarged and more friable livers with marked lobular pattern, congestion and multifocal pale or hemorrhagic foci of necrosis. Films and strands of fibrin covered the hepatic surface in 20.4% of the cases and in 27.8% of the cases the gall bladder was thickened by edema. Zonal or randomly distributed multifocal hepatic necrosis (93.5%) associated with intranuclear inclusion bodies were the most consistent microscopic findings. Intranuclear inclusion bodies were found in the liver in every case and their detection was the criterium for confirmation of the diagnosis. The most significant microscopic extra-hepatic lesions included hemorrhages and intranuclear inclusion bodies in endothelial and reticuloendothelial cells of the renal glomeruli (50.0%) lymph nodes (47.8%), brain (27.8%), tonsils (25.0%) and spleen (10.0%).

E-mail: claudioslbarros@uol.com.br Necropsy reports from 5,361 dogs necropsied over a 43-year period (1964-2006) were reviewed in search for cases of infectious canine hepatitis (ICH). Sixty two (1.2%) cases of the disease were found. Most of the 62 affected dogs (91.9%,) were 2-year-old or less. Clinical signs were recorded in the necropsy files of 45 ICH affected dogs and included anorexia (55.6%), apathy (35.6%), diarrhea (35.6%) (often with blood [43,8%]), neurological signs (33.3%), vomiting (26.7%), petechiae and echymosis in the mucous membranes and/or skin (24.4%), hypothermia (20.0%), abdominal pain (15.6%), icterus (13.3%), enlargement and congestion of the tonsils (11.1%), fever (11.1%) and ascites (6.7%). The clinical courses lasted from few hours to 15 days. The most frequent necropsy findings included hepatic changes (87.1%), edematous, congested and hemorrhagic lymph nodes (51.6%), bloodstained fluid, clear fluid or whole blood in the abdominal cavity (35.5%), and petechial or paint-brush hemorrhages over the pleural (27.4%) and gastrointestinal (24.2%) serosal surfaces. In 12.9% of the cases there was a granularity to the intestinal serosa. Hemorrhages in the leptomeninges and in the substance of the brain were observed in 9.7% of the cases. Hepatic gross changes included moderately enlarged and more friable livers with marked lobular pattern, congestion and multifocal pale or hemorrhagic foci of necrosis. Films and strands of fibrin covered the hepatic surface in 20.4% of the cases and in 27.8% of the cases the gall bladder was thickened by edema. A significant difference in the response was observed between the two groups (p<0.05). Only one dog (5%) in the control group reacted to the intradermal test, whereas 14 dogs (56%) in the allergic group were positive for at least one extract (odds ratio = 24.2). Most of the positive reactions observed in the allergic group occurred against the extracts of T. putrescentiae or L. destructor, each inducing reactions in 10 dogs (40%). D. farinae, D. pteronyssinus e B. tropicalis extracts induced reactions in 7 (28%), 3 (12%) and 3 (12%) dogs, respectively. The allergenic extracts standardized for humans evaluated in the present study may be used as a tool to complement the diagnosis of the disease, as well as to select potential allergen candidates for allergen-specific immunotherapy.

E-mail: claudioslbarros@uol.com.br The files of 5,361 necropsies performed in dogs in the Veterinary Pathology Laboratory of the Federal University of Santa Maria during 1965-2006 were reviewed in search of cases of canine distemper. Six hundred and eighty three cases (12.7%) of the disease were found, 620 of which had neurological signs. From those 620, the following data on each case were retrieved: age, clinical signs, histopathology and concomitance or not of another disease. Age groups were classified as puppies (up to 1 year of age), adults (from 1 to 9 years) and aged (from 10 years on). In 565 out of the 620 (91.1%) neurological cases of canine distemper, histopathological brain changes were observed and in 554 of those 565 the age was registered in the files with following age group distribution: 45.9% of puppies, 51.4% of adults, and 2.7% of aged dogs. Neurological clinical signs encompassed a large spectrum of motor, postural and behavioral disturbances which could occur together or individually. Most frequent clinical signs were myoclonus (38.4%), motor incoordination (25.0%), seizures (18.5%), and paraplegia (13.4%). In 98.4% of the 565 dogs with histopathological changes in the brain demyelination, non-suppurative encephalitis or a combination of these two were found. Intranuclear eosinophilic inclusion bodies were observed in different brain cells of 343 of the 565 dogs with histopathological changes. In 170 (49.6%) the cellular type bearing the inclusions was not mentioned in the file and in the remaining cases the inclusions were seen in astrocytes (94.8% of the cases), neurons (3.5%), oligodendrocytes (1.1%), and ependyma cells (0.6%). Taking in consideration the type of lesions and the age groups, cases with combined demyelination and non-suppurative encephalitis occurred in 40.0% of the puppies, 51.2% of the adult dogs and 72.7% of the aged dogs. Demyelination alone occurred in 48.4% of the puppies, 41.3% of the adults and in 35.7% of the aged dogs. Non-suppurative encephalitis alone occurred 11.6% of the puppies, 7.5% of the adults and in 7.1% of the aged dogs.
