#161 - Glicogenose hereditária em bovinos Brahman no Brasil, p.210-214

The disease was diagnosed in a Brahman herd in the municipality of Porto Lucena, Rio Grande do Sul, Brazil. Affected calves, after one month of age, showed progressive difficulty to follow their mother, retarded growth, muscular weakness and tremors, lethargy and poor body condition. All affected calves were sired by the same bull. Necropsy was performed on three affected calves. The only gross lesion detected was paleness of the skeletal muscles of the trunk and limbs. Cytoplasmic vacuoles, the main histological lesion, were particularly evident in skeletal muscles, myocardium and Purkinje fibers, in neurons of the brain and spinal cord. Large amounts of periodic acid Schiff (PAS) positive granules were also observed in these most severely affected tissues. Pretreatment with diastase completely abolished the PAS reactivity. The 1057?TA, a lethal mutation in the gene of the acid alpha-glucosidase, which causes generalized glycogenosis in Brahman cattle, was detected by PCR in paraffin embedded tissues of affected animals on which post-mortem examination was performed. Clinical, histological and molecular findings were similar to previous descriptions of generalized glycogenosis in Brahman cattle in Australia. No previous indexed reports about generalized glycogenosis of Brahman cattle in Brazil could be found.

In 9 instances (60%) the disease occurred as sporadic cases affecting 1-3 cattle whereas in six farms (40%) MCF occurred as epizootics involving several cattle in each affected herd. Morbidity rates ranged from 2.4% to 20% and lethality rates were 83.3% and 100%. Cattle of all ages and both sexes were affected. Where the information was available (9 farms) sheep were in contact with affected cattle and cases of MCF occurred more frequently in spring and summer. Clinical courses were acute or subacute and clinical signs included fever, nasal and ocular discharges, conjunctivitis, drooling, hematuria, necrosis and blunting of buccal papillae, enlargement of lymph nodes, diarrhea and neurological disturbances. Necropsy findings included opaque corneas, reddening, erosions and ulcerations in several mucous membranes of the alimentary, respiratory and urogenital tracts, and the conjunctiva; enlargement and haemorrhage of lymph nodes and multiple white foci in the renal cortices and in the hepatic portal triads. Crustous dermatitis was observed in some cases. Main histopathological findings included vasculitis, necrosis of the surface epithelia and accumulation of inflammatory cells in several organs. Vasculitis were associated with fibrinoid necrosis of the medial layer of arteries and inflammatory cells included lymphoblasts, lymphocytes, plasma cells and macrophages.

Pesquisa Veterinária Brasileira 25(1):15-20. Departamento de Patologia Clínica Veterinária, Faculdade de Veterinária, Universidade Federal do Rio Grande do Sul, Porto Alegre, RS 91540-000, Brazil. E-mail: alseitz@terra.com.br. Seven sheep received dry crushed Sida carpinifolia L.f. One of them died at 18 and other at 53 days of the experiment. Four others were euthanatized and necropsied at 30, 45, 75 and 100 days. For one sheep the supply of S. carpinifolia was interrupted on the 80th day of the experiment, and 70 days later the animal was euthanized and necropsied. The minimal amount of the dry plant consumed was 11 g/kg and the maximum was 30 g/kg. The progression of clinical findings was similar in six animals with slight diarrhea at 20 days of experiment. Neurological signs were observed at 25 days and included ataxia with dysmetria, muscle tremors of the head, atypical postural reactions, frequent falls, sluggish of movements, difficulty in grazing and swallowing. These signs were enhanced when the animals were forced to walk. Four of the animals presented progressive emaciation. The sheep whose supply of the plant was interrupted recovered gradually, and 11 days after the animal returned to normal. During necropsy, only enlarged mesenteric lymph nodes were observed. The histological alterations were more significant in the central nervous system, with multiple and severe cytoplasmic distention and vacuolation which affects specially Purkinje cells of the cerebellum, neurons of cerebral cortex, thalamus, midbrain and the ventral horn of spinal cord. Axonal spheroids in the brain, more frequently in the granular layer of cerebellum were also observed. The cytoplasmic vacuolation was also found in pancreatic acinar cells, renal tubules, thyroid follicular epithelium, hepatocytes and macrophages of lymphoid organs. The ultrastructural lesions observed were cytoplasmic vacuolation, some surrounded by membranes in Purkinje cells of cerebellum and thyroid follicular cells. The sheep, which had S. carpinifolia withdrawn from its diet for 70 days, had no significant histological alterations.

The course of the disease was acute or subacute. The most important post-mortem findings were enlarged lymphnodes, non-collapsed lungs, with consolidated areas mainly in the cranial lobes. Histological lesions consisted mainly of lymphohistiocytic infiltration with multinucleate giant cells in lymph nodes, spleen, Peyer’s patches, kidney, lung and liver, depletion or lymphoid hyperplasia, as well as lymphohistiocytic interstitial pneumonia and areas of secondary bronchopneumonia. The diagnosis was established through observations of the symptoms and typical lesions, and was confirmed by immunohistochemical examination and PCR. The objective of this study was to characterize the epidemiological, clinical and pathological aspects of the outbreak of PMWS, because of the severe direct or indirect economical losses caused by the disease to the world pig industry.

[Swine aflatoxicosis outbreak in the state of Rio Grande do Sul, Brazil.] Surto de aflatoxicose em suínos no Estado do Rio Grande do Sul. Pesquisa Veterinária Brasileira 24(4):207-210. The corn, which contained the micotoxin and has been used to feed the animals, was produced and processed on the farm. Seven sows and 8 growing pigs died, and 2 cases of abortion were reported. The clinical signs observed were apathy, anorexia, icterus, yellowish urine stained with blood, and photosensitization. Gamma glutamyltransferase and total bilirubin were elevated in the 4 pigs on which post-mortem examination was done. The main macroscopic findings were generalized icterus, an orange-yellow liver, edema of the gall bladder wall and yellowish effusion in the abdominal and pericardial cavities. The most important microscopic changes were found in the liver and included tumefaction, degeneration and individual necrosis of the parenchyme, with ductal proliferation and cholestasis. The diagnosis was based on the clinical signs, macroscopic and histological findings and on the high levels of B1 aflatoxin found in the corn and ration.

Petechial hemorrhages in serosal membranes and edema in the gall bladder were frequently seen. Pale kidneys with red spots in the cortex were observed in one animal. Microscopically, the most striking lesion in the liver was massive coagulative necrosis, associated with centrolobular haemorrhages, observed in four animals. In the liver of one bovine centrolobular necrosis was observed . Tubular renal necrosis was noted in two animals. Additional microscopic lesions were found in the central nervous system of five bovines, especially in the frontal cortex, and included perineuronal and perivascular edema with basophilia and retraction of the neurons. E-mail: davetpat@vortex.ufrgs.br The study assessed differences of pathogenicity among 19 strains of Brachyspira pilosicoli isolated from cases of diarrhea in swine in the State of Rio Grande do Sul, Brazil, using an one-day-old chick model. The chicken inoculated with live bacterial cultures were submitted to euthanasia 21 days later, and the cecum was examined histologically using hematoxylin-eosin, silver staining and immunohistochemistry with a polyclonal antibody anti-Brachyspira pilosicoli. With silver staining and immunohistochemistry, respectively, 21.59% and 70.96% of the chicken showed colonization of the cecal epithelium with B. pilosicoli. Differences in the type of colonization characterized by continuous adhesion, focal adhesion or free bacteria in the intestinal lumen were observed. Immunohistochemistry was more efficient to assess intestinal colonization, because 49.37% more colonized chicken could be detected with silver staining. With three strains, elongated figures inside the cytoplasm of epithelial cecal cells in the inoculated chicken were observed.

The outbreak occurred from January to March of 2004, in Lagoa Vermelha county, state of Rio Grande do Sul, Brazil. The poisoning was experimentally reproduced by oral administration of E. deciduum fruits to 5 sheep.Three of them developed neurological signs and died. Poisoning was caused with a single dose of 60g/kg or when the dosage was fractioned into at least 4 doses of 17 g/kg given every 12 hours. The main clinical signs in natural and experimentally poisoned sheep were ataxia, hyperexcitability and muscular tremors which where more pronounced when the animals were moved. At the final stage, dyspnea with abdominal breathing and cyanosis was observed. The most significant alterations found at necropsy were pronounced edema, lung congestion and presence of the fruits or seeds of E. deciduum in the ruminal content of all animals. Histologically, except edema and pulmonary congestion, there were no other significant changes observed.

Four received the leaves in fresh green stage and the fifth dried ones. Clinical signs were observed in four of the bovines that died; the fifth did not show signs of poisoning. The fresh green plant was proved to be toxic from a dose of 25g/kg on. Dried leaves fed at the dose of 30 g/kg were also toxic. All the animals that died showed clinical signs from 13h30min to 45h after the ingestion of the plant and and death followed within about 48h. The clinical course lasted for about 8h30min until death. The main symptoms where apathy, anorexia, slight tenesmus, muscle trembling, difficulties to keep consciousness, pressing the head against obstacles, lateral recumbency, paddling movements, coma and death. The most significant macroscopic alterations were observed in the liver, with accentuation of the lobular pattern, dark-red areas interspersed with yellowish areas. Petechiae were found in serosal membranes of the abdominal and thoracic organs as well as the intestines. The main microscopic change was hepatic centrolobular coagulative necrosis, associated with congestion and hemorrhages.

These two isolates are representative of the BHV-1 subtypes prevalent in Brazil. Viral subtypes were confirmed by monoclonal antibody analysis and by restriction enzyme digestion of viral genomes. The viruses were inoculated intranasally into seven 3 months old calves (four with BHV-1.1, three with BHV-1.2a). Three other calves of identical age and condition were kept as uninfected controls. In both groups of infected calves, the clinical signs observed were consistent with typical infectious bovine rhinothracheitis (IBR), including pyrexia, apathy, anorexia, nasal and ocular mucopurulent discharges, erosions on the nasal mucosa, conjunctivitis, lachrymation, redness of nasal mucosa, dyspnoea, coughing, tracheal stridor and enlargement of retropharingeal, submandibular and cervical lymphnodes. No significant differences were observed between the clinical scores attributed to both groups. Virus shedding in nasal and ocular secretions were also similar, apart from a significant difference in nasal virus shedding on day 1 to 3 post-inoculation, which was higher for BHV-1.1 than for BHV-1.2a. Following corticosteroid induced reactivation of the latent infection, recrudescence of clinical signs was also observed, with no significant differences on both groups. It was concluded that both subtypes BHV-1.1 and BHV-1.2a were able to induce clinically undistinguishable respiratory disease in calves, either subsequent to a primary infection or following reactivation.
