#21 - Clinical and anatomopathological findings of lipid-related lesions in wild and pet birds from the State of Paraíba, Northeastern Brazil

Abstract in English:

Diseases related to lipid metabolism disorders are reported in several orders of birds, especially in psittacines, and include obesity, atherosclerosis, hepatic lipidosis, egg yolk coelomitis, lipomas, liposarcomas, xanthomas and xanthogranulomas. This study describes epidemiological, clinical and anatomopathological aspects of 28 cases involving lipid-related lesions in wild and pet birds from Northeastern Brazil. The cases were selected from 313 avian patients referred to the UFPB Veterinary Hospital from 2018 to 2022. Lipid-related tumors were the most frequent lesions, followed by obesity, hepatic steatosis, egg yolk coelomitis, atherosclerosis and lipemia. The Psittaciformes order was the most affected, and an erroneous diet (excess consumption of sunflower seeds, bread, crackers, rice, etc.) was identified as the main risk factor for the development of these lesions, which reinforces the need to warn bird owners and caretakers about the importance of adequate nutrition. Forty-nine percent of the reported tumors were in the pericloacal region, which makes lipomas and xanthomas an important differential diagnosis for nodules in this location of birds, especially psittacids.

Doenças relacionadas à desordens no metabolismo dos lipídeos são reportadas em diversas ordens de aves, especialmente em piscítacídeos e incluem obesidade, aterosclerose, lipidose hepática, celomite por gema de ovo, lipomas, lipossarcomas, xantomas e xantogranulomas. Este estudo descreve aspectos epidemiológicos, clínicos e anatomopatológicos de 28 casos envolvendo lesões relacionadas a lipídeos em aves silvestres e pet no Nordeste Brasileiro. Os casos foram selecionados de um total de 313 aves atendidas entre 2018 e 2022 no Hospital Veterinário da UFPB. Tumores relacionados a lipídeos foram as lesões mais relatadas, seguido por obesidade, esteatose hepática, celomite por gema de ovo, aterosclerose e lipemia. A ordem Piscitaciforme foi a mais acometida e dieta errônea (Excesso de sementes de girassol, pão, bolacha, arroz, etc.) foi identificado como o principal fator de risco para o desenvolvimento das lesões, o que reforça a necessidade de advertir os tutores quanto a importância da alimentação adequada. Quarenta e nove por cento dos tumores relatados estavam na região pericloacal, o que torna lipomas e xantomas um diagnóstico diferencial importante para nódulos nessa localização em aves, principalmente psitacídeos.


#22 - Bovine genital leptospirosis: Findings in bulls maintained in Caatinga biome conditions

Abstract in English:

Leptospirosis is a disease that causes economic and social impact, as it affects wild and domestic animals and humans. There may be peculiarities in the epidemiology of this disease in the Caatinga biome, Brazil, where the environment is adverse and the etiologic agent, Leptospira spp., requires alternative transmission routes. Considering that in bovine leptospirosis the genital carrier is constantly neglected and the lack of reports on the role of bulls in the epidemiology of the bovine genital leptospirosis (BGL) syndrome, mainly in semiarid conditions such as Caatinga biome, this study aimed to investigate bulls maintained in Caatinga biome conditions as genital carriers of leptospires. Urinary tract (urine, bladder, and kidney) and reproductive tract (vas deferens, cauda epididymis, and vesicular gland) samples were collected from 42 slaughtered bulls. Microscopic agglutination test (MAT), polymerase chain reaction (PCR), and microbiological isolation were included as diagnostic methods. Anti-Leptospira spp. antibodies were found in 17 (40.48%) animals, while 26 animals (61.90%) had at least one organ or urine with leptospiral DNA, and 10 animals (23.81%) were positive at bacteriological culture. Sequenced samples targeting the LipL32 gene showed 99% similarity with Leptospira borgpetersenii. Molecular analysis of samples from the vas deferens and cauda epididymis is recommended for the diagnosis of genital leptospirosis in bulls and, if it is impossible to collect these tissues, semen can be used. In conclusion, this study provides important information relating to bulls from the Caatinga biome, Brazil, as carriers of genital leptospirosis. The results indicate that, even under adverse environmental conditions, leptospires may survive and propagate, mainly due to the characteristic of genital carriers for the sexually spreading of adapted Leptospira species without influence by external variables. Thus, prevention and control strategies for bovine leptospirosis need to include actions aimed at the genital carrier.

A leptospirose é uma doença que causa impacto econômico e social, pois afeta animais silvestres, domésticos e humanos. É possível que existam peculiaridades na epidemiologia desta doença no bioma Caatinga, Brasil, onde o ambiente é adverso e o agente etiológico, Leptospira spp., requer vias alternativas de transmissão. Considerando que na leptospirose bovina o portador genital é constantemente negligenciado e a falta de relatos sobre o papel dos touros na epidemiologia da síndrome da leptospirose genital bovina (BGL), principalmente em condições semiáridas como no bioma Caatinga, este estudo teve como objetivo investigar touros mantidos em condições do bioma Caatinga como portadores genitais de leptospiras. Amostras do trato urinário (urina, bexiga e rim) e do trato reprodutivo (ducto deferente, cauda do epidídimo e glândula vesicular) foram coletadas de 42 touros abatidos. Teste de soroaglutinação microscópica (SAM), reação em cadeia da polimerase (PCR) e isolamento microbiológico foram incluídos como métodos de diagnóstico. Anticorpos anti-Leptospira spp. foram encontrados em 17 (40,48%) animais, enquanto 26 animais (61,90%) apresentaram pelo menos um órgão ou urina com DNA leptospírico, e 10 animais (23,81%) foram positivos na cultura bacteriológica. As amostras sequenciadas a partir do gene LipL32 apresentaram 99% de similaridade com Leptospira borgpetersenii. Recomenda-se a análise molecular de amostras de ducto deferente e cauda do epidídimo para o diagnóstico de leptospirose genital em touros e, na impossibilidade da coleta desses tecidos, o sêmen pode ser utilizado. Em conclusão, este estudo fornece informações importantes sobre touros do bioma Caatinga, Brasil, como portadores de leptospirose genital. Os resultados indicam que, mesmo em condições ambientais adversas, as leptospiras podem sobreviver e se propagar, principalmente devido à característica de os portadores genitais disseminarem sexualmente espécies adaptadas de Leptospira sem influência de variáveis externas. Assim, as estratégias de prevenção e controle da leptospirose bovina precisam incluir ações voltadas para o portador genital.


#23 - Diseases of the digestive system of agoutis (Dasyprocta leporina) raised in captivity in the Brazilian semiarid region

Abstract in English:

The objective of this study was to describe the clinical and pathological aspects of diseases of the digestive system in agoutis (Dasyprocta leporina Linnaeus, 1758) diagnosed by the “Laboratório de Patologia Veterinária” (Veterinary Pathology Laboratory) of the “Universidade Federal Rural do Semi-Árido” (UFERSA), from January 2018 to February 2020. During the study period, necropsy and a survey of the clinical history of 27 agoutis were performed, 25.93% (7/27) of which were diagnosed with digestive system diseases. The percentages of digestive tract diseases among the diagnosed were: acute carbohydrate overload (11.12%), gastric ulcer (7.41%), gastric volvulus (3.70%), and intestinal volvulus (3.70%). Studies on the occurrence rate of these diseases, as well as the description of their clinical and anatomopathological aspects, may serve as a basis for guiding the appropriate management in the breeding of these animals.

O objetivo deste estudo foi descrever os aspectos clínicos e patológicos das doenças do aparelho digestivo em cutias (Dasyprocta leporina Linnaeus, 1758) diagnosticadas pelo Laboratório de Patologia Veterinária da Universidade Federal Rural do Semiárido (UFERSA), de janeiro 2018 a fevereiro de 2020. Durante o período do estudo, foram realizadas necropsias e levantamento da história clínica de 27 cutias, sendo 25,93% (7/27) diagnosticadas com doenças do aparelho digestivo. Os percentuais de doenças do aparelho digestivo foram: sobrecarga aguda de carboidratos (11,12%), úlcera gástrica (7,41%), vólvulo gástrico (3,70%) e vólvulo intestinal (3,70%). Estudos sobre a taxa de ocorrência dessas doenças, bem como a descrição de seus aspectos clínicos e anatomopatológicos, podem servir de base para orientar o manejo adequado na criação dessa espécie.


#24 - Could dysregulation of RASSF1 expression be a mechanism of tumorigenesis in CTVT?

Abstract in English:

Canine transmissible venereal tumor (CTVT) is the oldest known somatic cell lineage. It is a transmissible cancer that propagates naturally in dogs and reportedly contains gene mutations. RASSF1 participates in DNA damage repair, and its downregulation, results in tumor progression. Hence, RASSF1 is a tumor suppressor gene. Its expression was quantified in tumors from seventeen animals and three cell cultures derived from tumors. In general, RASSF1 was underexpressed in 65%, and absent in 35% of tumor samples. Cells from tumor tissue cultures showed decreased expression of RASSF1 in 67% and elevated expression in 33% of samples tested. The tumor tissues showed significantly lower levels of RASSF1 expression compared to cultured cells. Previously we reported that both the tumor microenvironment and the host immune system appear to influence the tumorigenesis and stage of CTVT. This is the first article to demonstrate the expression of RASSF1 in CTVT. Decreased RASSF1 possibly helps tumor progression.

O tumor venéreo transmissível canino (TVTC) é a linhagem de células somáticas mais antiga conhecida. É um câncer transmissível que se propaga naturalmente em cães e mutações genéticas já foram relatadas. O gene RASSF1 atua no reparo de danos ao DNA e presume-se que, quando suprimido ou com expressão gênica reduzida, o TVTC tende a progredir. A expressão do gene supressor de tumor, como RASSF1, foi quantificada em tecidos de dezessete animais e três culturas de células de tecidos tumorais. Em geral, o gene RASSF1 apresentou prevalência de subexpressão (65%) e ausência em 35% dos demais tecidos analisados. Células isoladas de culturas de tecidos tumorais também demonstraram 67% com expressão diminuída e 33% com expressão elevada, com diferença significativa entre os níveis de expressão gênica em amostras de tecido quando comparadas às culturas de células, com tecidos apresentando níveis mais baixos de expressão gênica em comparação com células. Anteriormente, relatamos que tanto o microambiente tumoral quanto o sistema imunológico do hospedeiro parecem influenciar a tumorigênese e o estágio do TVTC. Este é o primeiro artigo a demonstrar a expressão de RASSF1 no TVTC, possivelmente alterando sua tumorigênese e auxiliando no aumento da progressão tumoral.


#25 - Bovine tuberculosis in safari park in Brazil

Abstract in English:

Bovine tuberculosis (bTB) is an infectious disease caused by Mycobacterium bovis, affecting domestic animals, wild animals and humans. In captivity, for wild animals, bTB represents a risk to animal keepers and zoo visitors, in addition to the possibility of spreading the infection to domestic animals or through the trade of infected wild animals. Sambar (Cervus unicolor), red deer (Cervus elaphus) and fallow deer (Dama dama) from a safari park in the State of Rio Grande do Sul, Brazil, showed a clinical condition of dyspnea and weight loss. Some animals died and showed lesions suggestive of tuberculosis (LST), which were confirmed by histopathology. After the interdiction of the safari park by the state veterinary authorities, 281 deer were euthanized with the authorization of the “Instituto Brasileiro do Meio Ambiente e dos Recursos Naturais Renováveis” (IBAMA). Retropharyngeal and submandibular lymph nodes and viscera were collected from 21 animals, which were grown in Stonebrink medium for up to 90 days. After DNA extraction from the bacterial colonies, PCR was performed for targets flanking the region of differentiation 4 (RD4). Of the 21 samples, 14 (66.7%) presented LST with a granulomatous appearance, a whitish coloration, and caseous or calcified consistency, and seven samples (33.3%), showed no lesions. In the culture of 14 samples with LST, 13 (92.8%) presented bacterial growth compatible with M. bovis. In the cultivation of the seven samples without LST, four (57.1%) presented colonies compatible with M. bovis. PCR and DNA sequencing of the PCR amplicons detected as positive all the 17 (100%) bacteriological cultures suggestive of M. bovis, thus confirming the outbreak of bTB in deer. Decisions about positive tested and suspicious animals should be taken based on the evaluation of the risk of transmission to the rest of the zoological animals, animal welfare, conservation considerations and, the zoonotic potential of this pathogen.

A tuberculose bovina (bTB) é uma doença infecciosa causada por Mycobacterium bovis, afetando animais domésticos, animais selvagens e humanos. Para animais selvagens em cativeiro, a bTB representa um risco para os tratadores de animais e visitantes do zoológico, além da possibilidade de espalhar a infecção para animais domésticos ou por meio do comércio de animais silvestres infectados. Cervídeos sambar (Cervus unicolor), veado-vermelho (Cervus elaphus) e gamo (Dama dama) de um parque safári no Estado do Rio Grande do Sul, Brasil, mostraram uma condição clínica de dispneia e perda de peso. Alguns animais morreram e apresentaram lesões sugestivas de tuberculose (LST), as quais foram confirmadas por histopatologia. Após a interdição do parque safári pelas autoridades veterinárias estaduais, 281 veados sofreram eutanásia com a autorização do Instituto Brasileiro do Meio Ambiente e dos Recursos Naturais Renováveis (IBAMA). Os linfonodos retrofaríngeos e submandibulares e vísceras foram coletados de 21 animais, que foram cultivados em meio Stonebrink por até 90 dias. Após extração de DNA das colônias bacterianas, foi realizada PCR para alvos que flanqueavam a região de diferenciação 4 (RD4). Das 21 amostras, 14 (66,7%) apresentaram LST com aspecto granulomatoso, coloração esbranquiçada e consistência caseosa ou calcificada, e sete amostras (33,3%) não apresentaram lesões. Na cultura de 14 amostras com LST, 13 (92,8%) apresentaram crescimento bacteriano compatível com M. bovis. No cultivo das sete amostras sem LST, quatro (57,1%) apresentaram colônias compatíveis com M. bovis. A PCR e o sequenciamento de DNA dos fragmentos de PCR detectaram como positivo todas as 17 (100%) culturas bacteriológicas sugestivas de M. bovis, confirmando assim o surto de bTB em cervídeos. As decisões sobre animais positivos testados e suspeitos devem ser tomadas com base na avaliação do risco de transmissão para o restante dos animais zoológicos, bem-estar animal, considerações de conservação e no potencial zoonótico desse patógeno.


#26 - Clinical and post mortem examination of white worm lizards (Amphisbaena alba) in the State of Paraíba, Northeastern Brazil: morphological, pathological and radiographic findings of a secretive species

Abstract in English:

Amphisbenians are limbless reptiles that belong to the order Squamata. Due to their fossorial and secrevie habits, little is known about their morphology, ecology and pathological conditions that may affect them. In this manuscript, we present a brief guide for identification of normal structures as well as traumatic injuries on radiography and necropsy of Amphisbaena alba. From April to September 2019, three cases of A. alba with suspected trauma were referred to the Veterinary Hospital of the Federal University of Paraíba (UFPB). In the clinical evaluation, traumatic injuries were observed, and support therapy was instituted, but they did not resist and died shortly after. Bone fractures and organ ruptures, in addition to specific structures of this species were identified on radiography. A systematic necropsy was performed of all amphisbaenians in order to evaluate external and internal structures, not only to identify lesions but also to investigate the morphological aspects of amphisbenids. Macroscopically, multiple organ fractures and ruptures observed in radiographs were confirmed, in addition to the presence of the cestodes Semenoviella amphisbaenae in the large intestine. Histologically, it was possible to identify normal characteristics and microscopic lesions in the tissues. This is the first study to incorporate morphological, clinical, and pathological aspects of A. alba. This manuscript brings essential information for wildlife veterinarians and pathologists who may have to treat or perform a necropsy on these unique reptiles.

Anfisbenas são répteis desprovidos de membros que pertencem a ordem Squamata. Devido a seus hábitos fossoriais e reclusos, pouco se conhece sobre sua morfologia, ecologia e condições patológicas que possam afetá-las. Neste artigo, um guia sucinto para a necropsia, identificação de tecidos e órgãos e lesões encontradas em Amphisbaena alba, é apresentado. Durante março e dezembro de 2019, três casos de A. alba com suspeita de trauma foram encaminhados ao Hospital Veterinário da Universidade Federal da Paraíba. Na avaliação clínica foram observadas lesões traumáticas e instituído um suporte terapêutico, porém não resistiram e morreram em seguida. Foram realizadas projeções radiográficas onde identificaram fraturas e ruptura de órgãos, além de identificar estruturas específicas dessa espécie. Uma avaliação completa de estruturas externas e internas foi conduzida para investigar aspectos morfológicos dos anfisbenídeos. Macroscopicamente foram confirmadas múltiplas fraturas e rupturas de órgãos observadas anteriormente nas radiografias, além disso evidenciou no intestino grosso presença de parasitos anoplocéfalos denominados Semenoviella amphisbaenae. Na histologia foi possível identificar características normais e lesões microscópicas nos tecidos. Esse é o primeiro estudo a incorporar aspectos morfológicos, clínicos e patológicos de A. alba. Esse manuscrito traz informações essenciais para clínicos e patologistas de animais selvagens que podem se deparar com casos clínicos ou de necropsia de Amphisbaena alba.


#27 - Isolation and genotyping of Mycobacterium bovis in suggestive lesions of tuberculosis in cattle slaughtered in the state of Ceará, Brazil

Abstract in English:

Bovine tuberculosis (BTB) is a zoonosis caused by the bacterium Mycobacterium bovis, which induces the development of nodular and granulomatous lesions in various animal tissues. The recognition of these suggestive gross lesions during postmortem sanitary inspection in slaughterhouses provides a presumptive diagnosis, which requires the use of complementary tests to confirm the disease. This study aimed to verify the occurrence of BTB in cattle slaughtered in slaughterhouses in the state of Ceará, Brazil, using bacteriological and molecular methods. To this end, suggestive lesions were analyzed on carcasses condemned by the “Serviço de Inspeção Estadual” (SIE). The samples were submitted to microbiological analysis using culture media and specific staining followed by spoligotyping molecular technique for identification and genotyping of the mycobacteria. Occurrence of lesions suggestive of BTB was verified in bovine carcasses (0.071%) from different municipalities of the state. These lesions were located mainly in the lung (95.12%), lymph nodes (58.53%), and liver (36.58%). Microbiological culture showed bacterial isolation (17.94%), with the growth of colonies showing morphological and tannic characteristics belonging to genus Mycobacterium spp. Genetic polymorphism analysis identified M. bovis in all isolates, which were discriminated into six spoligotypes (SB0121, SB0295, SB1064, SB0120, SB0870, and SB0852). These profiles have been described in Brazil and several areas of the world, except for profiles SB1064 and SB0852, which were described in the country for the first time. The results show that the association of the diagnostic methods used was the basis for the first study on identification of mycobacteria found in the state, which may provide a database for the epidemiological study of BTB in the state of Ceará.

A tuberculose bovina (TB) é uma zoonose causada pelo Mycobacterium bovis, o qual induz ao desenvolvimento de lesões nodulares e granulomatosas em vários tecidos do animal. O reconhecimento dessas lesões macroscópicas sugestivas durante a inspeção sanitária post mortem em matadouros fornece um diagnóstico presuntivo, sendo necessário a utilização de testes complementares para confirmação da doença. O objetivo deste trabalho foi verificar a ocorrência da TB em animais abatidos em matadouros-frigoríficos no estado do Ceará através da utilização de métodos bacteriológicos e moleculares. Para tanto, foram analisadas lesões sugestivas de TB em carcaças condenadas pelo Serviço de Inspeção Estadual (SIE). As amostras foram submetidas à análise microbiológica, utilizando meios de cultivo e de coloração específicos, seguida pela técnica molecular spoligotyping para identificação e tipificação genética da micobactéria. Verificou-se a ocorrência de lesões sugestivas de TB em carcaças bovinas (0,071%) oriundas de diferentes municípios do estado do Ceará. Essas lesões estavam localizadas principalmente no pulmão (95,12%), linfonodos (58,53%) e fígado (36,58%). O cultivo microbiológico obteve isolamento bacteriano (17,94%), com o crescimento de colônias apresentando características morfológicas e tintoriais pertencentes ao gênero Mycobacterium spp. A análise do polimorfismo genético identificou a presença de M. bovis em todos os isolados, que foram discriminados em seis espoligotipos (SB0121, SB0295, SB1064, SB0120, SB0870 e SB0852), descritos no Brasil e em diversas áreas do mundo, exceto os perfis SB1064 e SB0852 que foram descritos pela primeira vez no país. Os resultados obtidos demonstram que a associação dos métodos diagnósticos utilizados foram a base do primeiro estudo de identificação das micobactérias encontradas no estado do Ceará, o que pode contribuir para a criação de um banco de dados para o estudo epidemiológico da TB no estado.


#28 - Morphological description of ovary and uterus of the nurse shark (Ginglymostoma cirratum) caught off at the Fortaleza coast, Northeast Brazil

Abstract in English:

The nurse shark, Ginglymostoma cirratum (Bonnaterre, 1778) is one of the most studied species of elasmobranchs. However, the knowledge of their reproductive biology is still relatively rare, particularly in the western South Atlantic. This study aimed to describe the morphology of the uterus and the ovary of G. cirratum, based on specimens caught off at the Fortaleza/CE coast, northeast Brazil. Samples were collected from September 2012 to June 2013, from regular landings of artisanal fishing, which commercialize this species freely. A total of ten females were collected. The methodologies followed for analyzing the ovaries and uterus of those females included both macroscopic and histological analysis. G. cirratum has internal type ovary morphology, with invaginations of connective tissue, which defines compartments and separate oocyte groups in ovigerous lots. The epithelium lining the ovary changes from simple columnar ciliated in the area without ovigerous lots, which turns into a simple cubic epithelium in the coating portion of the epigonal organ where ovarian tissue is absent. The uterine mucosa has secretory cells denoted by Alcian Blue staining, indicating the production of mucopolysaccharides, even in immature individuals. This lecithotrophic shark has a uterine vascularized mucosa that is one characteristic of viviparous elasmobranch species.

O tubarão-lixa, Ginglymostoma cirratum (Bonnaterre, 1778) é uma das espécies mais estudadas de elasmobrânquios. O conhecimento de sua biologia reprodutiva, no entanto, ainda é relativamente raro, particularmente no Oeste do Atlântico Sul. Este estudo objetivou a descrição morfológica do útero e ovário de G. cirratum, baseado em espécimes capturados na costa de Fortaleza/CE, no Nordeste do Brasil. Um total de dez fêmeas foi coletado de setembro de 2012 a junho de 2013, nos desembarques regulares da pesca artesanal, que comercializam essa espécie livremente. A metodologia foi de acordo com as analises macroscópica e histológica dos ovários e útero. G. cirratum tem o tipo de morfologia de ovário interno, com invaginações de tecido conjuntivo, que define compartimentos e separa grupos de oócitos em lotes ovígeros. O epitélio que reveste o ovário varia de epitélio simples colunar ciliado na área que não apresenta lotes ovígeros para epitélio simples cúbico na porção de revestimento do órgão epigonal, onde o tecido ovariano é ausente. A mucosa uterina possui células secretoras, evidenciadas pela coloração em Alcian Blue, indicando a produção de mucopolissacarídeos, mesmo em indivíduos imaturos. Este tubarão lecitotrófico apresenta a mucosa uterina vascularizada, sendo esta uma característica de espécies de elasmobrânquios vivíparos.


#29 - A taste of pathology

Abstract in English:

Adult learning, or andragogy, provides a novel way of appreciating using food analogies as an effective learning tool in veterinary pathology. Facilitation of adult learning requires that new concepts be presented in a way that draws on the learner’s experience. Because veterinary students will have had considerable experience with a plethora of food items prior to enrolling in a pathology course, food analogies can provide an easy conduit for incorporating key learning concepts regarding veterinary pathology. In this paper, many of these analogies are presented, along with the mechanisms responsible for each of the characteristic lesions, in the hopes that their usefulness in the classroom can be highlighted to create a more engaging and facilitated learning environment.

A aprendizagem de adultos, ou andragogia, é uma nova maneira de apreciar o uso de analogias de alimentos como uma ferramenta eficaz no aprendizado em patologia veterinária. A facilitação da aprendizagem de adultos requer que novos conceitos sejam apresentados de uma forma que se baseie na experiência do aluno. Como os estudantes de medicina veterinária já terão sido expostos a um número considerável de tipos de alimentos antes de se matricularem na disciplina de patologia, as analogias de alimentos podem fornecer um ótimo conduto para incorporar os conceitos-chave na aprendizagem da patologia veterinária. Neste artigo, muitos dessas analogias são apresentadas juntamente com os mecanismos responsáveis por cada uma das lesões características, na esperança de que sua utilidade na sala de aula possa ser destacada para criar um ambiente de aprendizado mais envolvente e favorável.


#30 - Hepatoprotective and antineoplastic potencial of red propolis produced by the bees Apis mellifera in the semiarid of Rio Grande do Norte, Brazil

Abstract in English:

The objective of this study was to evaluate the hepatoprotective effect of the honey bee Apis mellifera ethanolic extract of the red propolis, obtained in four municipalities of the Rio Grande do Norte semi-arid region, through an in vitro evaluation of the antineoplastic potential in human hepatic carcinoma (HepG2) and normal cell lines (L929), and from the comet assay in hepatic cell lines (ZF-L hepatocytes) to evaluate the genoprotective potential of the extract. The hepatoprotective effect was also evaluated in vivo by the induction of chronic experimental hepatic lesions in rodents (Rattus norvegicus Berkenhout, 1769), Wistar line, by intraperitoneal administration of thioacetamide (TAA) at the dose of 0.2g/kg. The animals were distributed in the following experimental groups: G1 (control), G2 (treated with 500mg/kg ethanolic extract of propolis), G3 (treated with 500mg/kg of ethanolic extract and TAA) and G4 (treated with TAA). All rats were submitted to serum biochemical, macroscopic, histological and stereological biochemical exams of the liver. It was verified the genoprotective effect of red propolis since the mean damages promoted to DNA in cells tested with the extract were significantly lower than the mean of the positive control damage (hydrogen peroxide). The red propolis extract did not present cytotoxic activity to the tumor cells of human liver cancer, as well as to normal ones. The absence of cytotoxicity in normal cells may indicate safety in the use of the propolis extract. The results of the serum biochemical evaluation showed that the serum levels of the aminotransferase enzymes (AST) did not differ significantly between G1, G2 and G3 when compared to each other. G4 showed significant increase in levels compared to the other groups, indicating that the administration of the extract did not cause liver toxicity, as well as exerted hepatoprotective effect against the hepatic damage induced by TAA. The G3 and G4 animals developed cirrhosis, but in G3 the livers were characterized by the presence of small regenerative nodules and level with the surface of the organ, whereas in G4 the livers showed large regenerative nodules. The livers of the G1 and G2 animals presented normal histological appearance, whereas the livers of the G3 animals showed regenerative nodules surrounded by thin septa of connective tissue, and in G4 the regenerative nodules were surrounded by thick septa fibrous connective tissue. The analysis of the hepatic tissues by means of stereology showed that there was no statistical difference between the percentage of hepatocytes, sinusoids, and collagens in G1 and G2. In G3 the percentage of hepatocytes, sinusoids, and collagen did not differ significantly from the other groups. It was concluded that the ethanolic extract of the red propolis exerted a hepatoprotective effect, because it promoted in vitro reduction of the damage to the DNA of liver cells, antineoplastic activity in human hepatocellular carcinoma cell line (HepG2) and did not exert cytotoxic effect in normal cells or was able to reduce liver enzyme activity and the severity of cirrhosis induced by TAA in vivo.

Este estudo objetivou avaliar o efeito hepatoprotetor do extrato etanólico da própolis vermelha da abelha Apis mellifera, obtido em quatro municípios do semiárido do Rio Grande do Norte, mediante avaliação in vitro do potencial antineoplásico em linhagens de células de carcinoma hepático humano (HepG2) e em linhagens de células normais (L929), além do ensaio cometa em linhagens de células hepáticas (hepatócitos ZF L) para avaliar o potencial genoprotetor do extrato. O efeito hepatoprotetor também foi avaliado in vivo através da indução de lesões hepática experimental crônica em roedores da espécie Rattus norvegicus (Berkenhout, 1769), linhagem Wistar, pela administração intraperitoneal de tioacetamida (TAA) na dose de 0,2g/kg. Os animais foram distribuídos nos seguintes grupos experimentais: G1 (controle), G2 (tratados com 500mg/kg de extrato etanólico da própolis), G3 (tratados com 500mg/kg de extrato etanólico e TAA) e G4 (tratados com TAA). Todos os ratos foram submetidos aos exames bioquímico sérico, anatomopatológico macroscópico, histológico e esteriológico do fígado. Foi constatado o efeito genoprotetor da própolis vermelha uma vez que as médias dos danos promovidos ao DNA em células testadas com o extrato foram significativamente inferiores à média dos danos do controle positivo (peróxido de hidrogênio). O extrato da própolis vermelha não apresentou atividade citotóxica para células tumorais de câncer de fígado humano, bem como para normais. A ausência de citotoxicidade em células normais, tal como constatado, pode indicar segurança no uso do extrato da própolis. Os resultados da avaliação bioquímica sérica demonstraram que os níveis séricos das enzimas aminotransferase (AST) não diferiram significativamente entre G1, G2 e G3, quando comparadas entre si. No G4 houve aumento significativo dos níveis em relação aos demais grupos, indicando que a administração do extrato não causou toxicidade hepática, bem como exerceu efeito hepatoprotetor frente ao dano hepático induzido pela TAA. Os animais dos G3 e G4 desenvolveram cirrose, porém no G3 os fígados caracterizaram-se pela presença de pequenos nódulos regenerativos e nivelados com a superfície do órgão, enquanto que no G4 os fígados apresentaram grandes nódulos regenerativos. Os fígados dos animais G1 e G2 apresentaram aspecto histológico normal, enquanto que os fígados dos animais do G3 apresentaram nódulos regenerativos circundados por finos septos de tecido conjuntivo, e nos do G4 os nódulos regenerativos foram circundados por espessos septos de tecido conjuntivo fibroso. A análise dos tecidos hepáticos por meio de estereologia mostrou que não houve diferença estatística entre o percentual de hepatócitos, sinusoides e colágenos nos G1 e G2. No G3 o percentual de hepatócitos, sinusoides e colágeno não diferiu significativamente dos demais grupos. Concluiu-se que o extrato etanólico da própolis vermelha exerceu efeito genoprotetor, por promover in vitro redução do dano ao DNA de células hepáticas, atividade antineoplásica em linhagem celular de carcinoma hepatocelular humano (HepG2) e não exerceu efeito citotóxico em células normais ou efeito hepatoprotetor in vivo com diminuição da gravidade da cirrose induzida por TAA.
