E-mail: jpacosta00@hotmail.com The distribution of cells involved in the immune response in accessory sex glands of rams experimentally infected with Actinobacillus seminis was studied. Twelve one-year old rams were experimentally infected by intraurethral (IU) (n=4) and intraepididymal (IE) (n=4) route, and four control (CON) animals were used. The animals were slaughtered 35 days post-inoculation, samples were taken from accessory sex glands, and bacteriology and histopathology tests were performed. The presence of CD4, CD8 and TCRγδ (WC1) lymphocytes, CD45RO cells, macrophages (CD14), dendritic cells (CD1b), IgA-, IgG- and IgM-containing cells (IgCC) was determined. Animals of the IE group developed clinical epididymitis. No lesions were seen in rams of the IU group; two of the intraepididymal inoculated CON developed small lesions in the epididymis. A. seminis isolates were achieved from 6:16 (37.5%) accessory sex glands in the IE group, but not in the IU and CON groups. In the CON group, IgA- and IgM- containing cells predominated in the bulbourethral glands and the disseminated prostate, and they were scarce or null in the vesicles and ampullae. A significant increase of IgA-, IgG- and IgM- containing cells was confirmed in the seminal vesicles, the ampullae and the bulbourethral glands in the IE group. In the IE and IU groups, an increase in CD4, CD8, WC1, CD45RO and CD14 was evidenced in the vesicles and ampullae. CD1b dendritic cells were present in the ampullae and vesicles with inflammatory processes. A. seminis triggered a local immune response in the IE and IU groups. These results indicate a different pattern of infiltrating immune cells in the accessory sex glands of infected A. seminis rams.

E-mail: sandrafigueiredo@ufmt.br Yacare Caiman meat consumption has become a marketing trend and a commodity on the rise in Mato Grosso state in Brazil. In the last four years, cuts from the trunk represented 28.40% of total meat sales. Although evolutionary studies, morphophysiological ontogenetic and technology research have been carried out, characterization of muscle and bone bases of cuts from the torso has not been previously reported. The aim of this research is to describe the muscles and corresponding bones related to sirloin, filet mignon and meat trims cuts. To describe the bones, we used six boned carcasses from juvenile Yacare Caiman, as well as an adult specimen, obtained by donation after death from the Federal University of Mato Grosso Zoo. The bones were macerated, bleached and their anatomical details recorded. In order to study the muscle, 24 juvenile specimens were obtained after slaughter and skinning and dissected on both sides. The sirloin cut consists of the semispinal, longissimus and iliocostalis muscles, which are inserted on thoracic vertebrae and ribs, as well as lumbar and sacral ribs. The meat trims cut is formed by latissimus dorsi, serratus, pectoral and abdominal (external oblique, internal oblique, transversus and rectus) muscles, based in various bones: bone ribs are the thoracic, lumbar, and sacral ribs, the gastralia, the sternum and epipúbis. The filet mignon cut is formed by the internal puboischiofemoralis cranial (sublumbar) muscle and by the troncocaudal (ventral surface of the pelvis) muscle.

O corte aparas é constituído pelos músculos grande dorsal, serrátil, peitoral e abdominais (oblíquo externo, oblíquo interno, transverso e reto), cuja base óssea corresponde as costelas torácicas, lombares e sacrais, a gastrália, o esterno e o epipúbis.

ABSTRACT.- Campos A.P., Miranda D.F.H., Alves G.B.B., Carneiro M.S., Prianti M.G., Gonçalves L.M.F., Castro V. & Costa F.A.L. 2015. The role of alveolar type II cells in swine leptospirosis. Pesquisa Veterinária Brasileira 35(7):620-626. Setor de Patologia Animal, Departamento de Clínica e Cirurgia Veterinária, Universidade Federal do Piauí, Campus da Socopo S/N, Teresina, PI 64.049-550, Brasil. E-mail: angelapiauilino@hotmail.com This study aimed to investigate a possible relationship between alveolar type II cells and the inflammatory response to infection with Leptospira spp., and thus comprise a further element that can be involved in the pathogenesis of lung injury in naturally infected pigs. The study group consisted of 73 adult pigs that were extensively reared and slaughtered in Teresina, Piauí state, and Timon, Maranhão state, Brazil. The diagnosis of leptospirosis was made using the microscopic agglutination test (MAT) aided by immunohistochemistry and polymerase chain reaction. The MAT registered the occurrence of anti-Leptospira antibodies in 10.96% (8/73) of the pigs. Immunohistochemistry allowed for the visualization of the Leptospira spp. antigen in the lungs of 87.67% (64/73) of the pigs. There was hyperplasia of bronchus-associated lymphoid tissue and circulatory changes, such as congestion of alveolar septa, parenchymal hemorrhage and edema within the alveoli. Lung inflammation was more intense (p = 0.0312) in infected animals, which also showed increased thickening of the alveolar septa (p = 0.0006). Evaluation of alveolar type II (ATII) cells using an anti-TTF-1 (Thyroid Transcription Factor-1) antibody showed that there were more immunostained cells in the non-infected pigs (53.8%) than in the infected animals (46.2%) and that there was an inverse correlation between TTF-1 positive cells and the inflammatory infiltrate. There was no amplification of Leptospira DNA in the lung samples, but leptospiral DNA amplification was observed in the kidneys. The results of this study showed that a relationship exists between a decrease in alveolar type II cells and a leptospire infection. Thus, this work points to the importance of studying the ATII cells as a potential marker of the level of lung innate immune response during leptospirosis in pigs.

E-mail: marina_marchesi@hotmail.com The concentration of heavy metals (Cr, Fe, Al, As, Cd, Cu, Pb, Mo, Ni, Se and Zn) was evaluated in the blood of nestling blue macaws (Anodorhynchus hyacinthinus) captured in the Pantanal, Mato Grosso do Sul (n=26) in 2012; this was based on the hypothesis that these birds exhibit levels of these heavy metals in their organism and that these interfere in hatching success, weight and age of the chicks. Blood samples were digested with nitric acid and hydrochloric acid and the quantification of metals was performed by ICP-OES (Optical Emission Spectroscopy and Inductively Coupled Plasma). Blood samples of nestlings showed concentrations of Cr (0.10μg/g) Fe (3.06μg/g) Al (3.46μg/g), Cd (0.25μg/g) Cu (0.74μg/g), Mo (0.33μg/g), Ni (0.61μg/g), Se (0.98μg/g), and Zn (2.08μg/g). The levels of heavy metals found were not associated with weight, age and hatching success of the chicks.

#25 - Hematological profile of Saanen and mixed-breed (½ Saanen x ½ Anglo-Nubian) raised in the tropical climate of Ceará, Brazil, 35(1):99-104

Abstract in English:

ABSTRACT.- Souza P.T., Salles M.G.F., Costa A.N.L., Carneiro H.A.V., Souza L.P., Rocha D.R. & Araújo A.A. 2015. [Hematological profile of Saanen and mixed-breed (½ Saanen x ½ Anglo-Nubian) raised in the tropical climate of Ceará, Brazil.] Paranjana 1700, Campus do Itaperi, Fortaleza, CE 60714-903, Brazil. E-mail: priscilatsvet@yahoo.com.br The aim of this study was to determine the influence of climatic conditions on the hematological profile of Saanen and mixed-breed (½ Saanen x ½ Anglo-Nubian) goats, as well as to define reference values for these animals bred in Ceará, Brazil. Thirty goats were utilized and blood samples were collected monthly during the rainy (February to May) and dry (August to November) periods to obtain an erythrogram a leukogram. The averange were compared by the t-Student test and Mann Whitney test, with parametric and non-parametric distribution of the data, respectively, where p<0.05 was considered significant. A study was carried out of simple Pearson correlations of the hematological parameters with environmental and physiological variables. The number of red blood cells (RBCs) was higher in the Saanen goats and in the rainy period, while the hematocrit was higher in the dry period (p<0.05). The leukocytes and lymphocytes were higher in the ½S½AN goats in the two periods (p<0.05). In the two genotypes, the leukocytes and lymphocytes were higher in the dry season and the segmented neutrophils higher in the rainy season (p<0.05). The other parameters did not differ (p>0.05). The RBCs and segmented neutrophils displayed a negative correlation with air temperature, but positive with relative humidity and rectal temperature (p<0.05). The hematocrit positively correlated with air temperature and respiratory rate (p<0.05). The leukocytes and lymphocytes showed a negative correlation with rectal temperature (p<0.05). It was therefore concluded that Saanen females are more affected by climatic variations and that the rainy season has a greater negative impact on hematological parameters. The values obtained could serve as a reference for these genotypes in Ceará.

E-mail: clauciahonorato@yahoo.com.br This study aimed to evaluate possible histopathological liver of tilapia fed diets containing fish silage and level protein. Sample of 180 fed with tilapia fingerlings fed with diets containing three protein levels (20, 24 and 28% CP), and Proportions residue fermented silage of tilapia (¼ and ½) of were analyzed during 75 days. The tissue fragments were fixed in Bouin and included in Histosec®. After that, between 2 to 5μm slices were done in a rotation microtome. The methods used for tissue analysis were hematoxilin/eosin and PAS. The histological slices were examined under light microscope (Olympus BX-50). The disarray in the morphology of the liver of fish fed biological silage was influenced by high protein levels, and increased proportions of ½ of animal protein diets. It was observed that the variation of hepatocytes is directly related to the type of diet for fish. In fish fed diets containing 28% CP, the liver showed disruption of the structure cordon of hepatocytes, necrosis and shifting points of the core to the periphery. Elevated levels of biological fish silage cause deleterious changes in the liver. The level of protein required to maintain the health of the associated development fish is 24% crude protein.

E-mail: assiralm@gmail.com Leptospirosis is an endemic worldwide anthropozoonosis, affecting humans and several species of domestic and wild animals. At the beginning of infection is the production of IgM to control the infection and after a few days, IgG is produced and cause lysis of circulating leptospires. The objective of this study was to identify deposits of immunoglobulins and antigens of leptospires in kidney tissue, to assess the role of immunoglobulins in the pathogenesis of leptospirosis nephropathy in pigs. We collected 139 blood samples and kidney of pigs from the cities of Timon/MA and Teresina/PI, to be evaluated by SAM, immunohistochemistry and PCR. Interstitial nephritis, fibrosis, vasculitis; swollen glomeruli hypercellularity and diffuse in a pig were main pathological changes found. Immunohistochemistry leptospira antigen detected in 60 pigs. Deposits of IgG, IgM and IgA were observed in the endothelium of glomerular capillaries, the capillaries intertubulares and the Bowman’s capsule, marked focal, diffuse, global and segmental. The deposition of IgM and IgA was significantly higher in infected pigs, strangely deposits of IgG was significantly higher in non-infected pigs, where there was no presence of antigen leptospires nor tubulointerstitial injury. We conclude that Leptospira antigen in porcine kidney relates to deposits of IgM and IgA but not IgG deposits.

E-mail: jpacosta00@hotmail.com The distribution of some of the cells involved in the immune response at the level of the prepuce and the pelvic urethra was quantified in four one-year-old rams seronegative for B. ovis and A. seminis and without apparent lesions in the testicles, the epididymis, and the prepuce. At the moment of slaughter, samples were taken from the preputial fornix and the pelvic urethra and placed in 10% formalin and under freezing conditions. CD4, CD8, WC1, CD45RO, CD14 and CD1b cells were demonstrated by immunohistochemistry, and immunoglobulin-containing cells (ICC) of the IgA, IgG and IgM classes were demonstrated by immunofluorescence. The labeled cells present in the mucosa of both organs were counted with an image analyzer. The total number of cells was compared between both tissues and differentially between the epithelium and the connective tissue of the mucosa. Significant differences were found in the total number of CD4, CD45RO, and WC1 lymphocytes, in CD14 macrophages, and CD1b dendritic cells, with mean values being greater in the fornix than in the urethra (p<0.05) in all cases. Only dendritic cells were found in the prepuce. No differences were found in the number of CD8 lymphocytes between both organs. The ratio between each cell type in the connective and the intraepithelial tissues and between organs was 10/1 for CD4 in the fornix (p<0.05), against 7/1 in the urethra (p<0.05), while CD8 had a 1/1 distribution in both mucosae. The WC1 ratio was 5/1 in both mucosae (p<0.05). CD45RO labeling was 19/1 in the prepuce (p<0.05) and 1/1 in the urethra. IgA-containing cells did not show differences in the total number of cells in both tissues. In the urethra, no IgG-containing cells were observed and IgM-containing cells were scarce; in contrast, both cell types were present in the prepuce, in amounts greater than in the urethra (p<0.05). IgA-, IgG-, and IgM-containing cells were located in both organs in the mucosal connective tissue. The presence of antigen-presenting cells, macrophages, and dendritic cells, as well as of lymphocytes CD4, CD8 TCR γδ (WC1), IgA-, IgG and IgM positive cells, and CD45RO cells suggests that both mucosae may behave as inductive and effector sites for the mucosal immune response.

RESUMO.- Acosta-Dibarrat J., Buendía-J. A., Soriano-Vargas E., Oca-Jiménez R.M. & Tórtora-Pérez J. 2014. Distribution of immune response cells in the pelvic urethra and the prepuce of rams.

Email: moleta@ufmt.br In the county of Colniza, Mato Grosso, the main limitation for livestock production is the occurrence of “sudden death” in cattle, which affects in some farms up to 50% of the herd. In visits to some of the farms where the problem occurred, in 2004, 2011 and 2012, the presence of Amorimia pubiflora on the pastures was associated with the occurrence of “sudden deaths” in cattle. The deaths occurred throughout the year, however more frequently at beginning of the rainy season, when A. pubiflora sprouts in the grazing areas. The poisoning was experimentally reproduced in sheep and cattle by the administration of young leaves of the plant collected during two seasons, and in sheep by the administration of mature leaves and fruits. In the sheep that died, the first clinical signs were observed between 34min and 17h34min after the administration of the plant, and the clinical course varied from 3min to 15h20min, with a final peracute phase of 3 to 21 minutes. The main clinical signs were tachycardia, engorgement of the jugular veins, muscle trembling, apathy and reluctance to move, which were more evident when the animals were moved. The peracute final phase was characterized by generalized tremors and muscle contractions mainly of limbs, head and neck, respiratory distress and abdominal respiration, sternal and quick lateral recumbence or falling to the ground with peddling movements, opisthotonus, nystagmus, nystagmus and cyanosis of the oral mucosa, followed by death. The young leaves of A. pubiflora, independent of the collection period, were more toxic and caused death of sheep and cattle after ingestion of 2g/kg and 3g/kg respectively. Mature leaves caused death at the dose of 20g/kg, and the fruits at 5g/kg. The young leaves contained 0.015% of sodium monofluoracetate which is responsible for clinical signs of the “sudden death”. These findings show the importance of Amorimia pubiflora for cattle raising in Midwestern Brazil. The plant is toxic also for sheep causing a clinical picture similar to that reported in cattle poisoned by monofluoracetate-containing plants.
