Brucella spp. are intracellular facultative gram-negative bacteria which are pathogenic for many species of mammals, causing brucellosis, a worldwide spread zoonosis. Therefore the search for more efficient alternatives of control, as the development of new potential immunogens is necessary. In this study, we knockouted virB10 from Brucella abortus S2308 strain, generating a mutant strain probably incapable to produce the corresponding native protein. The gene virB10 is part of an operon that codifies for type IV secretion system, which is essential for the intracellular survival and multiplication of the bacteria in host cells. The knockout was carried through by the construction of the suicidal plasmid pBlue: virB10: kan and eletroporation in eletrocompetent cells of B. abortus S2308, leading to the exchange of the wild gene for the interrupted gene, containing the gene of resistance to kanamycin, for double homologous recombination. BALB/c mice were inoculated with S19, RB-51, DvirB10 strains of B. abortus and S2308 wild strain; the results demonstrated that the BALB/c mice inoculated with S19 and BALB/c mice inoculated with S2308 presented faster fall of trend line, when compared with the too much groups, for bacterial recovery (BR) and esplenic weight (EW) respectively. The groups that received DvirB10 S2308 B. abortus and RB-51 demonstrated similar behavior for both the characteristics. In the sixth week postinoculation, the results for BR (log UFC ± standart deviations) and EW (esplenic weight ± standart deviations), respectively, showed: groups inoculated with strains S2308 (4,44±1,97 and 0,44±0,11), S19 (1,83±2,54 and 0,31±0,04), RB-51 (0,00±0,00 and 0,20±0,01) and DvirB10 S2308 (1,43±1,25 and 0,19±0,03). Considered the bacterial clearance, all the groups differed statistical from the group that received S2308 (p<0,0001), the group inoculated with DvirB10 S2308 B. abortus was similar to the S19 group (p=0,4302) and different of group RB-51 (p=0,0063). The evaluation of the persistence of the strains showed that virB10 is essential for the maintenance of the virulence. These results support other studies concerning the immunogenic potential of this mutant strain.

Brucella spp. são bactérias gram-negativas, intracelulares facultativas que são patogênicas para muitas espécies de mamíferos causando a brucelose, uma zoonose difundida mundialmente. Por isso a busca de alternativas de controle mais eficientes se faz necessário como o desenvolvimento de novas cepas que possam ser testadas como potenciais imunógenos. Neste estudo realizou-se a deleção do gene virB10 da cepa S2308 de Brucella abortus gerando uma cepa knockout provavelmente incapaz de produzir a proteína nativa correspondente. O gene virB10 faz parte de um operon que codifica para um sistema de secreção do tipo IV, essencial para a sobrevivência intracelular e multiplicação da bactéria em células hospedeiras. A deleção foi realizada pela construção do plasmídeo suicida pBlue:virB10:kan e eletroporação deste em células eletrocompetentes de B. abortus S2308, ocorrendo a troca do gene selvagem pelo gene interrompido, com o gene de resistência a canamicina, por recombinação homóloga dupla. Camundongos BALB/c foram inoculados com as cepas S19, RB-51, DvirB10 de B. abortus e B. abortus S2308 selvagem; os resultados demonstraram que camundongos BALB/c inoculados com S19 e camundongos BALB/c inoculados com S2308 apresentaram queda mais rápida de linha de tendência, quando comparadas aos demais grupos, para recuperação bacteriana (RB) e peso esplênico (PE) respectivamente. Os grupos que receberam DvirB10 S2308 de B. abortus e RB-51 demonstraram comportamento semelhante para ambas as características. Na sexta semana após a inoculação, os resultados para RB (log de UFC ± desvio padrão) e PE (peso esplênico ± desvio padrão), respectivamente, mostraram: grupos inoculados com as cepas S2308 (4,44±1,97 e 0,44±0,11), S19 (1,83±2,54 e 0,31±0,04), RB-51 (0,00±0,00 e 0,20±0,01) e DvirB10 S2308 (1,43±1,25 e 0,19±0,03). Considerado o clearance bacteriano, todos os grupos diferiram estatisticamente do grupo que recebeu S2308 (p<0,0001), o grupo inoculado com DvirB10 S2308 de B. abortus foi semelhante ao grupo S19 (p=0,4302) e diferente do grupo RB-51 (p=0,0063). A avaliação da persistência revelou que o gene virB10 é essencial para a manutenção da virulência da bactéria. Os resultados obtidos possibilitarão que outras pesquisas sejam realizadas avaliando o potencial imunogênico desta cepa mutante.

As formas de imobilização em bovinos mais utilizadas levam a uma manipulação excessiva e consequentemente estressante para o animal. A dor é uma experiência emocional de sensação desagradável que vem associada a um prejuízo tecidual. Surge no mercado um método de contenção física para bovinos que propõe a utilização de um aparelho que funciona com ondas eletromagnéticas de baixa intensidade promovendo imobilização sem estresse e ou estímulo doloroso para o animal. O objetivo deste projeto foi avaliar os parâmetros cardiorrespiratórios e hemogasométricos bem como os níveis de cortisol de bovinos submetidos à imobilização eletromagnética, a fim de observar se tal método é eficaz na diminuição do estresse e/ou estímulo doloroso. Foram utilizados 6 bovinos, mestiços, peso médio de 300,3 kg (±85,76). Os animais foram alocados aleatoriamente em dois grupos (controle dele mesmo): grupo controle (GC), introdução da sonda via retal (desligada); grupo Imobilizador (GI) introdução da sonda e permanência desta ligada por 5 minutos. Análise estatística utilizou o Teste T pareado entre os grupos (P<0,05) e a avaliação entre tempos dentro de cada grupo análise de variância de uma única via com repetições múltiplas (ANOVA - RM). Momentos avaliados M0: basal; M1: imediatamente após a retirada da sonda; M2 e M3 (30 e 60 minutos após M1 respectivamente). Houve um aumento significativo nas variáveis: frequências cardíaca e respiratória em (M1), na Pressão arterial média (M1, M2, M3) e níveis de cortisol nos momentos M1 e M2 no grupo imobilizador quando comparado ao grupo controle. Com relação à resposta ao estímulo, o GI teve resposta negativa no momento da estimulação, comparado ao GC (resposta máxima). De acordo com os resultados obtidos, podemos concluir que o imobilizador eletromagnético é um método, exclusivamente, de contenção física, não oferecendo nenhum suporte anestésico, para o conforto ou bem estar ao animal.

The ways of immobilization generally used in cattle provide excessive manipulation and stress for the animal. The pain is an emotional experience and unpleasant sensation in combination with tissue damage. The new method of physical restraint for cattle uses low intensity electromagnetic waves, promising immobilization without stress or painful stimulation for the animals. The aim of this study is to evaluate of cardiorespiratory effects and plasmatic cortisol concentration of cattle submitted to electromagnetic immobilization. Six healthy cows with average weight of 300.3 ± 85.76 kg (mean ± SD) were assigned to two treatments in a randomized design with a 15-day interval between experiments GC (control group) and GI (immobilization group). Data are presented as mean + SD. The statistical analyses used the t-Test between groups, and ANOVA random block design between times into same group. All statistical analyses were performed using P<0.05 as a significant level. Evaluation moments: M1 (basal); M1: after withdrawal probe, M2 and M3 (30 and 60 minutes after M1 respectively). There was a significant increase of FC and f in M1, PAM in the moments (M1, M2, M3) and in the levels of plasmatic cortisol concentration in M1 e M2 in the immobilization group when to compared with the control group. It could be concluded that this electromagnetic immobilization is a method of physic restraint only, offers no anesthetic support and favors comfort and animal welfare.

O presente trabalho visou avaliar a vitalidade e os níveis séricos de cortisol de bezerros neonatos, nascidos de partos eutócicos e auxiliados, bem como determinar os índices de partos distócicos em vacas receptoras de embriões nelores. Cerca de 310 bezerros foram avaliados. Observou-se a ocorrência de 38 partos distócicos (38/310) e de oito abortos (8/310). Os bezerros nascidos de partos distócicos mais duradouros, entre quatro e seis horas, apresentaram os maiores valores médios de frequência respiratória e dos níveis séricos de cortisol. A temperatura retal foi mais elevada em animais nascidos de partos laboriosos do que naqueles nascidos de partos normais. As taxas de morbidade e mortalidade em animais neonatos nelores foram de 77,93% e 3,44%, respectivamente. O emprego do escore APGAR, ao nascimento, demonstrou que 90% dos bezerros apresentavam-se deprimidos, independente do tipo de nascimento.

The purpose of present study was to evaluate the vitality and the serum levels of cortisol from newborn calves by normal and laborious deliveries, and to obtain the distocia register in Nelore embryo recipient cows. Three hundred ten calves were evaluated. The register of 38 dystocic calvings (38/310) and 8 abortions (8/310) were observed. In newborn animals the morbity and mortality rates were 77.93% and 3.44%, respectively. Calves born after long dystocic calving, lasting 4-6 hours, showed the highest mean values of respiratory frequency and serum cortisol levels. Animals born after laborious calving presented higher rectal temperature than those born at normal calving. The APGAR score showed that 90% calves had signs of dullness independent of parturition type.

The purpose for using lobectomy or pneumonectomy in dogs and cats, is to cure or palliate of bronchopulmonary diseases whenever conservative clinical treatment proves ineffective. Considering the marked changes caused by pneumonectomy, new experimental studies have to be done to assess the advantages of this surgical intervention allow its performance without any risk. The aim of the current study was to evaluate the electrocardiographic alterations in ten adult mongrel dogs of both sexes with between 10 and 30 Kg that were submitted to right pneumonectomy. The clinical parameters of the dogs were evaluated in a daily basis and the alterations in every electrocardiogram derivation were recorded. All dogs presented a good post-operative outcome. In most cases there were no electrocardiographic alterations; when these alterations were observed they were of no clinical significance and included premature ventricular complexes in one dog, most likely resulting from a cardiorespiratory arrest that was reverted to successfully, and a decreased width in the QRS complex amplitude on the first 0-14 days post surgery which returned to normal after sixty days post surgery.

O propósito das ressecções pulmonares em cães e gatos, quer sejam por lobectomia ou pneumonectomia, é a cura ou paliação de processos broncopulmonares sempre que os meios conservadores de tratamento clínico sejam considerados ineficientes. Tendo em vista as significativas alterações resultantes da pneumonectomia, novos estudos experimentais devem ser feitos para avaliar as vantagens dessa intervenção cirúrgica e determinar a maneira como aplicá-la com segurança. O presente estudo tem como objetivo avaliar as alterações eletrocardiográficas em dez cães adultos de ambos os sexos, sem raça definida, com 10-30 kg, submetidos à pneumonectomia direita. Foram avaliados diariamente os parâmetros clínicos de cada cão e as alterações em todas as derivações do eletrocardiograma. Todos os cães apresentaram um bom desenlace pós-operatório. Apenas um cão apresentou alteração de relevância clínica, um caso de complexos ventriculares prematuros, possivelmente decorrente da parada cardiorrespiratória, que foi revertido com sucesso. Houve diminuição da amplitude dos complexos QRS nos primeiros 14 dias, retornado ao normal após 60 dias de pós-operatório.

Chronic renal disease (CRD) is the most common form of renal disease in cats. Several factors contribute to disease progression. Proteinuria is an important marker of renal disease progression. The protein-creatinine ratio in a single urine sample correlates well with urinary protein loss in 24 hours. The aim of this investigation was to determine the urine protein-creatinine (UPC) ratio in cats with natural acquired chronic renal disease. The UPC ratio was performed in nine clinically normal cats and in 30 cats with chronic renal disease. Mean UPC ratio in normal cats was 0.16±0.10, and mean UPC ratio in the cats with chronic renal disease was 0.53± 0.59. In the group with renal disease there was positive correlation between UPC ratio and serum creatinine levels. The results obtained from this study demonstrate that UPC ratio in cats with CRD is variable and that, in accordance to what has previously been described, approximately one third of the cats with CRD are considered proteinuric according to the criteria established in literature (UPC ratio > 0.4).

Doença renal crônica (DRC) é a forma mais comum de doença renal em gatos. Vários fatores têm sido citados como importantes na progressão da doença, dentre eles a proteinúria. A relação proteína-creatinina (RPC) urinária em uma única amostra de urina apresenta boa correlação com a perda de proteína urinária em 24 horas. O objetivo dessa investigação foi determinar a RPC urinária em gatos com DRC adquirida naturalmente. A determinação da RPC foi realizada em nove gatos saudáveis (Grupo I) e em trinta gatos com DRC (Grupo II). Os gatos do Grupo I apresentaram RPC de 0,16±0,10 e os gatos do Grupo II apresentaram RPC de 0,53± 0,59. No Grupo II encontrou-se correlação positiva e significante da RPC com o nível de creatinina sérica. Os resultados deste estudo demonstram que a RPC urinária em gatos com DRC é bastante variável e que, à semelhança do que já havia sido previamente descrito, aproximadamente um terço dos gatos com DRC são considerados proteinúricos segundo critérios estabelecidos pela literatura (RPC urinária >0,4).

Attempting to avoid difficulties associated to triple pelvic osteotomy (TPO), the sacroiliac wedge method was experimentally developed to increase the acetabular ventroversion. The goal of this study was to apply the techniques of sacroiliac wedge and TPO to canine cadavers and radiographically evaluate the efficacy of acetabular ventroversion. Ten cadavers of adult dogs weighting 15-25 kg were used. To each right hemipelvis the technique of TPO with 20° and 40° plates was applied. In the left hemipelvis, 20° and 40º wedges in the sacroiliac joint were applied. Radiographic evaluations in standard projections were carried out for measurement of the acetabular covering of the head of the femur in the two techniques. There was no significant differences between TPO and the use of sacroiliac wedge using implants of 20° and 40°, however a significant difference (p<0,05) could be observed before and after application of the 20° and 40° implants, as well as between those that received 20° and of 40° implants. The application of sacroiliac wedge produced similar results of those observed in TPO, and it is more feasible.

Na tentativa de evitar algumas das dificuldades associadas à osteotomia pélvica tripla (OPT), foi desenvolvido experimentalmente o método de aplicação de cunha na junção sacroilíaca para aumentar a ventroversão acetabular. O objetivo deste estudo foi aplicar as técnicas de cunha sacroilíaca e OPT em cadáveres e avaliar radiograficamente a eficácia da ventroversão acetabular. Para tal, foram utilizados 10 cadáveres de cães, adultos, com 15-25 kg. Em cada hemipelve direita foi realizada OPT com placas de 20° e 40°. Na hemipelve esquerda foram aplicadas cunhas nas articulações sacroilíacas de 20° e 40°. Avaliações radiográficas em projeções ventrodorsais foram realizadas para mensuração da cobertura acetabular à cabeça femoral nas duas técnicas. De acordo com os dados obtidos pode-se observar que não houve diferença entre a técnica de OPT e o uso de cunha sacroilíaca utilizando implantes de 20° e 40°, mas ocorreu diferença significativa (p<0,05) entre os cães antes e após a aplicação dos implantes de 20° e 40°, e também entre os que receberam implantes de 20° e os de 40°. A aplicação de cunha sacroilíaca produziu resultados semelhantes à OPT, e também se mostrou como de mais fácil aplicação.

The aim of this study was to establish an immunological profile by marking and quantification of T and B lymphocytes in Curraleiro breed by immunocytochemistry. A total of 116 cattle was used, males and females of different ages, from two farms situated in the Goiás State, Brazil. The animals were allotted into groups according to age, sex and origin. Blood samples were collected and processed in accordance with immunocytochemistry standard technique using lymphoid markers species-specific, as anti-CD3 (MM1A - BoCD3) and anti-LB (LCTB16A-clone B-B14), for T and B lymphocyte counting, respectively. The data were submitted to descriptive statistics, and to Kruskall Wallis and Mann-Whitney tests. The results showed decreased levels of leukocytes, lymphocytes, T lymphocytes and B lymphocytes along the age advance. Absolute values of leukocytes, lymphocytes and T lymphocytes were higher in males than females. None of the evaluated parameters were affected by differences of the management carried out at the two farms.

The objective of the study was to evaluate the effect of platelet-rich plasma (PRP) in the treatment of induced tendinitis in the superficial digital flexor tendon (SDFT) of horses through ultrasonographic evaluation. Thus, six 8 to15-year-old healthy gelding horses (=12 years) were used. Tendinitis of the SDFT was provoked in both forelimbs via intratendineous administration of 2.5mg of collagenase (2.5mg.mL-1), what was considered as beginning of the experimental phase. Twelve days after induced tendinitis, the horses were submitted to two treatments: (1) in the lesion caused in the right SDFT (treated group, TG), 2.5mL of PRP activated with calcium chloride at 0.0125mol.L-1, at concentrations from 320,000 to 500,000 platelets.µL-1, were administered; (2) in the tendinitis of the left SDFT (control group, CG), 2.5mL of 0.9% saline solution was administered. After 5 days, the animals were submitted to controlled and progressive physical activity during 30 days. Ultrasonographic examinations were carried out before and after tendinitis induction (48 hours after and on experimental days 7, 12, 14, 21 28, 35 and 42), with tendon cross-sectional area (CSA), lesion cross-sectional area (L-CSA), L-CSA percentage, severity and echogenicity of the lesion, as well as collagen fiber alignment being evaluated. The result revealed reduction (P<0.05) in the L-CSA and in the degree of lesion echogenicity in function of time, but with difference (P<0.05) between groups only for L-CSA, with smaller values in TG. The treatment with PRP promotes greater reduction in the area of the lesion measured by ultrasound.

“Inflow Occlusion” technique can be used in heart surgeries when heart is required to be opened just for few minutes, to allow quick repairs. However, circulatory arrest, event occasioned by this technique, may produce serious metabolic and neurological consequences to the patient. In this study, 12 mongrel dogs were used, divided into two groups, A and B, which were submitted to 7 and 8 minutes of total circulatory arrest, respectively, using “Inflow Occlusion” technique. Normothermia was tried during surgical procedures. Clinical and behavior evaluation were performed after surgery to both groups, and biochemical data were collected to compare pre and post-operatory moments. There were two transoperatory deaths in Group B. Transitory clinical problems were observed in group A until moment M7 (48 hours after surgery), and in Group B these problems were more intense and seen even after M7; and permanent blindness in one animal of Group B was present during all follow up period. Despite all alterations found during the study, it might be safe to use “Inflow Occlusion” technique for periods up to 7 minutes, however, it is contra-indicated for longer periods.

The objective of this investigation was to evaluate the in vitro sensibility of Staphylococcus spp. to several commercially available disinfectants used for pre and post dipping. A total of 60 isolates of Staphylococcus spp., identified as Staphylococcus aureus (50) and Positive coagulase Staphylococcus (10) were obtained from the mammary glands of dairy cows with subclinical mastitis in the regions of Metropolitan Recife, the Agreste and the Zona da Mata of the State of Pernambuco, Brazil. As active ingredients were used a chlorine base (25%), iodine (0.6%), chlorhexidine (2.0%), quaternary ammonium (4.0%), and lactic acid (2.0%) at four specific intervals (15", 30", 60", and 300"). One hundred percent of S. aureus was found to be sensitive to iodine, 93.3% to chlorhexidine, 80% to ammonia, 35.6% to lactic acid, and 97.8% were resistant to chlorine at a 60-minute interval. With respect to the Positive coagulase Staphylococcus (SCP), 100.0% of the isolates were sensitive to iodine, 81.8% to quaternary ammonium, 99.9% to lactic acid, 72.7% to chlorhexidine, and 100% was resistant to chlorine at an interval of 60 minutes. It can be concluded that the highest disinfectant activity in vitro was found to be with iodine and chlorhexidine for S. aureus, and with iodine and lactic acid for SCP. A further conclusion was that it is important to undertake a periodic evaluation of the disinfectants used on the dairy properties in the regions studied, given the variety of sensibilities and resistance to disinfectants used, which may prejudice the control of bovine mastitis caused by Staphylococcus spp.
