The aim of this study was to determine the clinical-pathological alterations and laboratory findings in cattle inoculated with Bothrops alternatus venom, with the intention of providing information for the establishment of diagnosis and differential diagnosis procedures, as well as to elucidate some obscurities observed in the pertinent literature. The lyophilized venom was diluted in 1 ml of physiologic solution. It was administered to 5 bovines by the subcutaneous route at doses of 0.0625, 0.125 and 0.25mg/kg body weight, and to 2 bovines by the intramuscular route at doses of 0.25 e 0.45mg/kg. Six bovines died and the only animal that survived, who had subcutaneously received the venom at a dose of 0.0625mg/kg, recovered. The first clinical signs were observed from 25min to 5h30min after the inoculation. The clinical evolution time varied from 7 hours 18 minutes to 92 hours. Regardless of the dose, the clinical picture was characterized by swelling (hemorrhage/hematoma) at the site of inoculation, increase in bleeding time and capillary refill time, paleness of mucous membranes and apathy. The laboratory exams revealed progressive normocytic normochromic anemia, thrombocytopenia, reduction in fibrinogen and total plasma proteins, decreased hematocrit and hemoglobin, and slight increase in creatine kinase and lactate dehydrogenase. When subjected to necropsy, the animals that received the venom through the subcutaneous route showed large hematomas and hemorrhagic areas in the subcutaneous tissue, extending from the site of inoculation. The animals in which inoculation was performed intramuscularly exhibited, in addition, intramuscular hemorrhage. The left endocardium showed extensive hemorrhagic lesions, and petechiae were found on the serosae of rumen, omasum, abomasum and gall bladder. The colon, rectum and perirenal areas were surrounded by clotted blood in three animals. Besides hemorrhage, the histological examination revealed hemorrhage and coagulative muscle necrosis in the vicinities of the inoculation site in the animals that received the venom by the intramuscular route. These lesions were mild in the muscles close to the site of inoculation in the animals inoculated by the subcutaneous route. The swelling on the site of inoculation and its surroundings was a consequence of blood accumulation and not merely edema. Myoglobinuria and gross or microscopic lesions were not observed in the kidneys. This study indicates that B. alternatus can lead adult bovines to death if they release all their venom during a bite. Conversely, since venomous ophidians can control the amount of poison they inoculate and they probably do not identify bovines as potential preys, it is likely that the number of accidents involving this species is small, which is in agreement with the findings of the majority of the centers for anatomical-pathological diagnosis in Brazil.

However, some aspects of their macro anatomy need a more detailed description, especially the abdominal and pelvic arterial vascular system, which has a huge variability in distribution and trajectory. Thirty cadaveric adult New Zealand rabbits, 13 male and 17 female, with an average weight and rostrum-sacral length of 2.5 kg and 40cm, respectively, were used. The thoracic aorta was cannulated and the vascular system was filled with stained latex S-65. The celiac artery and its proximal branches were dissected and lengthened in order to evidence origin and proximal ramifications. The celiac artery emerged between the 12th and 13th thoracic vertebra in 11 (36.7%) rabbits; at the level of the 13th thoracic vertebra in 6 (20%) rabbits; between the 13th thoracic vertebra and the 1st lumbar vertebra in 12 (40%) rabbits; and at the level of the 1st lumbar vertebra in only one (3.3%) rabbit. The mean length of the celiac artery was 0.5cm. The celiac artery first branch was the lienal artery, the second branch was the left gastric artery and the hepatic artery arose from the left gastric artery in all the dissected rabbits. No relation was observed between the celiac artery length and the rostrum-sacral length in rabbits. The number of left gastric and lienal artery branches and the distribution of celiac artery origin are not gender dependent.

Twenty pairs of agouti (Dasyprocta prymnolopha Wagler, 1831) kidneys were studied to describe the arterial anatomical-surgical segments. The renal arteries were injected with stained acetate vinyl, followed by procedures of acid corrosion in order to obtain vascular casts. It was found that the renal artery is always single and bifurcated into ventral and dorsal sectorial arteries. The sectorial arteries reached the kidneys (100% of the cases) through the hilus. These vessels gave origin to segmental branches responsible for kidney irrigation. At the right kidney, the ventral sectorial arteries gave origin to 3 (60% of the cases), 4 (35%) and 5 (5%) segmental branches; the dorsal sectorial arteries gave origin to 3 (30%), 4 (45%), 5 (20%) and 6 (5%) segmental arteries separated by a vascular sector. At the left kidney, the ventral sectorial arteries originated 2 (10%), 3 (55%) or 4 (35%) segmental branches; the dorsal sectorial arteries gave origin to 3 (25%), 4 (50%) and 5 (25%) segmental branches. Based on the arterial distribution of agouti kidneys, independent sections and arterial segments were found, so that it is possible to accomplish partial kidney resection surgery.

The aim of this study was to evaluate the use of a locally made biosynthetic cellulose membrane after experimental trochleoplasty, in order to verify whether its use could support migration of chondrogenic cells. Twelve male and female adult healthy dogs and without claudication were used. All dogs were submitted to trochleoplasty in both pelvic limbs after sedation and epidural anesthesia. In the left hind limb, the biosynthetic cellulose membrane was fixed with simple suture using Polyglactin 910 6-0 after performing trochleoplasty (treated group); whereas in the right limb (control group) only trochleoplasty was performed. The dogs were subdivided into 4 subgroups for postoperative evaluation at 15, 30, 60 and 90 days post-surgery. Biopsy was performed after exploratory arthrotomy for histopathologic and morfometric evaluation. At 30 and 60 days post-surgery, more condrocyte-like cells of immature aspect were observed in lesions treated with the cellulose membrane. At 90 days post-surgery the reparative tissue was characterized as mature fibrocartilage-like tissue without difference between the groups. In the control group there was a progressive increase of the number of cells until the end of the evaluation period. Otherwise, when compared to the initial period (15 days), there was an increase in the number of cells until 60 days, followed by a return the initial values at 90 days in the treated group. In comparison to controls, the number of cells was greater in the treated group from 15 to 60 days. Initially, the neoformed repair tissue was thicker in the treated group. From the results of this study, it was concluded that the cellulose membrane shortened the initial tissue repair process in the trochleoplasty area, showing good integration of the neoformed tissue with the adjacent cartilage.

To determine the reversibility of neurological and cardiac lesions in Ateleia glazioviana and Tetrapterys multiglandulosa poisoning, 3 groups of four sheep each were fed orally with the plants. In Group 1, 2 and 3, when sheep with nervous signs showed considerable increase of symptoms and risk of death, the plant administration was suspended. Group 4 with two sheep was used as control. Sheep from Group 1, fed 10g/kg during 6 days of fresh A. glazioviana collected in autumn were euthanized 8, 11, 16, and 21 days after the start of the experiment; they had shown regression of nervous signs, but had progressive cardiac lesions. Sheep from Group 2, fed during 8 days the same dose of fresh A. glazioviana collected in spring, were euthanized on days 9, 23, 38 and 68; they had shown regression of nervous signs and had no cardiac lesions. This suggests that the plant is less toxic in spring. Sheep from Group 3, fed 10g/kg during 11 days dry and ground T. multiglandulosa mixed with concentrated food, were euthanized on days 33, 33, 92 and 92; they had shown regression of nervous signs, and cardiac lesions were less severe on day 33 than on day 92. These results indicate that nervous lesions are reversible after the end of feeding, but cardiac lesions are progressive after discontinuation of the plant administration; these can be afterwards reversible if the animals do not anymore show clinical signs or die as a consequence of the poisoning. Results of those and previous experiments show that lesions of the nervous system are induced by lower doses than cardiac lesions and occur within a shorter period, what suggests that in spontaneous cases cardiac signs are always preceded by nervous signs, and that nervous signs can occur in the absence of cardiac lesions.

Venereal infection of heifers and cows with bovine herpesvirus type 1.2 (BoHV-1.2) may result in vulvovaginitis and transient infertility. The acute infection is followed by the establishment of latent infection which can be periodically reactivated. We herein describe the virology and clinico-pathological aspects of acute and recrudescent vulvovaginitis in heifers inoculated with a Brazilian BoHV-1.2 isolate recovered from an outbreak of balanoposthitis. Genital inoculation of isolate SV-56/90 (108.1TCID50/animal) in four eight-months-old heifers resulted in efficient virus replication in the genital mucosa and the development of moderate to severe vulvovaginitis. The inoculated heifers shed virus in genital secretions in titers up to 107.3TCID50/mL until day 10 pi and developed genital congestion, swelling, vesicles and pustules. The vesicles and pustules increased in size eventually coalesced and became covered with a yellowish exsudate. These signs appeared at day 2 pi, increased in severity up to days 5 - 8 pi and progressively subsided thereafter. Dexamethasone administration at day 55 pi resulted in virus shedding in vaginal secretions for up to 10 days. Virus reactivation in all animals was accompanied by clinical recrudescence of the disease, yet less severe than during acute infection. Examination of sacral ganglia and lymph nodes by PCR at day 36 post-reactivation revealed the presence of latent viral DNA in the pudendal (4/4), genito-femoral, sciatic and rectal caudal (3/4) and obturator nerve ganglia (1/4); in addition to several regional lymph nodes. These results demonstrate the virulence of isolate SV-56/90 for heifers and pave the way for its use in further pathogenesis studies and vaccine-challenge trials.

Venereal infection of heifers and cows with bovine herpesvirus type 1.2 (BoHV-1.2) may result in vulvovaginitis and transient infertility. The acute infection is followed by the establishment of latent infection which can be periodically reactivated. We herein describe the virology and clinico-pathological aspects of acute and recrudescent vulvovaginitis in heifers inoculated with a Brazilian BoHV-1.2 isolate recovered from an outbreak of balanoposthitis. Genital inoculation of isolate SV-56/90 (108.1TCID50/animal) in four eight-months-old heifers resulted in efficient virus replication in the genital mucosa and the development of moderate to severe vulvovaginitis. The inoculated heifers shed virus in genital secretions in titers up to 107.3TCID50/mL until day 10 pi and developed genital congestion, swelling, vesicles and pustules. The vesicles and pustules increased in size eventually coalesced and became covered with a yellowish exsudate. These signs appeared at day 2 pi, increased in severity up to days 5 - 8 pi and progressively subsided thereafter. Dexamethasone administration at day 55 pi resulted in virus shedding in vaginal secretions for up to 10 days. Virus reactivation in all animals was accompanied by clinical recrudescence of the disease, yet less severe than during acute infection. Examination of sacral ganglia and lymph nodes by PCR at day 36 post-reactivation revealed the presence of latent viral DNA in the pudendal (4/4), genito-femoral, sciatic and rectal caudal (3/4) and obturator nerve ganglia (1/4); in addition to several regional lymph nodes. These results demonstrate the virulence of isolate SV-56/90 for heifers and pave the way for its use in further pathogenesis studies and vaccine-challenge trials.

Extracellular matrix plays an important role in chronic hepatic lesions and has been studied in experimental intoxication models. However in cattle, studies on chronic disease have focused on the hepatocellular damage and extracellular matrix (ECM) changes are usually overlooked. There are no specific studies on the hepatic ECM in either normal or chronically damaged bovine liver. Thus an experimental model of hepatic toxicity model using Senecio brasiliensis poisoned calves was designed. Senecio brasiliensis contains pyrrolizidine alkaloids which cause either acute or chronic progressive dose dependent liver damage. Five calves were orally fed with 0.38g of dry leaves of S. brasiliensis/kg/day for 24 days. Liver needle biopsy specimens were obtained every 15 days for 60 days. Clinical signs of digestive complications appeared at 3rd week. One calf died on 45th day and four were evaluated up to 60th day. Biopsy samples were processed for routine light microscopy, immuno-histochemistry and transmission electron microscopy. From 30th day on progressive liver damage characterized by hepatocellular ballooning, necrosis, apoptosis and megalocytosis, centrilobular, pericellular and portal fibrosis were seen by light microscopy. Quantitative and semi-quantitative measurements of hepatic ECM components were performed before and after the onset of lesions. Morphometric analysis of total collagen and elastic fiber system was conducted. Total collagen and I and III collagen types progressively increased in throughout the liver of affected calves. Changes in location, amount and disposition of the elastic fiber system were also observed. Then numbers of Kupffer cells were significantly increased at 30th day and total numbers of sinusoidal cells were significantly increased at 45th and 60th days. Liver damage was progressive and irreversible even after the exposure to the plant was discontinued. Severe fibrotic lesions occurred mainly in portal tracts, followed by veno-occlusive and pericellular fibrosis. Collagen types I and III s were present in every normal and damaged liver, with predominance of type I. In affected calves the increase of total collagen and elastic fibers system paralleled the number of total sinusoidal cells.

For the experimental reproduction of the disease, fresh green leaves, weekly harvested in the same region where spontaneous case occurred, were force-fed to five cattle at doses of 10 and 20g/kg for 120 days, 40g/kg for 30 days, and 30 and 40g/kg body weight for 150 days. One animal died and the others were euthanatized at the end of the experiment. Clinical signs and lesions varied from mild to severe in the experimentally poisoned cattle and depended on dose and length of the period of consumption. Main histological and ultrastructural lesions consisted of vacuolation and distension of neuronal perikarya (mainly of Purkinje cells), and of the cytoplasm of acinar pancreatic cells and thyroid follicular cells. It is concluded that ingestion of even small amounts S. carpinifolia for prolonged periods of time cause lisosomal storage disease in cattle.

Mammillitis caused by bovine herpesvirus type 2 (BoHV-2) is an important disease in dairy herds yet its pathogenesis remains largely unknown. This report describes the reproduction and characterization of acute mammillitis in lactating ewes inoculated with BoHV-2 in the skin of the udder and teats. Five out of eight inoculated ewes developed large plaques, with focal necrosis, small vesicles and crust formation in the inoculated areas. The lesions were first observed on day 4 post-inoculation (pi), progressed in size and severity up to days 7-8pi and subsided progressively thereafter. Infectious virus was isolated from the lesions at days 7 and 8pi. Viral antigens and herpesvirus-like particles were demonstrated by electron microscopy in lesions examined at days 5, 6 and 10pi. Histological findings included epithelial necrosis, erosions and ulcers, and formation of syncytial cells. Intranuclear inclusions bodies in epithelial, syncytial and inflammatory cells and lymphoplasmacytic inflammatory infiltrate in the dermis were also observed. In a second experiment, seven out of ten lambs inoculated into the nostrils and muzzle developed nasal hyperemia and discharge, vesicles, and erosions in the nose. Infectious virus was isolated from lesions during up to three days and all lambs seroconverted to BoHV-2. Attempts to reactivate the latent infection by dexamethasone administration on day 40pi failed, since virus shedding, clinical recrudescence or seroconversion were not observed. The reproduction of acute infection and mammillitis resembling that occurring in cattle paves the way for the use of sheep to study several aspects of the biology of BoHV-2 infection.

The immunoglobulin variation profile in lavages from the broncoalveolar and tracheo-bronchial regions of 20 healthy newborn Holstein male calves was studied. They were fed with colostrum and distributed into 2 groups, 10 animals each. Group 1 underwent the nasotracheal catheterization technique to get the bronchoalveolar lavage (BAL), and Group 2 underwent the tracheocenthesis to collect the tracheobronchial lavage (TBL), both procedures being carried out at a 7-day-interval, starting on the first days up to about one month of life. Higher IgG contents, as compared to IgA, were noted across the respiratory tract. These immunoglobulins were impacted by the site of the respiratory tract washed, as well as by the calves’ life time in weeks. Higher immunoglobulin contents were detected in TBL, as well as higher IgM and IgA rates, as compared to BAL. The BAL immunoglobulins showed a tendency to be reduced in TBL.
