In the present study we investigated the presence of infections by vaccinia-like viruses in dairy cattle from 12 counties in the state of Rio de Janeiro in the last 9 years. Clinical specimens were collected from adult animals with vesicular/pustular lesions mainly in the udder and teats, and from calves with lesions around the nose and mouth. A plaque reduction neutralization test (PRNT) was applied to search for antibodies to Orthopoxvirus; the vesicular/pustular fluids and scabs were examined by PCR, electron microscopy (EM) and by inoculation in VERO cells for virus isolation. Antibodies to Orthopoxvirus were detected in most cases. The PCR test indicated a high nucleotide homology among the isolates and the vaccinia viruses (VACV) used as controls. By EM, typical orthopoxvirus particles were observed in some specimens. The agents isolated in tissue culture were confirmed as vaccinia-like viruses by EM and PCR. The HA gene of the vaccinia-like Cantagalo/IOC virus isolated in our laboratory was sequenced and compared with other vaccinia-like isolates, showing high homology with the original Cantagalo strain, both strains isolated in 1999 from dairy cattle. Antibodies to Orthopoxvirus were detected in one wild rodent (genus Akodon sp.) collected in the northwestern region of the state, indicating the circulation of poxvirus in this area. Nonetheless, PCR applied to tissue samples collected from the wild rodents were negative. Vesicular/pustular lesions in people in close contact with animals have been also recorded. Thus, the vaccinia-like virus infections in cattle and humans in the state seem to be an expanding condition, resulting in economic losses to dairy herds and leading to transient incapacitating human disease. Therefore, a possible immunization of the dairy cattle in the state should be carefully evaluated.

Bovine herpesvirus type 1 (BoHV-1) is recognized as a major cause of economic losses in cattle. Vaccination has been widely applied to minimize losses induced by BoHV-1 infections. We have previously reported the development of a differential BoHV-1 vaccine, based on a recombinant glycoprotein E (gE)-deleted virus (265gE-). In present paper the efficacy of such recombinant was evaluated as an inactivated vaccine. Five BoHV-1 seronegative calves were vaccinated intramuscularly on day 0 and boostered 30 days later with an inactivated, oil adjuvanted vaccine containing an antigenic mass equivalent to 107.0 fifty per cent cell culture infectious doses (CCID50) of 265gE-. Three calves were kept as non vaccinated controls. On day 60 post vaccination both vaccinated and controls were challenged with the virulent parental strain. No clinical signs or adverse effects were seen after or during vaccination. After challenge, 2/5 vaccinated calves showed mild clinical signs of infection, whereas all non vaccinated controls displayed intense rhinotracheitis and shed virus for longer and to higher titres than vaccinated calves. Serological responses were detected in all vaccinated animals after the second dose of vaccine, but not on control calves. Following corticosteroid administration in attempting to induce reactivation of the latent infection, no clinical signs were observed in vaccinated calves, whereas non vaccinated controls showed clinical signs of respiratory disease. In view of its immunogenicity and protective effect upon challenge with a virulent BoHV-1, the oil adjuvanted preparation with the inactivated 265gE- recombinant was shown to be suitable for use as a vaccine.

A condition of cattle and sheep characterized by ulcerous and granulomatous skin lesions of the legs was studied. Epidemiological data and the nature of the lesions indicate that they are caused by the thorns of Mimosa pudica (Leg. Mimosoideae). The disease was observed only in pastures largely infested by the plant, and animals recover quickly when moved from them.

The objective of this paper was to study the molecular performance and phenotypic characterization of Burkholderia mallei isolated from horses with clinical and serological diagnosis of glanders, originating from the Metropolitan District of Recife and Zona da Mata of Pernambuco and Alagoas. The isolation and biochemical identification of B. mallei was carried out by microbiological and molecular techniques of PCR-fingerprinting and RAPD-PCR. From the eight samples studied, four showed little phenotype variations. In the molecular tests, the samples formed 4 groups of different ribotype profiles and 4 genotype profiles. There was some association of PCR-fingerprinting with RAPD-PCR results. It was concluded that the slight biochemical variations were not associated with different molecular profiles. They also indicated that these differences show heterogeneity associated with the origin of the sample, indicating that the infection was caused by clones of different strains and that the polymorphism of DNA observed could make it

Duchenne muscular dystrophy (DMD) is a neuromuscular disorder characterized by a continuous muscle necrosis and degeneration with eventual fibrosis and fatty tissue infiltration. Progressive increase in muscle interstitial fibrosis prevents the movement of myogenic cells, which is necessary for myotube formation. Canine model is the best phenocopies of the disease in humans when comparing with others animal models with dystrophy. Anti-fibrotic treatment of DMD patients, targeting the cytokine mediators, TGF-beta, and the treatment with antiinflammatories, may limit muscle degeneration and contribute for the improvement of the course of the illness. This work aimed to verify the possible adverse effects in renal physiology by means of evaluation sanguineous biochemist and arterial pressure, in order to verifying the viability of Losartan (a TGF-beta inhibiter) in affected dogs by muscle dystrophy. It was used four adults dogs, two of each gender. A dose of 50mg of Losartan was orally given once a day. The clinical exams, the kidney function, arterial blood pressure and potassium level did not show any adverse effect through the experimental period. Losartan utilization showed to be a safe therapy for the antifibrotic treatment in adults dogs, not affecting neither the kidney function nor the arterial blood pressure.

This paper deals with a comparative histologic and morphometric study of the testes of domestic cats distributed into two groups: Group 1, cats until 1 year of age, and Group 2, cats over 1 year. It was found that: (1) at 4 months of age the seminiferous tubules were poorly developed, appeared as seminiferous cords without lumen, lined by a low epithelium, and showed undifferentiated Sertoli cells and scarce interstitial tissue; (2) at 5 months the seminiferous tubules began to differentiate with increase in tubular diameter and lumen, the other tubular structures remaining similar to those previous referred; (3) at 6 and 7 months of age spermatocytogenesis began to appear, Leydig cells were large, polyhedral in shape, with vacuolated cytoplasm and clear nuclei, resting on a sparse interstitial tissue with few blood vessels; (4) 1-year-old cats showed testicular histological features of an adult animal, had seminiferous tubules of large diameter and high seminiferous epithelium with small lumen, and Leydig cells of different sizes, with polyhedral shape, vacuolated cytoplasm, clear nuclei and evident nucleoli resting in a sparse interstitial tissue with some blood vessels; (5) in Group 1 the average diameter of the seminiferous tubules was 160.58µm, and 185.94µm in Group 2; (6) the height of the seminiferous epithelium was 49.51µm for Group 1 and 63.29µm for Group 2; (7) the largest measures of the analyzed parameters were found in animals of Group 2, with functional reproductive organs.

The coronary arteries of the goats have been used to researches about to pharmacological actions, heart failure after connection of coronary arteries and others. Twenty-seven goat hearts, both sexes, fixed in formalin at 10% has been used in this study that analyzed the coronary arteries pattern in goats and if the same is similar to the human coronary arteries pattern. The left coronary artery is dominant and in relation to the length demonstrated average 1.15cm emitting two branches: interventricular paraconalis and circumflex. The interventricular paraconalis branch with average length 10.5cm issued branches to the both ventricles fairly, ending more frequently in the interventricular subsinuosis sulcus. The circumflex branch with average length 8.5cm issued branches to left ventricle and right atrium fairly. The left marginal branch is not constant. The right coronary artery, with average length 6.1cm issued branches to right ventricle and right atrium fairly, but the interventricular subsinuosis branch with average length 3.1cm is variable and can present one long branch, one short branch or one double branch, with the huge part for the right ventricle.

Chlamydophila abortus is a recognized cause of bovine epizootic abortion. Abortion, premature birth and weak lamb/calf, stillbirth and repeat breeding in irregular intervals are the most frequent disease manifestations. The complement fixation test is the recommended by the World Organization of Animal Health (OIE) for Chlamydophila spp. serologic diagnosis. The aim of this study was estimate the prevalence of antibodies against Chlamydophila spp. in cattle herds with abortion, selected inside the sampling design of National Program of Control and Erradication of Brucellosis in Paraná state. Serum samples of 3,102 cows (age ³ 24 months) from 373 herds were analyzed by complement fixation test. Totally, 44 (1.42%) animal were positive with titers ³ 32. The seroprevalence of Chlamydophila spp. in the herds was 8.82% (6.15%-12.17%). Four variables were associated with seroprevalence for Chlamydophila spp. in the final model of logistic regression: confined or semi-confined breeding (OR=3.339, P=0.004), farms with less than 35 cows (OR=3.339, P=0.017), abortion in the pasture (OR=2.372, P=0.037) and pasture rent (OR=3.398, P=0.006) were risk factors for Chlamydophila spp. This bacterium infected a small number of cattle from herds with abortion in Paraná state. Chlamydophila spp impact as abortion cause is reduced in this state.

The objective of the study was to evaluate the effect of platelet-rich plasma (PRP) in the treatment of induced tendinitis in the superficial digital flexor tendon (SDFT) of horses through ultrasonographic evaluation. Thus, six 8 to15-year-old healthy gelding horses (=12 years) were used. Tendinitis of the SDFT was provoked in both forelimbs via intratendineous administration of 2.5mg of collagenase (2.5mg.mL-1), what was considered as beginning of the experimental phase. Twelve days after induced tendinitis, the horses were submitted to two treatments: (1) in the lesion caused in the right SDFT (treated group, TG), 2.5mL of PRP activated with calcium chloride at 0.0125mol.L-1, at concentrations from 320,000 to 500,000 platelets.µL-1, were administered; (2) in the tendinitis of the left SDFT (control group, CG), 2.5mL of 0.9% saline solution was administered. After 5 days, the animals were submitted to controlled and progressive physical activity during 30 days. Ultrasonographic examinations were carried out before and after tendinitis induction (48 hours after and on experimental days 7, 12, 14, 21 28, 35 and 42), with tendon cross-sectional area (CSA), lesion cross-sectional area (L-CSA), L-CSA percentage, severity and echogenicity of the lesion, as well as collagen fiber alignment being evaluated. The result revealed reduction (P<0.05) in the L-CSA and in the degree of lesion echogenicity in function of time, but with difference (P<0.05) between groups only for L-CSA, with smaller values in TG. The treatment with PRP promotes greater reduction in the area of the lesion measured by ultrasound.

[Implantation of the autologous auricular cartilagem in the repair of penile deviation in cattle.] Implante de cartilagem auricular autóloga no reparo de desvio de pênis de bovinos. Eighteen 18 to 24-month-old mixed breed bullocks were submitted to removal of the apical ligament of the penis to induce penile deviation. After 60 days from the surgery the steers were put together with cows in heat. A ventral and right lateral penis deviation leading to incapacity to copulate was observed. The bullocks were then submitted to an autolgous transplant of a cartilage segment from the ear pinna placed on top of the tunica albuginea, replacing the penile ligament apical tha had been removed. The implanted material was well tolerated; histologically with only infiltration few mononuclear cells, polymorpho nucleated cells and giant cells, and fibrosis, which caused strong adherence and provided ability to sustain the free penile extremity. The bullocks that were cows in heat did not show any degree of penile deviation nor any change in sexual behavior. The surgical method described here was shown to be satisfactory as to be indicated for correction of ventral and right lateral penile deviation in bullocks.
