#681 - Histochemical study of the oviducal gland and analysis of the sperm storage tubules of Mustelus schmitti Springer, 1939 (Chondrichthyes, Triakidae), 35(8):741-748

Findings indicate that these glands possessed the same zonation as in most chondrichthyans from anterior to posterior: club, papillary, baffle and terminal zones. The whole gland is composed by simple tubular glands that connect with transverse grooves all along the organ. The club zone presents a typical indian club shape with a simple columnar and ciliated epithelium including secretory cells PAS (+) and AB (+). The papillary zone is characterized by lamella forming small and long cones in numbers of three. The epithelium of this zone contains ciliated cells with apical nuclei and secretory cells with basal nuclei that stain AB (+)The baffle zone consists of apically flattened lamellae alternating with spinnerets which are small projections disposed by both sides of the plateau. This whole structure is present in number of 8 or 9 units. A simple columnar ciliated epithelium covers the plateau and spinnerets and no AB or PAS staining is observed. The epithelium of the terminal zone is PAS (-) and AB (+), and elongated tubules, that run adjacent to the baffle zone are the site where groups of spermatozoa are clearly observed in the lumen. The epithelium of the sperm storage tubules do not stain with any of the dyes tested. Sperm was also observed in the baffle zone, presumably in its way to the fecundation in the oviduct because it displays no aggregation pattern and was between the folds of the epithelium. By scanning electron microscopy sperm was observed in the club and baffle zones in a gland which belonged to a pregnant female.

E-mail: sandrafigueiredo@ufmt.br Yacare Caiman meat consumption has become a marketing trend and a commodity on the rise in Mato Grosso state in Brazil. In the last four years, cuts from the trunk represented 28.40% of total meat sales. Although evolutionary studies, morphophysiological ontogenetic and technology research have been carried out, characterization of muscle and bone bases of cuts from the torso has not been previously reported. The aim of this research is to describe the muscles and corresponding bones related to sirloin, filet mignon and meat trims cuts. To describe the bones, we used six boned carcasses from juvenile Yacare Caiman, as well as an adult specimen, obtained by donation after death from the Federal University of Mato Grosso Zoo. The bones were macerated, bleached and their anatomical details recorded. In order to study the muscle, 24 juvenile specimens were obtained after slaughter and skinning and dissected on both sides. The sirloin cut consists of the semispinal, longissimus and iliocostalis muscles, which are inserted on thoracic vertebrae and ribs, as well as lumbar and sacral ribs. The meat trims cut is formed by latissimus dorsi, serratus, pectoral and abdominal (external oblique, internal oblique, transversus and rectus) muscles, based in various bones: bone ribs are the thoracic, lumbar, and sacral ribs, the gastralia, the sternum and epipúbis. The filet mignon cut is formed by the internal puboischiofemoralis cranial (sublumbar) muscle and by the troncocaudal (ventral surface of the pelvis) muscle.

Objetivou-se descrever os músculos e correspondentes bases ósseas dos cortes filé de lombo, filé mignon e aparas. Na descrição óssea, utilizaram-se seis carcaças desossadas de exemplares juvenis de jacaré-do-Pantanal, além de um exemplar adulto, obtido por doação após óbito, do Zoológico da UFMT. Os ossos foram macerados em água corrente, clareados e descritos. Para a descrição muscular, 24 exemplares juvenis foram abatidos e esfolados, conservados em freezer e descongelados quando utilizados, sem qualquer fixação. Após a evisceração, foram dissecados em ambos os antímeros. Os músculos semiespinhal, longuíssimo e iliocostal, fixados nas vértebras e costelas torácicas, lombares e sacrais, formam o filé de lombo. O corte aparas é constituído pelos músculos grande dorsal, serrátil, peitoral e abdominais (oblíquo externo, oblíquo interno, transverso e reto), cuja base óssea corresponde as costelas torácicas, lombares e sacrais, a gastrália, o esterno e o epipúbis. Por sua vez, o m. puboisquiofemoral interno cranial, localizado na região sublombar e o m. troncocaudal, da superfície ventral da pelve, compreendem o filé mignon.

E-mail: moacir@ufersa.edu.br Galea spixii is a typical rodent of the caatinga belonging to the Caviidae family. Considering the lack of data on the aortic arch of the galea, this study aimed to describe the collateral branches of the aortic arch in this specie and to contributes to knowledge of its biology. Twenty males from previous studies were used which had been stored in a freezer at Multiplication Center of Wild Animals (CEMAS/UFERSA). The animals were thawed, the thoracic cavity was opened and the aorta cannulated vasculature was washed with saline and then injected with Neoprene latex colored with red pigment, yellow or white. Subsequently, the animals were fixed in formalin and after 72 hours dissected and analyzed, to obtain schematic drawings and the most representative samples were photographed. The cavy aortic arch issued as collateral branches, the brachiocephalic trunk and the left subclavian artery. The brachiocephalic trunk afforded in most of the specimens the left common carotid artery and the brachiocarotid trunk, where the right subclavian and right common carotid arteries arise. The right and left subclavian arteries in all animals studied issued the vertebral artery, internal thoracic artery, superficial cervical artery costocervical trunk and axillary artery. The pattern of formation of the aortic arch of galea was similar to that observed in other rodents, such as the kerodon, the guinea pig and chinchilla.

Considerando a inexistência de dados sobre o arco aórtico do preá, foi realizado este estudo tendo como objetivo descrever os ramos colaterais do arco aórtico neste cavídeo, e dessa forma, contribuir com dados para biologia da espécie. Foram utilizados vinte preás machos provenientes de estudos anteriores e encontravam- se armazenados em freezer no Centro de Multiplicação de Animais Silvestres (CEMAS/UFERSA). Os animais foram descongelados, a cavidade torácica foi aberta, a aorta canulada e o sistema vascular lavado com solução salina e em seguida, injetado látex Neoprene corado com pigmento vermelho, amarelo ou branco. Posteriormente, os animais foram fixados em formol e depois de 72 horas, dissecados e analisados, sendo obtidos desenhos esquemáticos e os exemplares mais representativos fotografados. O arco aórtico do preá emitiu como ramos colaterais, o tronco braquiocefálico e a artéria subclávia esquerda. O tronco braquiocefálico originou na maioria das peças estudadas, a artéria carótida comum esquerda e o tronco braquiocarotídeo, do qual surgem as artérias subclávia direita e carótida comum direita. As artérias subclávias direita e esquerda em todos os animais estudados emitiram a artéria vertebral, a artéria torácica interna, a artéria cervical superficial, o tronco costocervical e a artéria axilar. O padrão da formação do arco aórtico do preá assemelhou-se ao observado em outros roedores, tais como no mocó, no porquinho-da-índia e na chinchila.

E-mail: flavialopes@fmva.unesp.br In vitro production (IVP) of bovine embryos is not only of great economic importance to the cattle industry, but is also an important model for studying embryo development. The aim of this study was to evaluate the histone modification, H3R26me2 during pre-implantation development of IVP bovine embryos cultured with or without serum supplementation and how these in vitro treatments compared to in vivo embryos at the morula stage. After in vitro maturation and fertilization, bovine embryos were cultured with either 0 or 2.5% fetal bovine serum (FBS). Development was evaluated and embryos were collected and fixed at different stages during development (2-, 4-, 8-, 16-cell, morula and blastocyst). Fixed embryos were then used for immunofluorescence utilizing an antibody for H3R26me2. Images of stained embryos were analyzed as a percentage of total DNA. Embryos cultured with 2.5% FBS developed to blastocysts at a greater rate than 0%FBS groups (34.85+5.43% vs. 23.38+2.93%; P<0.05). Levels of H3R26me2 changed for both groups over development. In the 0%FBS group, the greatest amount of H3R26me2 staining was at the 4-cell (P<0.05), 16-cell (P<0.05) and morula (P<0.05) stages. In the 2.5%FBS group, only 4-cell stage embryos were significantly higher than all other stages (P<0.01). Morula stage in vivo embryos had similar levels as the 0%FBS group, and both were significantly higher than the 2.5%FBS group. These results suggest that the histone modification H3R26me2 is regulated during development of pre-implantation bovine embryos, and that culture conditions greatly alter this regulation.

O objetivo deste estudo foi avaliar a modificação de histona, H3R26me2 durante o desenvolvimento pré-implantacional em embriões bovinos produzidos in vitro, cultivados com ou sem suplementação de soro fetal bovino (SFB), bem como comparar essa modificação específica entre mórulas produzidas in vitro e in vivo. Após a maturação in vitro e fertilização, embriões foram cultivados com suplementação de 0 ou 2,5% SFB. O desenvolvimento embrionário foi avaliado e embriões foram coletados e fixados em diferentes fases durante o desenvolvimento (2, 4, 8 e 16 células, mórula e blastocisto). Os embriões fixados foram avaliados por imunofluorescência utilizando um anticorpo para H3R26me2. Imagens de embriões corados foram analisadas baseadas na porcentagem do DNA total. Embriões cultivados com 2,5% SFB tiveram uma taxa de desenvolvimento ao estágio de blastocisto maior que o grupo que não recebeu suplementação com SFB (34.85±5,43% vs 23.38±,93%; P<0,05). Níveis de H3R26me2 variaram para ambos os grupos ao longo do desenvolvimento. No grupo 0% SFB, a marcação para H3R26me2 foi mais intensa nos estágios de 4 células (P<0,05), 16 células (P<0,05) e mórula (P<0.05). No grupo 2.5% SFB, apenas os embriões de 4 células tiveram marcação significativamente maior que todas as outras fases (P<0,01). Mórulas produzidas in vivo apresentaram níveis de H3R26me2 semelhantes ao grupo 0% SFB, e ambos foram significativamente maiores que o grupo 2.5% SFB. Estes resultados sugerem que a modificação de histona H3R26me2 é regulada durante o desenvolvimento pré-implantacional de embriões bovinos, e que as condições de cultura alteram de maneira importante esta regulação.

E-mail: angelapiauilino@hotmail.com This study aimed to investigate a possible relationship between alveolar type II cells and the inflammatory response to infection with Leptospira spp., and thus comprise a further element that can be involved in the pathogenesis of lung injury in naturally infected pigs. The study group consisted of 73 adult pigs that were extensively reared and slaughtered in Teresina, Piauí state, and Timon, Maranhão state, Brazil. The diagnosis of leptospirosis was made using the microscopic agglutination test (MAT) aided by immunohistochemistry and polymerase chain reaction. The MAT registered the occurrence of anti-Leptospira antibodies in 10.96% (8/73) of the pigs. Immunohistochemistry allowed for the visualization of the Leptospira spp. antigen in the lungs of 87.67% (64/73) of the pigs. There was hyperplasia of bronchus-associated lymphoid tissue and circulatory changes, such as congestion of alveolar septa, parenchymal hemorrhage and edema within the alveoli. Lung inflammation was more intense (p = 0.0312) in infected animals, which also showed increased thickening of the alveolar septa (p = 0.0006). Evaluation of alveolar type II (ATII) cells using an anti-TTF-1 (Thyroid Transcription Factor-1) antibody showed that there were more immunostained cells in the non-infected pigs (53.8%) than in the infected animals (46.2%) and that there was an inverse correlation between TTF-1 positive cells and the inflammatory infiltrate. There was no amplification of Leptospira DNA in the lung samples, but leptospiral DNA amplification was observed in the kidneys. The results of this study showed that a relationship exists between a decrease in alveolar type II cells and a leptospire infection. Thus, this work points to the importance of studying the ATII cells as a potential marker of the level of lung innate immune response during leptospirosis in pigs.

A SAM registrou a ocorrência de anticorpos anti-leptospiras em 10,96% (8/73) dos suínos. A imunoistoquímica permitiu a visualização de antígenos de Leptospira spp. em pulmões de 87,67% (64/73) dos suínos. Havia hiperplasia do tecido linfoide associado ao brônquio e alterações circulatórias como, congestão do septo alveolar, hemorragia parenquimatosa e edema no interior de alvéolos. Os focos de inflamações pulmonares eram mais numerosos (p=0,0312) nos animais infectados, bem como o espessamento do septo alveolar (p=0,0006). A quantificação de células alveolares tipo II marcadas pelo anticorpo anti-TTF-1 (Thyroid Transcription Factor-1) mostrou que existia mais células imunocoradas em suínos não infectados (53,8%) comparados aos infectados (46,2%) e uma correlação inversa em relação ao infiltrado inflamatório. Não houve amplificação de DNA de Leptospira spp. em amostras de tecido pulmonar, no entanto DNA leptospiral foi observado em rim. Os resultados deste estudo mostraram que existe uma relação entre a diminuição das células alveolares tipo II e a infecção por leptospiras. Dessa forma, este trabalho aponta para a importância do estudo dessas células, como um provável marcador da modulação da resposta imune inata do pulmão na leptospirose em suínos.

E-mail: yuli_melisasierra@yahoo.com The study was carried out to screen and analyze the genetic characteristics of antimicrobial resistance in Campylobacter spp. from poultry sources. A total of 141 strains of Campylobacter isolated from samples of broilers of slaughterhouses in southern Brazil was identified by phenotypic and genotypic methods. Campylobacter isolates were evaluated for its antimicrobial susceptibility and the presence of resistance genes. The strains were investigated for antimicrobial susceptibility against two agents (ampicillin and tetracycline) by disk diffusion method. PCR assay was used to confirm the specie and the presence of ampicillin (blaOXA-61), tetracycline tet(O), and the energy-dependent multi-drug efflux pump (cmeB) genes. Campylobacter jejuni was the most ubiquitous; its presence was determined in 140 samples out of 141 (99.3%), whereas Campylobacter coli was found only in one of the contaminated samples (0.70%). The results obtained showed 65% and 35.5% of Campylobacter isolates resistant to &#946;-lactams and tetracyclines, respectively. The cmeB gene responsible for multidrug resistance was detected in 26 isolates out 141 strains (18.5%). Moreover, 36 out of 141 Campylobacter strains (25.6%) were found to be resistant to at least two different antimicrobia resistance markers (&#946;-lactams and tetracyclines).

E-mail: yuli_melisasierra@yahoo.com O presente estudo foi realizado para examinar e analisar as características genéticas de resistência antimicrobiana de Campylobacter spp. a partir de fontes avícolas. Um total de 141 amostras de Campylobacter isolados em matadouros-frigoríficos de aves do estado do Rio Grande do Sul, Brasil, foi identificado por métodos fenotípicos e genotípicos. Foi analisada a susceptibilidade antimicrobiana e a presença de genes de resistência. As cepas foram testadas para detectar sensibilidade frente a dois antimicrobianos (ampicilina e tetraciclina) pelo método de difusão em disco. A seguir, usando a reação em cadeia da polimerase foi confirmada a espécie e a presença dos genes de resistência à ampicilina (blaOXA-61) e tetraciclina tet(O), assim como a detecção da bomba de efluxo (cmeB). Campylobacter jejuni foi a espécie mais isolada, sua presença foi determinada em 140 amostras (99,3%), e Campylobacter coli foi encontrada em uma única amostra (0,70%). Os resultados obtidos mostraram 65% e 35,5% de Campylobacter isolados resistentes a &#946;-lactâmicos e tetraciclinas, respectivamente. O gene cmeB responsável pela resistência a múltiplos antimicrobianos foi detectado em 26 amostras (18,5%). Neste contexto, 36 das 141 amostras (25,6%) foram consideradas resistentes a dois grupos diferentes de antimicrobianos (&#946;-lactâmicos e tetraciclinas).

E-mail: andrea@ufv.br Surgical site infection (SSI) has been indicated as the third cause of nosocomial infection. The present study aimed to determine the epidemiological profile of SSI and its association with the risk factors. It is a transversal study done at the São João Batista Hospital of Viçosa-MG and at the Surgery Service of the Small Animals Veterinary Hospital of the Universidade Federal de Viçosa-MG, from September 2012 to February 2013. Global SSI rates were 0.7% at the human and 3.46% at the veterinary hospitals. At the veterinary hospital, SSI rates were not related to contamination potential, with clean procedures presenting the greater rates. As for the type of surgery, orthopedic ones are the most common in both hospital and also the ones presenting the greater SSI rates. Surgeries during more than 120 minutes were 15.25% of the total of procedures at the human hospital and are even less common in the veterinary, with 1.26%. Rate of SSI does not seem to be related to surgery duration in this classification. Bacteria isolated from surgical wounds were multi-resistant and the obtained data indicated that no criteria of antibiotic prophylaxis existed, mainly for clean surgeries. This scenario shows that the action of a commission to control nosocomial infection are extremely relevant in order to guarantee reliable data so that the quality of service may be evaluated and thus, promoting a decrease the risk of in post-operative complications.

Trata-se de um estudo transversal, realizado no Hospital São João Batista de Viçosa-MG e na Clínica Cirúrgica de Cães e Gatos do Hospital Veterinário da Universidade Federal de Viçosa-MG, no período de setembro de 2012 a fevereiro de 2013. As taxas globais de ISC foram de 0,7% no hospital humano e 3,46% no veterinário. No hospital veterinário, a taxa de ISC não mostrou relação com o potencial de contaminação, apresentando a maior taxa nos procedimentos classificados como limpos. Quanto ao tipo de cirurgia, as ortopédicas são as mais comuns em ambos os hospitais e também as que apresentam maior taxa de ISC. Cirurgias com duração maior que 120 minutos corresponderam a 15,25% do total de procedimentos no hospital humano e são ainda menos comuns no veterinário, com 1,26%. A taxa de ISC não parece estar relacionada à duração da cirurgia nesta estratificação. As bactérias isoladas das feridas cirúrgicas foram multirresistentes e os dados levantados indicam que não houve critério quanto ao emprego da antibioticoprofilaxia, principalmente nas cirurgias limpas. Este cenário mostra que é de extrema relevância a atuação de uma comissão de controle de infecção hospitalar, a fim de garantir obtenção de dados fidedignos, para que se possa avaliar a qualidade do serviço prestado e assim promover a redução dos riscos de complicações pós-operatórias.

E-mail: nevesrita@hotmail.com.br Mites of the genus Otodectes are parasites found frequently in the ear of dogs and cats, being recognized as the main causers of external otitis in these animals. Three hundred and twenty dogs, deriving of the city of Cuiabá, Mato Grosso, had been examined through the otoscopy and of the examination of parasitological swab, with the objective to evaluate through the bayes’analysis for the estimate of the prevalence, sensitivity and especificity of these methods in the diagnosis of the infestation caused for the mite, using dates informative and not informative a priori.  Each animal was considered an experimental unit. Of the total of examined dogs, 142 (44.37%) had been presented positive for otocariosis. In 100 animals (31.25%) the infestation was diagnosed by the two methods, in 31 (9.69%) only for the examination of parasitological swab, in 11 (3.44%) only for the otoscopy and in 178 (55.62%) animal the result was negative for both the methods. The sensitivity of the otoscopy, when compared with the examination of parasitological swab, was considered lesser, however the especificity of tests they do not differ. The informative model with priori it demonstrated to be most reasonable and considerably reduced the interval of credibility of the estimates of the parameters. It can be concluded that Bayes’analysis is effective in the estimation of prevalence and characteristics of the diagnostic tests.

Conclui-se que a análise bayesiana é efetiva na estimativa da prevalência e características dos testes diagnósticos.

E-mail: bondan@uol.com.br Although ultrastructural characteristics of mature neuroglia in the central nervous system (CNS) are very well described in mammals, much less is known in reptiles, especially serpents. In this context, two specimens of Bothrops jararaca were euthanized for morphological analysis of CNS glial cells. Samples from telencephalon, mesencephalon and spinal cord were collected and processed for light and transmission electron microscopy investigation. Astrocytes, oligodendrocytes, microglial cells and ependymal cells, as well as myelin sheaths, presented similar ultrastructural features to those already observed in mammals and tended to maintain their general aspect all over the distinct CNS regions observed. Morphological similarities between reptilian and mammalian glia are probably linked to their evolutionary conservation throughout vertebrate phylogeny.

E-mail: bondan@uol.com.br Muito embora as características ultraestruturais da neuróglia madura do sistema nervoso central (SNC) sejam bem descritas em mamíferos, muito pouco é conhecido em répteis, especialmente em serpentes. Neste contexto, dois espécimes de Bothrops jararaca foram eutanasiados para análise morfológica das células gliais presentes no SNC. Amostras de telencéfalo, mesencéfalo e medula espinhal foram coletadas e processadas para investigação por microscopia de luz e eletrônica de transmissão. Astrócitos, oligodendócitos, células microgliais e ependimárias, bem como bainhas de mielina, apresentaram características ultraestruturais similares àquelas já observadas em mamíferos e tenderam a manter seu aspecto geral pelas diferentes regiões observadas no SNC. Similaridades morfológicas entre as células gliais de mamíferos e de répteis estão provavelmente ligadas a sua conservação evolutiva ao longo da filogenia dos vertebrados.

E-mail: sanbsantos@gmail.com The aim of the present study was to report the occurrence of members of the Mollicutes class in the reproductive system of dairy cattle in Brazil. Five farms containing dairy cattle were visited in January of 2012. In total, 100 cows of different ages, breeds and stages of lactation were examined in the present study. The cows were part of intensive or semi-intensive management systems and were submitted to mechanical milking or hand milking. The samples were collected after washing the vulvar region with water and soap, and then drying it with paper towels and disinfecting the area with alcohol (70°GL). Vaginal mucous was collected using a sterile alginate cotton swab, which was rubbed on the vagina, as well as the lateral and internal walls. Vulvovaginal mucous samples were cultured in both liquid and solid modified Hayflick´s medium, for mycoplasmas, and UB medium, for ureaplasmas. The PCR assays for Mollicutes and Ureaplasma spp. were performed according to the standard protocols described in the current literature. During isolation, the frequency of Mycoplasma spp. was of 13.0% (13/100) and for Ureaplasma spp. was of 6.0% (6/100). In the PCR assays the frequency of Mollicutes was of 26.0% (26/100) and for Ureaplasma spp. was of 13.0% (13/100) in the dairy cattle studied. This is the first report of these agents in reproductive system of bovine of the Pernambuco state. Further studies are necessary to determine the pathogenic potential and species of these field isolates.

Foram visitadas em janeiros de 2012 cinco fazendas de bovinos leiteiros. Um total de 100 vacas de diferentes idades, raças e estágios de lactação foram examinadas. Os animais foram mantidos em sistema de manejo intensivo e/ou semi-intensivo, sendo submetidos aos sistemas de ordenha manual ou mecânica. As amostras de muco foram colhidas após a lavagem da região vulvar com água e sabão, com posterior desinfecção com álcool (70°GL). O muco vaginal foi colhido com suabe alginado estéril que foi friccionado nas paredes internas da vagina. Em seguida, as amostras foram cultivadas em meio Hayflick´s modificado, para micoplasmas, e em meio UB, para ureaplasmas, ambos caldo e placa. Os ensaios da PCR para Mollicutes e Ureaplasma spp. foram realizados de acordo com protocolo padrão descrito na literatura. No isolamento, a frequência de Mycoplasma spp. foi de 13% (13/100) e para Ureaplasma spp. foi de 6% (6/100). Nas reações da PCR a frequência para Mollicutes foi de 26% (26/100) e para Ureaplasmas spp. foi de 13% (13/100) nos rebanhos bovinos leiteiros estudados. Este é o primeiro relato destes agentes no trato reprodutivo de bovinos no Estado de Pernambuco. Estudos adicionais são necessários para determinar as espécies e o potencial patogênico destes isolados de campo.
