#91 - Histomorphometricand histological evaluations of the bone lesions caused by fluoride in chickens

Abstract in English:

Sodium fluoride was administered in the water to 2 groups of 25 Shaver female poultry. Group A received fluoride from 1 to 120 days of age and group B from 61 to 120 days. Each group was divided into 5 treatments, with 5 chickens each, which received 0, 25, 50, 200, and 400 ppm of fluoride in the water, respectively. AII animals were killed at 120 days of age. For histomorphometric studies the left femur and tibia were used, and for histologic studies the right femur and tibia. In the cortical bone, cortical porosity was slightly increased by fluoride, but the differences with the control group were not significant. Cortical thickness increased in the animals treated with fluoride (p<0,05, r2 = 0,59 ). In the trabecular bone, of group A, trabecular thickness (TT) (p<0,05, r2 = 0,63) and trabecular bone volume (TBV) (p<0,05, r2 = 0,72) increased in treatments receiving 25-200 ppm, and decreased in the 400 ppm treatment, as it was demonstrated by quadratic regression analysis. In group B there was a positive linear correlation on TT (p<0,05, r2 = 0,98) and TBV (p<0,05, r2 = 0,77) with fluoride ingestion. The osteoid surface was also positively correlated with the amount of fluoride ingested by the animals (p<0,001, r2 = 0,80). These results suggest that fluoride improves osteoblastic function resulting in increased osteoid production and bone apposition. In the other hand, in the animals treated with 400 ppm of fluoride for 120 days bone apposition was reduced suggesting a functional alteration of the osteoblasts or improved bone resorption. In the histological study of bone tissue all animals that received fluoride showed an increased trabecular thickness which was more marked in the 200-400 ppm treatments. In chickens treated with 200-400 ppm of fluoride osteoblasts and osteoclasts were more numerous suggesting an increased bone turnover. In cortical bone Haversian canais were narrowed on the periosteal surfaces and resorption spaces were enlarged on the endosteal surfaces.

#92 - Streptomycin increases the adherence on oral epitelial cells of Bacteroides melaninogenicus involved in the periodontal lesions of

Abstract in English:

"Cara inchada" of cattle (CI) is a periodontitis which occurs in calves in several regions of Brazil. lts progressive lesions, caused mainly by Bacteroides melaninogenicus anel Actinomyces pyogenes, could lead to loss of teeth, malnutrition anel possibly death of affected animals. For further analysis of the pathogenesis of this disease the influence of streptomycin on the adherence of the causative bacteria to oral epithelial cells of cattle was determined. Production of streptomycin by soil bacteria (Actinomycetes) in Brazil had been suggested by several authors. ln our studies, streptomycin in subinibitory concentrations increased tenfold the rate of adherence with B. melaninogenicus (from 0.4 to 4.0 bacteria per bovine epithelial cell). On the contrary, streptomycin did not influence significantly the adherence of A. pyogenes nor streptococci of the viridans group. Thus, streptomycin could be a contributing factor to the development of the periodontal infections with B. melaninogenicus.

#94 - Histologic study and isolation of mycobacteria from tuberculosis-like lesions in cattle in southern Brazil

Abstract in English:

One hundred and fifty-nine lesions macroscopically similar to tuberculosis, observed in cattle slaughtered from 13 counties located in southem Rio Grande do Sul, were studied histologically. Tuberculosis was diagnosed in 147 (92.45%), lesions, club-forming granulomas in 7 (4.4%), eosinophilic abscess in 2 (1.26%), and abscess in 3 (1.89%). Fifty-nine specimens with histological lesions of tuberculosis were cultured for mycobacteria, obtaining 40 isolates, all of them classified as Mycobacterium bovis. Based in these results and the data related with condemnation due to tuberculosis in Rio Grande do Sul, the prevalence of the disease in beef cattle in the State during 1980-1987 was estimated in 0.6%. However, marked differences were observed between different areas. The higher prevalences (2.23% to 3.39%) occurred in the regions focated in the coastland of the "Lagoa dos Patos", middle prevalences (0.97% to 1.95%) in the regions located West and Northwest near Porto Alegre, and the lower prevalences in the other regions of the State (0.12% to 0.71 %). The annual prevalence of the disease has been decreasing sistematically from 1980 (0.95%) to 1987 (0.42).

#95 - Experimental poisoning by Cassia occidentalis (Leg. Caes). in horses

Abstract in English:

In experimental studies seven 12 to 30 year old horses were force-fed through a nasoesophagic tube with ground Cassia occidentalis seeds admixed in water. Four horses died after receiving seeds corresponding to 1.5, 1.75 and 2g of seeds per kg (respectively 0.15, 0.175 and 0.2 per cent) of their body weight. A fifth horse was killed in extremis after receiving 3g of seeds per kg of body weight. The clinical disease induced by the plant in these five horses had a course of 4 to 96 hours and included signs of depression, muscle tremors, incoordinated and swaying gait, tachycardia, dyspnea and increase in serum leveis of creatine phosphokinase, aspartate transaminase and gamma glutamyl transferase. The main necropsy f"mdings included a swollen liver with marked lobular pattern in the natural surface and a nutmeg aspect of the cut surface; reddening of the intestinal mucosa; hemorrhages in the adrenal glands; suffusions and hemorrhages in the epi- and endocardium; pulmonary congestion and edema. Histologically there was hepatocellular degeneration and necrosis associated, in some cases, with polimorphonuclear neutrophylic infiltrates. The skeletal muscles had variable degrees of degenerative and necrotic lesions. The clinic-pathological alterations observed in these five horses suggest hepatotoxic and myotoxic principies in the C. occidentalis seeds, and a cumulative effect in those animals which were exposed to doses over 1.75g/kg of their body weight, fractioned up to eigth administrations. Two horses to which 1 and 2g of the seeds per kg were fed, survived. The horse which received 1g of the ground seeds per kg presented a mild muscular disfunction and recovered completely. These two horses were killed for post-mortem examination 30 days after they had received the last administration of seeds. Necropsy findings were negative, and no lesions which could be attributed to the effects of the plant were observed on histopathological examination.

#96 - Tetrapterys spp. (Malpighiaceae) as the cause of death of cattle due to cardiac lesions

Abstract in English:

This study reports on a disease of cattle which occurs over large areas of southeastern Brazil, and whose main characteristics are severe regressive changes and fibrosis of the heart. Clinical signs included edema of the brisket, a prominent pulsating jugular vein and cardiac arrythmia. In the majority of the cases the course of the disease was subacute (1-5 weeks), at times it was more chronic (months), but only exceptionally was it peracute. The most important post-mortem findings, as determined from 14 necropsies, were clearer areas of the heart muscle as seen through the epicardium, and sharp white areas and streaks across most of the cut surface of the myocardium. At times the myocardium seemed harder than normal. In general, liver alterations varied from accentuated lobulation to a nutmeg appearance of the tissue. Subcutaneous edema in the sternal region and in the body cavities were frequently found. Histological examination of the heart revealed intra- and extracelular edema, lysis, an increase in tlie eosinophilia of groups of fibers (incipient necrosis), massive necrosis, apoptosis, fiber atrophy, inflammatory infiltrations, fibrosis and occasionaly myogenic giant cells. In the liver, congestion, cloudy swelling, vacuolization and lysis ofhepatocytes, edema of Disse's space, and portal and centrolobular fibrosis were seen. The disease was experimentally reproduced through daily administration of the fresh sprouts (5-20g/kg during 9 to 55 days) or dried sprouts (20g/kg during 25 days) of Tetrapterys spp. (T. acutifolia and T. multiglandulosa, fam. Malpighiaceae). Administration of the fresh sprouts in smaller daily doses (2.5g/kg during 130 days) or in a single large dose (l00g/kg), as well as repeated administrations of the fresh or dried ripe leaves (20g/k daily for 64 or 30 days) orof the fruits (2.7-10.9g/kg daily for 6 days), only occasionally caused mild symptoms of poisoning. The course of poisoning in the animals that died was subcaute (9-20 days) in six and chronic (1-3 months) in three. Clinically, the animals showed locomotor difficulties, slowness, weakness, edema of the brisket, a prominent pulsating jugular vein and cardiac arrythmia. Post-mortem examination sometimes showed clearer areas of the heart through the epicardium and, in all but one case, whitish areas and streaks on the cut surface of the myocardium. Subcutaneous and body cativy edemas were less common. Histological lesions were present mainly in the heart and were identical to those found in the naturally occurring disease, although they were not as intense. It is concluded that the 2 species of Tetrapterys are the cause of this disease based upon the following: 1) the disease occurs only in pastures where the plants are found; 2) the course of the disease and clinical signs of experimental cases are very similar to those of the spontaneous disease; and 3) in experimental animals it was possible to reproduce the sarne heart lesions as seen in the naturally occurring disease. Case histories and histopathological findings in organs of fetuses studied, suggest strongly that Tetrapterys spp. are also responsible for abortions in cattle.

#97 - Cutaneous pythiosis in horses from Rio Grande do Sul, Brazil

Abstract in English:

Seventeen cases of Pythiosis in horses from Rio Grande do Sul, Brazil, are described. The lesions consisted of subcutaneous granulomas in the limbs, ventral thoracoabdominal wall and face. Specimens from the lesions were obtained through biopsy or at post-mortem examination. The disease is caused by the Pythium sp., and yeast-like Protista organism and the lesions are similar to the cutaneous Habronimiasis. Most cases of Pythiosis were observed in summer and the characteristic lesions consisted of an exuberant chronic inflammatory reaction crisscrossed by numerous irregular channels containing a central yellow and firm necrotic core (kunker). A presumptive diagnosis of Pythiosis was made in H & E preparations based on the findings of linear or round spaces in the necrotic areas which represented ghost images of the organism. Identification of the fungal elements was achieved with the use of silver stains (all cases), direct examination (2 cases) and cultures (2 cases). The morphology of the agent and histopathological lesions in all cases are identical and permit the etiologic diagnosis even in non-cultured lesions.

#98 - Influence of the milk on periodontal lesions of "cara inchada" in calves

Abstract in English:

Five calves of mixed zebu breed, 3 to 4 months old, with active periodontal lesions of "cara inchada" (CI) were confined, during 4 and a half months, to a covered corral on a farm where the disease has been a problem, in the Rondonópolis area of northem Mato Grosso, Brazil. The calves were milk-fed on cows which were kept graúng on Panicum maximum pasture known to induce a relatively high prevalence of CI (more than 30 per cent). As roughage the calves received native grass cut daily in a near "cerrado" (savanna) area. On this type of natural pasture the disease has never been observed. The periodontal lesions of the five animals persisted or worsened, and two of them died. These observations, toghether with data from former studies, suggest that a determinating factor of CI may exist also in the milk of cows kept on pastures where the disease occurs.

#99 - Atypical mycobacteria isolated from intestinal lesions of Bovine Oesophagostomosis

Abstract in English:

It was found that 90% of 1 to 9 year old cattle from southwestem Brazil had nodular intestinal lesions of Oesophagostomosis in the final portion of the ileum and in the cecum. The number of nodules varied from 1 to 53 and their diameter from l to 19 mm. The cut surface of the larger nodules showed a dry yellowish caseous material, and some times fragments of Oesophagostomum radiatum larvae could be observed. Bacteriological examination of 400 samples taken from the intestinal wall with and without lesions resulted in isolation of 42 (5,2%) cultures of atypical mycobateria. Thirty cultures originated (71,4%) from nodular material and 12 (28,6%) from the portion without lesion. Twenty three (54,8%) cultures belonged to the Runyon group IV and 19 (45,2%) to group II and III. There were 4 cultures of Mycobacterium scrofulaceum, 1 of the M. terrae-complex and 8 of M. intracellulare. Fifteen (78,9%) of these cultures were isolated from the nodular lesions and only 4 (21,0%) from the part without nodules. The present results suggest that the nodular lesion caused by Oesophagostomun radiatum larvae is a predisposing factor for the penetration and multiplication of atypical mycobateria in the intestinal wall. Consequently inespecific allergic reactions to tuberculinization may develop.

#100 - Enzymatic and endotoxic activities of bacteria isolated from periodontal lesions of calves with "Cara inchada"

Abstract in English:

Bacteria isolated from periodontal lesions of calves with "cara inchada" were examined for enzymatic and endotoxic activities. Black-pigmented cultures of Bacteroides produced deoxiribonuclease, collagenease, chondroitin sulfatase, fibrinolysin, gelatinase, hyaluronidase, lipase and protease. Trypsin-like proteolytic activities were demonstrated in cultures of black-pigmented asaccharolytic Bacteroides that were also high producer of collagenase. Actinomyces israelii, A. pyogenes and Fusobacterium nucleatum showed weak production of hydrolytic enzymes. Biological activities of lipopolysaccharides extracted from Bacteroides melaninogenicus, Bacteroides spp. and F. nucleatum were examined by the ability to produce a Shwarzman-reaction in rabbits. Endotoxic lipopolysaccharides from F. nucleatum had higher biological activities than those from Bacteroides. These findings suggest a possible role of the bacterial enzymes and endotoxins in the development of periodontal lesions in young cattle.
