The enzootic bovine leukosis (EBL) virus is the etiological agent of a permanent and incurable infection in cattle. The virus integrates into the host’s DNA and replicates continuously, primarily infecting B lymphocytes and negatively affecting the immune response of infected cattle. With oncogenic potential, the virus is responsible for the development of lymphomas in adult cattle; however, neoplasia is uncommon, occurring in approximately 2–5% of infected animals. Most seropositive cattle remain subclinical, which may lead to decreased immunity, particularly when associated with other diseases. This study aimed to estimate herd-level and within-herd seroprevalence of antibodies against EBL in confined dairy cow herds in the northern region of Rio Grande do Sul, Brazil. Four commercial farms using Free Stall or Compost Barn systems were selected by convenience, and all cows older than six months of age were sampled, totaling 750 animals. Serum samples were tested using a commercial blocking enzyme-linked immunosorbent assay (ELISA) for detection of antibodies against EBL virus. Descriptive statistics were used to calculate herd-level and within-herd seroprevalences and their 95% confidence intervals (95% CI). All herds had at least one seropositive animal (herd-level prevalence: 100%). Within-herd seroprevalence varied across the four herds, ranging from 51% in the São Luiz Gonzaga herd to 65.2% in the Salvador das Missões herd, and the overall seroprevalence among all sampled cows was 55.1% (95% CI: 51.2–58.9). These results demonstrate a high level of EBL seropositivity among dairy cows in the four confined herds evaluated in this study in the northern region of Rio Grande do Sul and indicate similarities in management practices and seroprevalence across these farms.

O vírus da leucose enzoótica bovina (LEB) é o agente etiológico de uma infeção permanente e incurável nos bovinos. O vírus integra-se no DNA do hospedeiro e replica-se continuamente, infectando principalmente os linfócitos B e afetando negativamente a resposta imunitária dos animais infectados. Com potencial oncogênico, o vírus é responsável pelo desenvolvimento de linfomas em bovinos adultos; no entanto, a neoplasia é pouco frequente, ocorrendo em aproximadamente 2–5% dos animais infectados. A maioria dos bovinos soropositivos mantém-se assintomática, o que pode levar a uma diminuição da imunidade, sobretudo quando associada a outras doenças. Este estudo teve como objetivo estimar a soroprevalência de anticorpos contra o vírus da LEB em rebanhos leiteiros confinados na região norte do Rio Grande do Sul, Brasil, tanto a nível de rebanho como dentro de cada rebanho. Foram selecionadas quatro fazendas comerciais que utilizam sistemas de confinamento em “Free Stall” ou em “Compost Barn”, por conveniência, e foram amostrados todos os bovinos com mais de seis meses de idade, totalizando 750 animais. As amostras de soro foram testadas através de um ensaio de imunoabsorção enzimática (ELISA) de bloqueio comercial para detecção de anticorpos contra o vírus da LEB. Foram utilizadas estatísticas descritivas para calcular a soroprevalência ao nível do rebanho e dentro de cada rebanho, bem como os seus respetivos intervalos de confiança a 95% (IC 95%). Todos os rebanhos apresentaram pelo menos um animal soropositivo (prevalência ao nível do rebanho: 100%). A soroprevalência dentro de cada rebanho variou entre 51% no rebanho de São Luiz Gonzaga e 65,2% no rebanho de Salvador das Missões, e a soroprevalência global entre todas as vacas amostradas foi de 55,1% (IC 95%: 51,2–58,9). Estes resultados demonstram um elevado nível de soropositividade para o vírus da LEB entre vacas leiteiras nos quatro rebanhos confinados avaliados na região norte do Rio Grande do Sul e indicam similaridade de manejo e de soroprevalência entre as fazendas.

Enzootic bovine leukosis (EBL) is an infectious disease caused by bovine leukemia virus (BLV) that affects cattle worldwide. Agar gel immunodiffusion (AGID) was the reference test for EBL diagnosis for many years, but enzyme-linked immunosorbent assay (ELISA) showed higher sensitivity, was faster to perform, and resulted in an objective reading. However, the importation of ELISA kits is lengthy and expensive, and currently, no AGID kits are available in Brazil. The aim of this work was to standardize an indirect ELISA (iELISA) for EBL diagnosis using BLV antigens produced in Tadarida brasiliensis lung (Tb1Lu) cells, which are Bovine viral diarrhea virus (BVDV) free, unlike fetal lamb kidney (FLK) cells, currently used for this purpose. Following standardization, iELISA results were compared with those obtained by AGID and the commercial Chekit Leucose-Serum ELISA. Compared to AGID, iELISA had 94,44% sensitivity, 75.68% specificity, 79.10% positive predictive value (PPV) and 93.30% negative predictive value (NPV), with 84% concordance and a Kappa index of 0.699. Compared to the Chekit Leucose-Serum ELISA, iELISA showed 92.60% sensitivity, 87.09% specificity, 90.27% PPV and 90,00% NPV, with 90.27% concordance and a Kappa index of 0.801. Taking into account the high agreement with the traditional tests and the absence of non-specific reactions with BVDV, the developed assay could be used as diagnostic method to control EBL in Brazil.

A leucose enzoótica bovina (LEB) é uma doença infecciosa natural dos bovinos com distribuição mundial causada pelo “bovine leukemia virus” (BLV). A imunodifusão em gel de ágar (IDGA) foi considerada por muitos anos o teste de eleição, porém ensaios imunoenzimáticos (ELISA) apresentam sensibilidade mais elevada e leitura mais rápida e objetiva. No entanto, a importação de kits de ELISA é um processo dispendioso e demorado, e atualmente não há kits de IDGA comercialmente disponíveis no Brasil. Desta forma, o objetivo deste trabalho foi padronizar um ELISA indireto (iELISA) para diagnóstico da LEB utilizando antígenos produzidos a partir do cultivo do BLV em linhagem celular Tadarida brasiliensis “lung” (Tb1Lu) livre de “bovine viral diarrhea virus” (BVDV), diferentemente do que acontece com as linhagens “fetal lamb kidney” (FLK) atualmente utilizadas na produção desses antígenos para uso em ensaios sorológicos. Após a padronização do iELISA, os resultados foram comparados com aqueles obtidos por IDGA e pelo ELISA comercial “Chekit Leucose-Serum”. Comparado ao IDGA, o iELISA apresentou 94,44% de sensibilidade, 75,68% de especificidade, valor preditivo positivo (VPP) de 79,1% e valor preditivo negativo (VPN) de 93,3%, com concordância entre os testes de 84% e o índice Kappa 0,699. Quando comparado ao ELISA “Chekit Leucose-Serum”, o iELISA apresentou sensibilidade de 92,6%, especificidade de 87,09%, VPP de 90,27% e VPN de 90%, com concordância de 90,27% e o índice Kappa 0,801. Portanto, devido à alta concordância com os testes tradicionais e ausência da ocorrência de reações inespecíficas com BVDV, o ensaio desenvolvido pode ser utilizado como ferramenta diagnóstica para o controle da LEB no Brasil.

The article reports epidemiological data, clinical signs, and laboratory and pathological findings in 24 cattle with enzootic bovine leukosis observed in the Clinic of Garanhuns, at the Federal Rural University of Pernambuco. The 24 cases represented 0.5% of 4,758 cattle examined from 2000 to 2010. The disease affected 22 (91.7%) females and two males. Twenty one of the animals were dairy (six Holstein, 13 girolando, one Brown Swiss and one Jersey), and three were for meat production (Nelore). Twenty three animals were 3-8 years of age and one was younger. All were raised in confinement or semi-confinement. All animals showed enlarged superficial lymph nodes. Other frequent clinical signs were hyporexia, decreased milk production, progressive weight loss, dehydration, hypomotility of the fore stomachs, and altered scant feces. Exophthalmia, dyspnea, and enlarged uterus were observed with less frequency. Leukocytosis (mean of 34,082 leukocytes/mL) with lymphocytosis (21,814 lymphocytes/mL) and neutrophilia (10,906lymphocytes/mL) was observed in the white blood count. Thirteen bovines were necropsied and 11 were slaughtered. Gross lesions were observed on the superficial lymph nodes of all animals necropsied. Six had lesions in the mesenteric lymph nodes, six in the gut, three in the abomasum, two in the uterus, one in the heart, one in the rumen, one in heart, and one in the liver. Due to the importance of enzootic bovine leukosis it is necessary for the farmers to introduce animals free of the disease and to establish a strict health policy for its control.

Neste trabalho são descritos os sinais clínicos, patologia clínica e patologia de 24 bovinos com leucose bovina enzoótica atendidos na Clínica de Bovinos de Garanhuns da Universidade Federal Rural de Pernambuco. Esses casos representaram 0,5% de 4.758 bovinos atendidos entre os anos de 2000 e 2010. A doença afetou 22 (91,7%) fêmeas e dois machos. Vinte e um animais (87,5%) eram de raças leiteras (seis Holandês, 13 Girolando, um Jersey e um Pardo Suíça) e três (12,5%) eram da raça Nelore. Vinte e três animais (95,8%) tinham idade entre 3 e 8 anos e um era mais jovem. Todos eram criados em regime de confinamento ou semi-confinamento. Clinicamente todos os animais apresentaram aumento dos linfonodos superficias. Outros sinais frequentes foram hiporexia, diminuição da produção de leite, emagrecimento progressivo, escore corporal baixo, desidratação, hipomotilidade dos pré-estômagos e fezes alteradas e em pouca quantidade. Com menor frequência foram observados exoftalmia, dispneia e aumento de volume do útero. No leucograma foi constatada leucocitose média de 34.082/µL, com linfocitose de 21.814/µL e neutrofilia de 10.906/µL. Treze animais foram necropsiados e os demais foram enviados pelos proprietários para o abate. Dos treze animais abatidos todos apresentaram lesões nos linfonodos superficiais, seis nos linfonodos mesentéricos, seis no intestino, três no abomaso, um no coração, um no fígado, um no rúmen, um no útero e um no rim. Diante da importância desta enfermidade e dos prejuízos causados pela mesma é necessário alertar produtores sobre os cuidados a serem tomados durante a aquisição de animais, assim como da necessidade de implantar medidas que evitem a difusão da doença entre as fazendas.

The occurrence of the infection with Bovine Leukosis Virus (BLV) was examined in agar-gel immunodiffusion (AGID) test and with indirect enzyme-linked immunosorbent assay (ELISA), in the State of Pará, Brazil. The blood sera were collected from different breeds including Nelore, Piemontes, Si mental, Holstein Frisian, Indubrasil, Girolanda, Sim brasil and their crossbreedings. The majority of the animals were adults. The overall occurrence of infections was 49.8% (359/721) and 26.0% (174/668) for ELISA and AGID test, respectively. All animal groups examined showed infection in ELISA, however in the AGID test two groups were sera negative.

A ocorrência da infecção pelo Vírus da Leucose Enzoótica dos Bovinos (BLV) no Estado do Pará, foi estudada através do método de imunodifusão em ágar-gel (AGID) e por um ensaio imunoenzimatico (ELISA) indireto, paralelamente. Os exames foram realizados com amostras de soros sanguíneos oriundos de bovinos de diferentes raças sendo a maioria deles adultos. A prevalência observada foi de 49,8% (359/721) no ELISA e 26,0% (174/668) no AGID. Todos os 14 grupos dos animais estudados pelo ELISA indireto, mostraram a existência da infeção, enquanto que pelo método da AGID, dois grupos de animais foram negativos.

The prevalence of infections with Bovine Leukosis Virus in cattle of the Jersey breed, raised in the State of São Paulo, Brazil, was determined with glycoprotein antigen (gp 51) in agar-gel immunodiffusion tests. The over-all prevalence of infections was 49,2% (360/709). The sample arranged according to different age groups showed a higher prevalence in older animals. The values obtained were the following: up to 3 months 45,5% (36/77), 3-6 months 9,8% (6/61), 6-12 months 17.1% (13/76), 12-24 months 24.6% (30/122), 24-48 months 47.6% (81/170), 48-72 months 65.9% (89/135) and older than 72 months 86.2% (106/123). The results showed that there exist foci of the infection by the bovine leukosis virus within the 11 studied municipalities. The prevalence of female animals (51.1% - 346/677) was higher than that found in males (9.4% - 3/32).

Determinou-se a prevalência da infecção pelo Vírus da Leucose dos Bovinos através da prova de imunodifusão radial dupla em gel de àgar utilizando-se o antígeno glicoprotéico (gp- 51), em bovinos da raça Jersey, criados no Estado de São Paulo, encontrando-se prevalência igual a 49,2% (360/709). A estratificação da população em faixas etárias demonstrou ser a prevalência maior nos animais idosos. Os resultados obtidos foram os seguinte: bezerros até 3 meses de idade - 45,5% (35/77), de 3 a 6 meses - 9,8% (6/61), de 6 a 12 meses - 17,1% 03/76), de 12 a 24 meses - 24,6% (30/122), de 24 a 48 mese - 47,6% (81/170), de 48 a. 72 meses - 65,9% (89/135) e com idade maior do que 72 meses - 86,2% (106/123). Os resultados demonstraram a existência de focos de infecção pelo Vírus da Leucose dos Bovinos nos 11 municípios paulistas examinados. A prevalência em fêmeas bovinas (51,1% - 346/677) foi maior do que a observada nos machos (9,4% - 3/32).

Lymphocytosis caused by Enzootic Bovine Leukosis (EBL) has been known since the beginning of the century. For the clinical diagnosis of EBL "hematologic keys" have been largely used. The objective of this paper was to evaluate the hematologic condition of black and white holstein cows infected by the Bovine Leukosis Virus (BLV) giving special attention to lymphocytic morphology. Blood samples from 423 cows reacting to the Agar Gel Immunodiffusion Test (AGID) and from 374 non reacting cows were collected. No statistical differences in polymorphic nuclear leucocyte counts and monocyte counts between reacting and non reacting cows were found, but the AGID-reacting cows showed higher lymphocyte counts than the non AGID-reacting cows. When the morphology of the lymphocytes was analysed it was found that the monocytic cell counts and the double nucleus lymphocyte counts were higer in AGID-reagent animals.

Desde o início do século já era conhecido que os animais afetados pela Leucose Enzoótica dos Bovinos (LEB) apresentavam linfocitose. Esta alteração foi muito usada como recurso auxiliar no diagnóstico clínico da LEB através do emprego das chamadas "chaves leucométricas". O presente estudo procurou avaliar o perfil do quadro leucocitário de fêmeas bovinas da raça holandesa naturalmente infectadas pelo Vírus da Leucose Bovina (VLB) dando-se particular atenção à morfologia dos linfócitos. Para tanto, realizou-se o leucograma de 423 bovinos reagentes e 374 não reagentes à prova de Imunodifusão em Gel para a Leucose Bovina (IDLB). Os resultados obtidos mostraram que apenas os linfócitos apresentaram aumentos estatisticamente significantes nos animais reagentes à IDLB. No tocante ao aspecto morfológico dos linfócitos, conclui-se que os linfócitos monocitóides e os linfócitos com núcleo duplo apresentaram contagens significativamente maiores nos animais reagentes.

A representative sample of blood from dairy cattle in the Sta te of Parana was tested for the presence of antibodies against the virus of enzootic bovine leukosis (EBL) and it was found that the general prevalence of the virus was 20.7%, in 40.8% of the surveyed farms. The higher prevalences of the virus were found in the areas of the State where the economic significance of dairy production is greater and this prevalence was also higher in the older animals and in those of the Holstein breeds.

Uma amostragem representativa de sangue do rebanho bovino produtor de leite do Estado do Paraná foi submetida à prova específica para verificação de anticorpos contra o vírus da leucose enzoótica bovina (LEB), tendo sido encontrado um índice geral de prevalência de 20,7% em 40,8% das propriedades pesquisadas. As maiores prevalências do vírus foram encontradas nas áreas do Estado onde a importância econômica da bovinocultura de leite é maior e esta prevalência foi maior nos animais mais velhos e naqueles das raças Holandesas.

Fifty-five dairy cows from two herds located in the State of Rio de Janeiro, were evaluated monthly over a six month period for persistent lymphocytosis (PL) and for the presence of plasma antibody to the major glycoprotein (gp51) of bovine leukosis virus (BLV), as evidence of infection. Absolute lymphocyte counts were superimposed on Bendixen's hematological key for bovine leukosis. Animals were considered to have PL when, during the six month period, three absolute lymphocyte counts were above 7000 per mm3 of blood. In herd A, of 15 absolute lymphocyte counts from three antibody-negative cows, 13 (86.7%) were normal and two (13.3%) suspect. Counts from 17 antibody-positive cows from the sarne herd showed 29 (30.5%) normal, 36 (37.9%) suspect and 30 (31.6%) leukotic. In herd B, of 82 absolute lymphocyte counts from 14 antibody-negative cows, 45 (54.9%) were normal, 23 (28.1%) suspect and 14 (17.0%) leukotic. Counts from 21 antibody-positive cows showed that 53 (45.3%) were normal, 40 (34.2%) were suspect and 24 (20.5%) were leukotic. It is concluded that PL is an unreliable criterion to identify cattle infected with BLV in the subtropical environment, and must not be used in programs aimed at controlling or eradicating BLV from infected herds. However, the hematological keys will continue to be useful in the diagnosis of the clinical disease, that is, the bovine lymphosarcoma. The use of the immunodiffusion test to detect antibody to gp51 in the plasma or serum of infected cattle is a simple and sensitive tool in the control of BLV infection.

Cinqüenta e cinco vacas leiteiras de dois rebanhos localizados no Estado do Rio de Janeiro foram examinadas mensalmente, durante um período de seis meses. para determinar a linfocitose persistente (LP) e a presença, no plasma, de anticorpos contra a glicoproteina maior (gp51) do vírus da leucose bovina (VLB), como evidência da infecção. As contagens linfocitárias absolutas foram sobrepostas à chave hematológica de Bendixen para leucose bovina. Os animais foram considerados portadores de LP quando, durante o período de seis meses, três contagens linfocitárias absolutas estavam acima de 7000 por mm3 de sangue. No rebanho A, de 15 contagens linfocitárias absolutas de três vacas anticorpo-negativas, 13 (86,7%) eram normais e duas (13,3%) atingiram a faixa de suspeitas. De 95 contagens de 17 vacas anticorpo-positivas do mesmo rebanho, ,29 mostraram-se (30,5%) normais, 36 (37,9%) suspeitas e 30 (31,6%) leucóticas. No rebanho B, de 82 contagens linfocitárias absolutas de 14 vacas anticorpo-negativas; 45 (54,9%) eram normais, 23 (28,1 %) suspeitas e 14 (17,0%) leucóticas. As 117 contagens de 21 vacas anticorpo-positivas mostraram que 53 (45,3%) eram normais, 40 (34,2%) eram suspeitas e 24 (20,5%) eram leucóticas. Concluiu-se que a LP é um critério pouco confiável na detecção de bovinos infectados com o VLB num ambiente subtropical e não deve ser utilizada em programas que objetivam a controlar ou erradicar o VLB de rebanhos infectados. Porém, as chaves hematológicas continuarão sendo úteis no diagnóstico da doença clínica, o linfossarcoma bovino. Ficou também evidente que a prova de imunodifusão para detectar anticorpos contra gp51 no plasma ou soro de animais infectados é um método simples e sensível para o controle da leucose bovina.

Sixty cows out of a total of 482 animals imported from Uruguay by Dairy Cooperative of Curitiba (CLAC) were sampled in order to verify the presence of antibodies against enzootic bovine leukosis (EBL) virus in their blood. All cows were about 30 months old and many of them were calving for the first time. Eleven (18.3%) cows had antibodies against EBL virus in their plasma which were detected using the immunodiffusion test.

Sessenta vacas de um total de 482 animais importados do Uruguai pela Cooperativa de Laticínios de Curitiba (CLAC) foram amostrados com a finalidade de se verificar a presença de anticorpos no sangue contra o vírus da leucose enzoótica bovina (LEB). Todas as vacas tinham aproximadamente 30 meses de idade e muitas delas se apresentavam parindo pela primeira vez. Onze (18,3%) vacas apresentaram anticorpos contra o vírus da LEB quando testados pela prova de imunodifusão.

The agar gel immunodiffusion tests performed in plates and on slides were compared in their efficiency to detect antibodies against the major glycoprotein gp51 of enzootic bovine leukosis virus (BLV). Materiais tested were milk, plasma and serum obtained from cattle belonging to seven dairy herds located in the States of Rio de Janeiro and São Paulo. Both tests were equally inefficient in the detection of antibodies in the milk. The plate immunodiffusion test was more sensitive than the slide test in the detection of antibodies in plasma, although, when the tests were performed with serum, both were equally sensitive in the identification of infected cattle. Out of a total of 1031 sera tested, 361 (35.0%) were identified as positive using the plate test, while the slide test detected 355 (34.4%) positives. It is concluded that- the slide inununodiffusion test can efficiently substitute for the plate immunodiffusion test in the detection of antibodies against gp51 of BLV in the serum, with the additional advantage of utilizing smaller amounts of glycoprotein antigen.

As provas de imunodifusão em ágar gel, realizadas em placa e em lâmina, foram comparadas quanto à eficiência em detectar anticorpos contra a glicoproteína maior gp51 rio vírus da leucose enzoótica bovina (VLB). As amostras testadas constaram de leite, plasma e soro, obtidos de bovinos pertencentes a sete rebanhos leiteiros dos Estados do Rio de Janeiro e São Paulo. Ambas as provas foram igualmente ineficientes na detecção de anticorpos no leite. A prova de imunodifusão em placa foi mais sensível que a prova em lâmina, na detecção de anticorpos no plasma, porém, quando as provas eram realizadas com soro, as duas eram igualmente sensíveis na identificação de bovinos infectados. De um total de 1031 soros testados, a prova em placa detectou 361 (35,0%) soros positivos enquanto a prova em lâmina 355 (34,4%). Concluiu-se que a prova em lâmina pode substituir a prova em placa eficientemente na detecção de anticorpos contra gp51 do VLB no soro, com a vantagem de utilizar menores quantidades de antígeno glicoprotéico.
