#1 - Genotyping of the infectious bursal disease virus (IBDV) based on the hypervariable region of the vp2 gene in broiler chickens from the State of Rio de Janeiro, Brazil

Abstract in English:

Infectious bursal disease virus (IBDV) causes immunosuppression in broilers by affecting the bursa of Fabricius. IBDV strains were classified into seven genogroups based on the amino acid composition of the hypervariable region of the VP2 capsid protein. This study aimed to detect and genotype IBDV strains in bursal samples from broilers in Rio de Janeiro and evaluate the genetic variability of the vp2 gene. Sixty-five bursal tissue samples from broilers suspected to have IBD were collected from four farms in Rio de Janeiro and stored in RNAlater. RNA was extracted using TRIzol reagent, and genotyping was performed using RT-qPCR. The VP2 hypervariable region was amplified using conventional PCR, purified, and sequenced. Phylogenetic analysis was conducted using the maximum likelihood method with the Kimura 2-parameter substitution model. IBDV was detected in 75.38% (49/65) of the samples, with 8.16% (4/49) classified as very virulent (vvIBDV) and 91.84% (45/49) as non-vvIBDV strains. This study seems to be the first report of G-3 infection in the state of Rio de Janeiro. Sequence comparison identified multiple missense mutations, including 20 in genogroup 4 and eight in genogroup 3, relative to the reference strain Edgar-USA. The genetic distances among the isolates suggest potential viral evolution and adaptation. These findings highlight the genetic diversity of IBDV in this region and reinforce the importance of continuous molecular surveillance of the virus. The detection of vvIBDV in a previously unreported area underscores the need for monitoring strategies to mitigate the impact of emerging strains on poultry production. Further studies focusing on antigenic variation and vaccine efficacy are essential to ensure effective control of IBDV.

O vírus da doença infecciosa da bursa (VDIB) causa imunossupressão em frangos de carne, afetando a bursa de Fabricius. As estirpes do VDIB foram classificadas em sete genogrupos com base na sua composição de aminoácidos na região hipervariável da proteína VP2 do capsídeo viral. O objetivo deste estudo foi detectar e realizar a genotipagem de cepas de VDIB em amostras de bursa de frangos de corte no Rio de Janeiro e avaliar a variabilidade genética do gene vp2. Sessenta e cinco amostras de tecido da bursa de Fabricius de frangos com suspeita de DIB foram coletadas em quatro granjas do Rio de Janeiro e armazenadas em RNAlater. O RNA foi extraído usando o reagente TRIzol, e a genotipagem foi realizada usando RT-qPCR. A região hipervariável da VP2 foi amplificada por PCR convencional, purificada e sequenciada. A análise filogenética foi efetuada utilizando o método da máxima verosimilhança com o modelo de substituição Kimura-2 parâmetros. O VDIB foi detectado em 75,38% (49/65) das amostras, com 8,16% (4/49) classificadas como muito virulentas (vvVDIB) e 91,84% (45/49) como estirpes não vvVDIB. Este estudo parece ser o primeiro relato de infeção por vvVDIB no estado do Rio de Janeiro. A comparação de sequências identificou múltiplas mutações de sentido trocado, incluindo 20 mutações no genogrupo 4 e oito no genogrupo 3 em relação à estirpe de referência Edgar-USA. As distâncias genéticas entre os isolados sugerem uma evolução potencial e adaptação viral. Estes resultados realçam a diversidade genética do VDIB nesta região e reforçam a importância da vigilância molecular contínua. A deteção do vvVDIB numa área anteriormente não registada sublinha a necessidade de estratégias de monitorização para mitigar o impacto das estirpes emergentes na produção avícola. São essenciais mais estudos centrados na variação antigênica e na eficácia da vacina para garantir medidas de controle eficazes contra o VDIB.


#2 - Investigation of antimicrobial resistance genes in Staphylococcus aureus from bovine mastitis milk in Southwest Turkey

Abstract in English:

One of the most important pathogens of bovine mastitis, Staphylococcus aureus, often shows antibiotic resistance due to the widespread use of antibiotics in dairy herds. Here, we analyzed S. aureus isolates recovered from milk samples of mastitis-affected cows from various dairy farms for the presence of specific antibiotic resistance genes (mecA, blaZ, aacA-aphD, tetK, tetM, vanA, cfr, fusB, and ileS). To isolate and identify S. aureus, milk was inoculated onto Baird-Parker agar plates. The thermonuclease gene (nuc) was used for molecular identification of each S. aureus isolate. The chosen resistance genes were found in 14 verified isolates. The blaZ and tetM genes were found in all isolates (100%), whereas the aacA-aphD, mecA, and cfr genes were found in 42.9%, 14.3%, and 7.1% of the isolates, respectively. TetK, vanA, fusB, and ileS genes were not found in any of the isolates. Every isolate carried at least blaZ and tetM, demonstrating the widespread co-carriage of several genes. The findings show that S. aureus from bovine mastitis in this area exhibits broad genotypic resistance to β-lactams and tetracyclines, and emphasize the need for ongoing regional surveillance of resistance determinants.

Staphylococcus aureus, um dos patógenos mais importantes da mastite bovina, frequentemente apresenta resistência aos antibióticos devido ao uso generalizado de antibióticos em rebanhos leiteiros. Analisamos isolados de S. aureus recuperados de amostras de leite de vacas afetadas por mastite de várias fazendas leiteiras para a presença de genes específicos de resistência aos antibióticos (mecA, blaZ, aacA-aphD, tetK, tetM, vanA, cfr, fusB, e ileS). Para isolar e identificar S. aureus, o leite foi inoculado em placas de ágar Baird-Parker. O gene termonuclease (nuc) foi usado para a identificação molecular de cada isolado de S. aureus. Os genes de resistência escolhidos foram encontrados em 14 isolados verificados. Os genes blaZ e tetM foram encontrados em todos os isolados (100%), enquanto os genes aacA-aphD, mecA e cfr foram encontrados em 42,9%, 14,3% e 7,1% dos isolados, respectivamente. Os genes TetK, vanA, fusB e ileS não foram encontrados em nenhum dos isolados. Todos os isolados carregavam pelo menos blaZ e tetM, demonstrando o transporte conjunto generalizado de vários genes. Os resultados mostram que S. aureus da mastite bovina nesta região tem ampla resistência genotípica aos β-lactâmicos e tetraciclinas, e enfatizam a necessidade de vigilância regional contínua dos determinantes de resistência


#3 - Detection of anti-Toxoplasma gondii antibodies in Callithrix spp. in Espírito Santo, Brazil

Abstract in English:

Toxoplasmosis is an infectious disease caused by the obligate intracellular protozoan Toxoplasma gondii, which has a worldwide distribution and is recognized for frequently causing acute and fatal disease in neotropical primates. This study aimed to detect anti-T. gondii antibodies in Callithrix spp. from different locations in Espírito Santo, Brazil, and to describe the distribution of seroreactivity according to environmental and host-related variables. A total of 21 animals were evaluated from 2022 to 2025, both sexes and ranging in age from juveniles to adults, including individuals maintained under human care and free-ranging individuals. Seroreactivity was detected using the modified agglutination test in 23.8% (5/21) of the primates, with titers of 1:25 and 1:50. All seroreactive individuals were identified among those classified under human care at the time of sampling, while no reactivity was observed among free-ranging individuals. The distribution of seroreactivity did not suggest a clear pattern in relation to sex or age and should be interpreted with caution due to the structure of the sampled population. The detected pattern was associated with the environmental and management contexts of the municipalities of origin of the animals, with seroreactive individuals linked to areas with greater anthropogenic influence. The low antibody titers observed are consistent with previous reports involving neotropical primates; however, the exclusive use of a serological method and the high susceptibility of this group may limit the detection of infection, particularly in cases of acute disease progression. These findings provide insights into T. gondii exposure in neotropical primates from Espírito Santo and highlight the importance of integrating serological and molecular approaches in future studies to better characterize parasite circulation and epidemiological dynamics in the region.

A toxoplasmose é uma doença infecciosa causada pelo protozoário intracelular obrigatório Toxoplasma gondii, de distribuição mundial, reconhecido por causar com frequência condição aguda e fatal em primatas neotropicais. Este estudo buscou como objetivo detectar anticorpos anti-T.gondii em Callithrix spp. provenientes de diferentes locais do Espírito Santo, Brasil, e descrever a distribuição de sororreatividade de acordo com variáveis ambientais e relacionadas ao hospedeiro. Foram avaliados 21 animais, de 2022 a 2025, de ambos os sexos e com faixa etária variando de juvenil a adulto, sendo 10 mantidos sob cuidados humanos e 11 de vida livre. Foi detectada sororreatividade através do teste de aglutinação modificado em 23,8% (5/21) dos primatas, com titulações de 1:25 e 1:50. Todos os indivíduos sororreagentes foram identificados entre aqueles classificados como mantidos sob cuidados humanos no momento da coleta, enquanto nenhuma reatividade foi observada entre os indivíduos de vida livre. A distribuição da sororreatividade não sugeriu um padrão claro em relação ao sexo ou à idade e deve ser interpretada com cautela devido à estrutura da população amostrada. O padrão observado foi associado aos contextos ambientais e de manejo dos municípios de origemdos animais, com indivíduos sororreagentes associados a áreas com maior influência antrópica. As baixas titulações relatadas são consistentes com a literatura, no entanto, o uso exclusivo de um método sorológico e a alta sucetibilidade desse grupo podem limitar a detecção da infecção, especialmente em casos de progressão aguda da doença. Esses achados fornecem informações relevantes sobre a exposição ao T. gondii em primatas neotropicais do Espírito Santo e destacam a importância da integração de abordagens sorológicas e moleculares em estudos futuros, visando melhor caracterizar a circulação do parasito e a dinâmica epidemiológica na região.


#4 - (Online First) Zoonotic Anaplasma platys in Vietnamese family-kept dogs and dual infections with Anaplasma marginale in cattle

Abstract in English:

Anaplasmosis is a widespread zoonotic tick-borne disease caused by Anaplasma species. This study analyzed 107 blood samples (68 canine and 39 bovine) stained using the Diff-Quik method for Anaplasma morulae. Microscopic evaluation and molecular validation via 16S rRNA gene sequencing identified Anaplasma platys in eight animals (three indigenous cattle, two hybrid dairy cows, and three indigenous dogs) and Anaplasma marginale in six cattle. Notably, one hybrid dairy cow showed dual infection with both Anaplasma species. The pairwise mean genetic distances were largest between A. platys and Anaplasma capra (4.30%) and lowest between A. platys and Anaplasma phagocytophilum (1.59%). Phylogenetic trees confirmed monophyletic Anaplasma and Ehrlichia clades, with Vietnamese A. platys isolates clustering as a sister group to the A. phagocytophilum subclade. The findings in our study confirm the persistent circulation of A. platys in Vietnam, highlighting its emerging zoonotic potential. Consequently, large-scale surveillance across animal hosts, vectors, and humans is essential to clarify the epidemiological status of Anaplasmataceae in the country.

A anaplasmose é uma zoonose disseminada transmitida por carrapatos, causada por espécies de Anaplasma. Este estudo analisou 107 amostras de sangue (68 caninas e 39 bovinas) para detecção de mórulas de Anaplasma. A validação molecular por meio do sequenciamento do gene 16S rRNA e a avaliação microscópica identificaram Anaplasma platys em oito animais (três bovinos nativos, duas vacas leiteiras mestiças e três cães nativos), e Anaplasma marginale em seis bovinos. Notavelmente, uma vaca leiteira mestiça apresentou infecção dupla por ambas as espécies. As distâncias genéticas médias entre pares foram maiores entre A. platys e A. capra (4,30%) e menores com Anaplasma phagocytophilum (1,59%). As árvores filogenéticas confirmaram os clados monofiléticos de Anaplasma e Ehrlichia, com os isolados vietnamitas de A. platys agrupando-se como um grupo irmão do subclado de A. phagocytophilum. Esses resultados confirmam a circulação persistente de A. platys no Vietnã, destacando seu risco zoonótico emergente e a necessidade urgente de medidas aprimoradas de diagnóstico e controle em animais domésticos.


#5 - (Online First) Molecular identification and phylogenetic analysis of emerging foot-and-mouth disease virus type O, topotype ME-SA, in the southwestern region of Pakistan

Abstract in English:

Foot-and-mouth disease virus (FMDV) is among the most contagious and economically significant viral pathogens of livestock worldwide. In Quetta, Balochistan, recurrent FMDV outbreaks continue to affect cattle and buffalo populations, yet genomic data on circulating strains remain scarce. This study investigated the molecular characteristics and phylogenetic relationships of FMDV serotypes and topotypes detected during 2021 surveillance in Quetta, Pakistan. Twenty epithelial tissue samples, previously confirmed as FMDV-positive through sandwich ELISA, were analyzed for viral RNA using real-time reverse transcription polymerase chain reaction (RT-PCR). Positive samples underwent VP1 gene sequencing, followed by phylogenetic analysis. FMDV RNA was detected in 80% (16/20; 95% CI: 56%–94%) of samples. Among infected animals, 87.5% were female, 69% were cattle, and most (63%) were aged 1–4 years. The highest number of cases (25%) originated from the Jan Muhammad Road area. Phylogenetic analysis revealed all isolates belonged to FMDV serotype O, topotype Middle East-South Asia (ME-SA). These findings confirm FMDV serotype O (ME-SA) as the dominant strain causing endemic outbreaks in Quetta. Strengthening active surveillance and molecular monitoring systems is crucial for early detection, effective vaccination planning, and regulation of cross-border livestock movement with Afghanistan, Iran, and other regions of Pakistan.

O vírus da febre aftosa (FMDV) está entre os patógenos virais mais contagiosos e economicamente significativos para o gado em todo o mundo. Em Quetta, Balochistão, surtos recorrentes de FMDV continuam a afetar populações de bovinos e búfalos, porém dados genômicos sobre as cepas circulantes ainda são escassos. Este estudo investigou as características moleculares e as relações filogenéticas dos sorotipos e topótipos de FMDV detectados durante uma vigilância realizada em 2021 em Quetta, Paquistão. Vinte amostras de tecido epitelial, previamente confirmadas como FMDV-positivas por ELISA sanduíche, foram analisadas quanto ao RNA viral usando reação em cadeia da polimerase com transcrição reversa em tempo real (RT-PCR). As amostras positivas foram submetidas ao sequenciamento do gene VP1, seguido de análise filogenética. RNA do FMDV foi detectado em 80% (16/20; IC 95%: 56%–94%) das amostras. Entre os animais infectados, 87,5% eram fêmeas, 69% eram bovinos e a maioria (63%) tinha entre um e quatro anos de idade. O maior número de casos (25%) se originou da área da Jan Muhammad Road. A análise filogenética revelou que todos os isolados pertenciam ao sorotipo O do FMDV, topotipo Middle East-South Asia (ME-SA). Esses achados confirmam que o sorotipo O do FMDV (ME-SA) é a cepa dominante que causa surtos endêmicos em Quetta. Fortalecer a vigilância ativa e os sistemas de monitoramento molecular é crucial para a detecção precoce, planejamento eficaz da vacinação e regulamentação do movimento transfronteiriço de animais com o Afeganistão, Irã e outras regiões do Paquistão.


#6 - Insights into Staphylococcus aureus pathogenesis and genetic diversity in bovine mastitis

Abstract in English:

Bovine mastitis is the most common infectious disease in dairy herds and causes important economic losses. Various pathogens can cause the disease, with Staphylococcus aureus being one of the most significant. S. aureus can cause clinical, subclinical, and chronic forms of the disease due to a variety of virulence factors, including protein A, coagulase, toxins, and adhesins. The presence of methicillin-resistant S. aureus strains complicates treatment protocols and raises concerns about antimicrobial resistance. The genetic characterization of these strains, utilizing techniques such as molecular typing by pulsed-field gel electrophoresis (PFGE), reveals the existence of different genetic and virulence profiles, as well as the geographical variations in the distribution of these genotypes. The role of next-generation sequencing (NGS) technologies, functional genomics, and multi-omics approaches in the genetic research of bovine mastitis are highlighted, providing a complete understanding of its genetic basis and opening opportunities for targeted interventions and improved disease control strategies in the dairy industry. This review highlights the importance of understanding the genetic diversity and pathogenic mechanisms of S. aureus in bovine mastitis to develop effective disease management strategies and reduce economic losses in the dairy sector globally.

A mastite bovina é a doença infecciosa mais comum em rebanhos leiteiros e causa importantes perdas econômicas. Diversos patógenos podem causar a doença, sendo Staphylococcus aureus um dos mais significativos. S. aureus pode causar formas clínicas, subclínicas e crônicas da doença devido a uma variedade de fatores de virulência, incluindo proteína A, coagulase, toxinas e adesinas. A presença de cepas de S. aureus resistentes à meticilina complica os protocolos de tratamento e levanta preocupações sobre a resistência antimicrobiana. A caracterização genética dessas cepas, utilizando técnicas como a tipagem molecular por eletroforese em gel de campo pulsado (PFGE), revela a existência de diferentes perfis genéticos e de virulência, bem como as variações geográficas na distribuição desses genótipos. Destaca-se o papel das tecnologias de sequenciamento de nova geração (NGS), da genômica funcional e das abordagens multi-ômicas na pesquisa genética da mastite bovina, proporcionando uma compreensão mais completa de sua base genética e abrindo oportunidades para intervenções direcionadas e melhores estratégias de controle da doença na indústria de laticínios. Esta revisão destaca a importância de compreender a diversidade genética e os mecanismos patogênicos de S. aureus na mastite bovina para desenvolver estratégias eficazes de manejo da doença e reduzir as perdas econômicas no setor de laticínios globalmente.


#7 - Clinical-laboratory evaluation of overweight and obese cats seen in routine clinical practice

Abstract in English:

Feline obesity has become an increasingly common problem worldwide over the past decade. Excess weight in cats may predispose them to a range of conditions such as insulin resistance, type 2 diabetes mellitus, and hepatic lipidosis. However, few studies have conducted clinical-laboratory profiles of overweight and obese cats. Therefore, the aim of this study was to describe and correlate clinical and laboratory alterations in overweight and obese cats, comparing them with lean cats. Fifty-three cats were evaluated and divided into obese (OB), overweight (SP), and control (CT) groups. After a clinical assessment, the clinically selected cats underwent morphometric measurements and hematological and biochemical tests; their owners were also instructed to complete a questionnaire. Our primary findings included an increase in mean corpuscular volume and total proteins, a decrease in red blood cell count, and an increase in circulating concentrations of total cholesterol, triglycerides, and urea in the OB group; the SP group exhibited an increase only in total cholesterol and urea. Furthermore, the OB and SP groups exhibited a higher frequency of ad libitum feeding, were more likely to receive premium food, and generally had lower activity levels. We concluded that being overweight or obese altered the cats’ hematological and biochemical parameters. Moreover, factors related to the feeding and environmental management of cats may predict an increased risk of being overweight.

Em gatos, a obesidade tornou-se um problema global com prevalência crescente nos últimos 10 anos. O excesso de peso na espécie felina pode predispor a uma série de condições como a resistência à insulina, a diabetes mellitus tipo 2 e a lipidose hepática. Poucos estudos traçaram um perfil clínico-laboratorial de gatos com sobrepeso e obesos. Com isso, o objetivo deste trabalho foi descrever e correlacionar as alterações clínicas e laboratoriais em gatos com sobrepeso e obesos comparando-as com gatos magros. Foram avaliados 53 gatos, divididos nos grupos: obeso (OB), sobrepeso (SP) e controle (CT). Após realizar uma avaliação clínica, os gatos clinicamente selecionados foram direcionados para mensuração de medidas morfométricas, coleta de exames hematológicos e bioquímicos e preenchimento de questionário aplicado ao tutor. Os principais achados foram aumento VCM (volume corpuscular médio) e proteínas totais, redução do número de hemácias, aumento das concentrações circulantes de colesterol total, triglicerídeos e ureia no grupo OB, enquanto o grupo SP mostrou aumento somente de colesterol total e ureia. Ademais, os grupos OB e SP apresentaram maior frequência de alimentação ad libitum, categorizada como premium e gatos com menor nível de atividade. Assim, conclui-se que o sobrepeso e a obesidade alteraram parâmetros hematológicos e bioquímicos. Além disso, fatores relacionados ao manejo alimentar e o ambiental dos gatos podem ser preditivos para um risco aumentado de excesso de peso.


#8 - Phylogenetic analysis of Sporothrix brasiliensis isolated from feline sporotrichosis on São Luís Island, Maranhão, Brazil

Abstract in English:

Sporotrichosis is a zoonotic cutaneous mycosis caused by saprophytic fungi of the Sporothrix, affecting cats, horses, dogs, and humans. This study aimed to evaluate sporotrichosis in cats clinically and to phenotypically characterize and molecularly characterize Sporothrix species on São Luís Island, Maranhão, Brazil. From October 2022 to July 2023, clinical assessments and cytological examinations were performed on suspected feline sporotrichosis cases at the Francisco Edilberto Uchôa Lopes Veterinary Hospital, State University of Maranhão. Lesion exudates were collected via exfoliation or imprinting for fungal culture and species identification. Fungal cultures underwent species-specific polymerase chain reaction (PCR), genetic sequencing, and phylogenetic analysis. A total of 46 cats (33 males and 13 females) were assessed. Disseminated cutaneous sporotrichosis was observed in 70% of cases, with lesions predominantly on the face, ears, thoracic regions, and limbs. Initially, white fungal cultures gradually turned blackish with a coriaceous texture characteristic of Sporothrix spp. PCR amplification of the calmodulin (CAL) gene using Sporothrix brasiliensis-specific primers confirmed all 46 samples as S. brasiliensis. Phylogenetic analysis revealed genetic identity rates ranging from 90% to 100% with S. brasiliensis sequences. This seems to be the first molecular confirmation of S. brasiliensis causing feline sporotrichosis on São Luís Island.

A esporotricose é uma micose cutânea zoonótica causada por fungos saprófitas pertencentes ao gênero Sporothrix que acomete gatos, cavalos, cães e humanos. Este estudo teve como objetivo avaliar clinicamente a esporotricose em gatos e caracterizar fenotípica e molecularmente as espécies de Sporothrix sp. na ilha de São Luís, estado do Maranhão, Brasil. De outubro de 2022 a julho de 2023, avaliações clínicas e exames citológicos foram realizados em casos suspeitos de esporotricose felina no Hospital Veterinário Francisco Edilberto Uchôa Lopes, Universidade Estadual do Maranhão. Exsudados de lesões foram coletados por esfoliação ou imprint para cultura fúngica e identificação de espécies. As culturas fúngicas foram submetidas à reação em cadeia em polimerase (RCP) espécie-específica, sequenciamento genético e analise filogenética. Um total de 46 gatos (33 machos e 13 fêmeas) foram avaliados. Esporotricose cutânea disseminada foi observada em 70% dos casos, com lesões predominantemente na face, orelhas, regiões torácicas e membros. Inicialmente, culturas fúngicas brancas gradualmente tornaram-se enegrecidas com uma textura coriácea característica de Sporothrix spp. A amplificação por PCR do gene calmodulina (CAL) usando primers específicos de Sporothrix brasiliensis confirmou todas as 46 amostras como S. brasiliensis. A análise filogenética revelou taxas de identidade genética variando de 90% a 100% com sequências de S. brasiliensis. Esta parece ser a primeira confirmação molecular de S. brasiliensis causando esporotricose felina na Ilha de São Luís.


#9 - Safety and efficacy of allogeneic bone marrow mesenchymal stem cells for treatment of canine leukopenia induced by canine parvovirus infection

Abstract in English:

This study aims to establish a therapy strategy for canine leukopenia induced by canine parvovirus (CPV) infection through intravenous infusion of allogeneic bone marrow mesenchymal stem cells (BMMSCs) and to evaluate the therapeutic effect of BMMSCs on canine parvovirus. Forty healthy 2-month-old dogs were randomly divided into four groups including the BMMSC treatment group (A), conventional treatment group (B), CPV infection group (C), and a normal control group (D). Then the A, B, and C groups were orally infected with CPV (103.25 TCID50/mL) at 1mL/kg, and the D group received the same dose of saline. After the onset of infection, Group A received mesenchymal stem cells (MSCs) and rehydration as the treatment; Group B was treated with anti-inflammatory therapeutics and rehydration; and Group C and D were injected with the same dose of physiological saline. The level of leukocytes rebounded significantly after the treatment with BMMSCs and returned to reference numbers on Day 3 after treatment, which was significantly higher than that in the conventional treatment group. The concentrations of IL-2 and IFN-α were gradually increased during the treatment, and the BMMSC treatment group exhibited significantly higher IL-2 and IFN-α concentrations than the conventional treatment group on Days 3 and 4. The expression of the virus in the blood gradually decreased during the treatment, and the BMMSC treatment group displayed a faster decrease than the conventional treatment group. These results showed the advantages of BMMSC treatment over conventional treatment. This study provides a new BMMSC treatment strategy for canine leukopenia induced by CPV infection and reveals the mechanism by which BMMSC increases leukocytes after CPV infection.

Este estudo tem como objetivo estabelecer uma estratégia terapêutica de leucopenia canina induzida pela infecção por parvovírus canino (CPV) através de infusão intravenosa de células tronco mesenquimais da medula óssea alogênica (BMMSCs) e avaliar o efeito terapêutico de BMMSCs no parvovírus canino. Quarenta cães saudáveis de dois meses de idade foram divididos aleatoriamente em quatro grupos: o grupo de tratamento de BMMSCs (A), o grupo de terapia convencional (B), o grupo de infecção por CPV (C) e um grupo controle (D). Os grupos A, B e C foram infectados oralmente com CPV (103.25 TCID50/mL) a 1mL/kg e o D recebeu a mesma dose de soro fisiológico. Após o início da infecção, o grupo A recebeu uma dose de derivadas da medula óssea (CTMMO) e hidratação como tratamento, o grupo B foi tratado com terapia anti-inflamatória e hidratação, e grupos C e D foram injetados com a mesma dose de soro fisiológico. As concentrações de IL-2 e IFN-α aumentaram gradualmente durante o tratamento, e o grupo de tratamento BMMSC mostrou concentrações significativamente maiores de IL-2 e IFN-α do que o grupo de tratamento convencional nos Dias 3 e 4. A expressão de vírus no sangue diminuiu gradualmente durante o tratamento, e o grupo de tratamento BMMSC mostrou uma diminuição mais rápida do que o grupo de tratamento convencional. Esses resultados mostraram as vantagens do tratamento BMMSC em relação ao tratamento convencional. Este estudo fornece uma nova estratégia de tratamento da BMMSC para leucopenia canina induzida pela infecção por VCP e revela o mecanismo pelo qual a BMMSC aumenta os leucócitos após infecção por VCP.


#10 - Detection of virulence genes, antibiotic resistance genes and antibiotic resistance of Escherichia coli isolated from diarrheic lambs in Anhui Province, China

Abstract in English:

Lamb diarrhea seriously restricts the development of the sheep industry. Infectious pathogens often cause diarrhea, and E. coli isolates are highly distributed in infectious diarrhea. The study aimed to investigate the prevalence of pathogenic E. coli in diarrhea lambs and determine the distribution of virulence genes, antibiotic resistance genes, and antibiotic resistance. One hundred seventy-eight E. coli isolates were isolated from the feces of 204 diarrhea lambs. The virulence genes mdh, hlyF, iss, ompA, fimC, iucD, st, lt, stx1, stx2, and antibiotic resistance genes including tetA, tetB, aac (6′)-II, blaCMY-2, qnr, aadA1, sul1, blaTEM, blaCTX-M were detected by polymerase chain reaction (PCR). Antibiotic resistance was determined by Kirby-Bauer Disc Diffusion method. There were 109 (61.24%, 109/178) enterotoxigenic E. coli (ETEC), 119 (66.85%, 119/178) Shiga toxin-producing E. coli (STEC), and 95 (53.37%, 95/178) hybrid STEC/ETEC isolates. The highest prevalent virulence genes were ompA (80.90%, 144/178) and fimC (67.98%, 121/178), and the lowest was iucD (8.99%, 16/178). The most commonly detected antibiotic resistance genes were tetA/tetB (92.70%, 165/178), qnr (75.84%, 135/178), and sul1 (62.92%, 112/178), no blaTEM or blaCTX-M genes were detected. All isolates had high antibiotic resistance to lincomycin (96.63%, 172/178), tetracycline (88.76%, 158/178), and co-trimoxazole (80.34%, 143/178), and the multidrug resistant (MDR) rate reached 93.82% (167/178). The high prevalence of ETEC and STEC indicates that E. coli is one of the critical pathogenic agents leading to diarrhea in lambs in the region, and its high antibiotic resistance, especially MDR, should be brought to our attention.
