#661 - Freqüência de anticorpos contra Borrelia burgdorferi em cães na região metropolitana do Rio de Janeiro

E-mail: fabiolanc@ig.com.br Lyme disease is a worldwide zoonosis caused by Borrelia burgdorferi (Spirochaetae) and transmitted through ticks (Ixodidae). Many species of domestic and wild animals are affected. The dog is an important reservoir in the domiciliary environment. The disease affects the joints and causes alterations in other organs. Blood serum samples from 143 dogs, living in the counties of Seropédica, Itaguaí and Rio de Janeiro, were analyzed with the indirect ELISA test, to search for antibodies against Borrelia burgdorferi latu sensu. Average positive results were 48.25%, 41.9% in Seropédica, 4.9% in Itaguaí and 1.4% in Rio de Janeiro. The titres varied from 1:400 to 1:6400. The prevalence was lower (p< 0.05) in dogs up to one year of age than in older dogs, with significant difference. Between males and females was no statistical difference with positives titres. The result of this survey should draw attention to the occurrence of Borrelia sp in the area studied, considering the importance of Lyme disease as an emerging zoonosis.

E-mail: lafranco@vet.ufg.br With the objective to investigate a possible association between foot diseases, clinical mastitis and/or metritis and predisposing factors for foot diseases, 5300 dairy cows from 80 intensive and semi-intensive farms were used. In 325 (6.13%) cows only foot disease was diagnosed, in 35 (0.66%) foot disease and clinical mastitis, in 52 (0.98%) foot disease and metritis, in 28 (0.53%) foot disease, clinical mastitis and metritis, in 128 (2.42%) only metritis, in 165 (3.11%) only clinical mastitis, and in 89 (1.68%) cows metritis and clinical mastitis. Rapid changes in the diet, high exposure time of hoof horn to slurry and wet conditions, concrete floors, use of footbaths, low frequency of claw trimming, irregular quarantine, and no attention to health aspects during acquisition of the animals were considered as major risk factors for the occurrence of those diseases. The McNemar test for dependent samples showed significant statistical differences between the occurrence of foot disease, clinical mastitis and metritis, in spite of the poor association between those diseases. It was concluded that there was no expressive relationship between foot diseases, clinical mastitis and metritis in lactent cows.

E-mail: armando@cnpsa.embrapa.br An epidemiological study in 60 units of swine termination (UT), pertaining to producers integrated with the main Agribusiness of the States of Santa Catarina and Rio Grande do Sul. These units had been selected due to the occurrence of lymphadenitis in slaughtered pigs. The batch size studied in each UT varied from 22 to 960 pigs. The animals were considered positive or negative for lymphadenitis, following the evaluation routine of the Service of Federal Inspection (SIF) at slaughter. For data collection at the UTs, a questionnaire with 156 variables was applied at a single visit to the farms. In the 60 UT studied, the percentage of positive animals for lymphadenitis was 10.37%. Among the variables analyzed, nine risk factors were identified (explicative variables) and associated with the occurrence of lymphadenitis (objective variables). The explicative variables were: bad hygiene of the feeders and drinking fountains, untreated drinking water, bad conservation of the facilities, use of the same truck for ration and animal transportation, production of ration inside the farm, free access of other animals to the ingredients of ration, storage of ready usable ration in boxes or bags and handling of the production of animals in an continuous system.

E-mail: rmtmed@cstr.ufpb.br Three outbreaks ofnitrate poisoning are reported from the semiarid region ofthe state of Paràíba, northeastem Brazil. One outbreak caused by Echinochloa polystachya (capim-mandante) and two others caused by Pennisetum purpureum (capim-elefante) occurred at the end ofthe dry season, after the first raios. ln one ofthe outbreaks caused by Pennisetum purpureum part of the pasture had been fertilized with cattle manure. Five out of 11 cattle died on one farm, 27 out of81 on another, and 3 out of9 on a third one. Clinicai sigos were anorexia, respiratory distress, teeth grinding, depression or hyperexitability, tremors, abdominal contractions, salívation, nasal discharge, uncoordinated gait, cyanosis, and finally recumbency. Nitrates and nitrites in the blood of affected cattle and pastures were determined by the diphenylamine test. It is suggested that the main reason for nitrate accumulation in the grasses was the prolonged draught followed by rain. ln one ofthe outbreaks caused by Pennisetum purpureum, another reason was probably the fertilization of the soil with cattle manure.

Em um dos surtos causado por Pennisetum purpureum, uma parte da área onde estava o pasto que continha níveis altos de nitratos havia sido fertilizada com esterco de bovino. No primeiro surto morreram 5 bovinos de um total de 11, no segundo morreram 21 de um total de 81 e no terceiro morreram 3 de um total de 19 bovinos. Os sinais clínicos se caracterizaram por anorexia, dispnéia, ranger de dentes, depressão ou hiperexitabilidade, tremores, contrações abdominais, salivação, corrimento nasal, andar cambaleante, mucosas cianóticas e, finalmente, decúbito. A presença de nitratos e nitritos foi detectada no sangue dos animais e nos pastos por meio da prova de difenilamina. Parece que o principal fator que determinou a concentração de altos níveis de nitratos nas plantas foi a ocorrência de chuvas depois de um longo período de seca. Outro fator importante no surto causado por Pennisetum purpureum foi a fertilização do solo com esterco.

E-mail: proehe@vottex.ufrgs.br The antigenic profile of 45 herpesviruses (44 viruses from cattle, including six reference BHV-1 strains and 15 putative BHV-1; three reference BHV-5 strains and 20 putative BHV-5) and one butfalo isolate (BuHV) were examined with a panei of monoclonal antibodies (Mabs) prepared against bovine herpesvirus antigens. Tests were performed by immunoperoxidase (IPX) on infected cell cultures, with the Mabs as primary antibodies. lmmunostaining allowed the differentiation between types 1 and 5 viruses. Ali isolates from cases of encephalitis displayed BHV-5 profiles. Four BHV-5 isolates obtained from geographically distinct áreas displayed different and highly variable IPX patterns of reactivity. Two viruses with BHV-5 antigenic profile were isolated from semen of asymptomatic bulis. The results showed that the antigenic characterization with the Mab panei employed here is useful for typing BHV-1 and BHV-5 isolates.

Duas amostras de vírus com perfil antigénico de BHV-5 foram isoladas de sêmen de animais infectados. Estes resultados comprovam a utilidade da caracterização antigénica com este painel de Acms na tipagem de amostras de BHV-1 e BHV-5.


#666 - Experimental poisoning by the burs of Stryphnodendron obovatum (Leg. Mimosoideae) in cattle. 1. Clinical aspects, 21(1):9-17

Clinical aspects] Intoxicação experimental pelas favas de Stryphnodendron obovatum (Leg. Mimosoideae) em bovinos. 1. Caracterização do quadro clínico. Pesquisa Veterinária Brasileira 21 (1):9-17. Projeto Sanidade Animal Embrapa/UFRRJ, Km 47, Seropédica, RJ 23851-970, Brazil. In order to contribute to the characterization of the clinical-pathological picture of poisoning by Stryphnodendron obovatwn Benth. (Leg. Mimosoideae), the pods of the tree were given orally to 17 young bovines. Six animals received the pods only once: Those which received doses of 10 and 20 g/kg, did not show any symptoms of poisoning. Those which receiveel 30, 40 and 40 g/kg of pods, showeel respectively moelerate, severe anel slight symptoms of poisoning, but all recovereel. The animal that was feel 60 g/kg, elieel. Eleven bovines receiveel repeateel doses of the pods: Two of them, which received 2,5 g/kg during 30 days, showed discrete to slight symptoms. Two animals received 5 g/kg during 13 anel 14 days anel showed moderate to severe poisoning. Three animals received 10 g/kg during 20, 8 anel 6 days; the first of them showed slight symptoms, the second died anel the third showed moderate to severe symptoms. Two animals received 20 g/kg during 2 anel 3 days; the first showed slight symptoms, the second died. The two bovines which were fed 30 anel 40 g/kg during 2 days, elieel. First symptoms of poisoning were observed from the first day of the experiments on, anel the clinical course varied from 3 to 63 days. Digestive disorders preelominateel in the clinical-pathological picture. Symptoms were mainly loss of appetite up to anorexia, faeces slightly elry to liquid, elistention of the abdomen without tympanism, loss of ruminal tluid during rumination, decrease in ruminal activity up to atonia, ruminal acidosis, gastro-intestinal colics, sialorrhoea, apathy, loss of weight, elebility, erosions anel ulcers of the oral mucosa. There was congestion of the visible mucous membranes (without icterus) anel of the interdigital parts of the hoofs, areas of focal alopecia anel/or hypotrichia in the axillary regions, the lateral parts of the tighs anel members, loss of hair on the tip of the tail, alterations which can be interpreted as slight photosensitization. In two bovines aspiration pneumonia was verifiecl. In three animals intermittent prolapse of the prepuce was seen, as well as polacuria; sometimes these animals sighed during urination. In all experimental animals the urine was generally turbiel, aciel, dark ambar, with a sweat smell anel high elensity, anel there was proteinuria; in a few animals glicosuria was eliagnoseel. Bilirubine values were normal. Thus, the severe photosensitization reporteei in the literature could not be reproeluceel. Loss of weight, slight photosensitization anel absence of icterus, observed in our experiments, are in accordance with the symptoms reporteei by farmers of the Cerrado-regions (Savanna) where S. obovatum occurs.

Com o objetivo de melhor caracterizar o quadro clínico na intoxicação experimental por Stryphnodendron obovatum Benth., procedeu-se a administração das favas, por via oral, a seis bovinos, em doses únicas e a 11 outros, em doses repetidas. Doses únicas de 10 e 20g/kg não provocaram sintomas. Doses únicas de 30 e 40g/kg provocaram quadros clínicos desde leves até graves, porém só morreu o animal que ingeriu 60g/kg. Os animais que ingeriram doses repetidas de 2,5g/kg por 30 dias adoeceram levemente, já os que receberam 5g/kg durante 13 e 14 dias mostraram sintomatologia moderada e grave, respectivamente. Um bovino que recebeu 10g/kg por 8 dias, outro que ingeriu 20g/kg por 3 dias e dois outros que receberam 30 e 40g/kg por 2 dias morreram ela intoxicação. Os outros três bovinos que receberam doses repetidas (dois com doses de 10g/kg por 20 e 6 dias, outro com dose de 20 g/kg por 2 dias) adoeceram, mas se recuperaram. Os primeiros sintomas de intoxicação, em ambos os grupos, foram observados a partir do primeiro dia do experimento e a evolução variou de 3 a 63 dias. Os sintomas consistiram em anorexia, fezes levemente ressecadas a líquidas, distensão do abdômen, sem timpanismo, perda de fluido ruminal durante a ruminação, atonia e acidose ruminal, cólica, sialorréia, apatia, emagrecimento, fraqueza, erosões e úlceras na cavidade oral. Em alguns animais foram observadas congestão de mucosas visíveis (sem icterícia) e de partes despigmentadas da pele, do córion laminar e ela região interdigital cios quatro membros, áreas ele alopecia focal e/ou hipotricose nas regiões axilares, na face lateral elas coxas e membros e perda dos pêlos ela ponta ela cauda, alterações estas que podem ser interpretadas como as ele leve fotossensibilização. Em três bovinos se observaram relaxamento intermitente cio prepúcio, micção freqüente e em gotejamento e, por vezes, gemidos ao eliminar a urina que, em geral, se apresentava turva, ácida, ele cor âmbar-escura, com odor adocicado e com a densidade elevada; os níveis ele bilirrubina na urina foram normais.

Rabbits are susceptible to bovine herpesvirus type 5 (BHV-5) infection and often develop an acute and fatal neurological disease upon intranasal inoculation. The kinetics of viral infection of the central nervous system (CNS) was investigated by testing serial brain sections for infectivity at intervals after virus inoculation. The virus was first detected in the main olfactory bulb at 48h, followed by the olfactory cortex at 48/72h. At 72/96h infectivity was also detected in the trigeminal ganglia, pons and cerebral cortex. Two experiments were conducted to investigate the role of the olfactory system in the invasion of the rabbits&rsquo; CNS by BHV-5. In the first experiment, rabbits were inoculated with two BHV-5 isolates in the conjunctival sac. Rabbits inoculated by this route developed the neurological disease, yet with a reduced frequency and delayed clinical course. In a second experiment, twelve rabbits were submitted to surgical removal of the olfactory bulb and subsequently inoculated intranasally with BHV-5. Eleven out of 12 (91.6%) of the control rabbits developed the disease, against four out of 12 (33.3%) of the animals lacking the olfactory bulb. These results suggest that the olfactory system is the main pathway utilized by BHV-5 to reach the CNS of rabbits after intranasal inoculation. Nevertheless, the development of neurological infection in rabbits inoculated in the conjunctival sac and in rabbits lacking the olfactory bulb indicate that BHV- 5 may utilize an alternative route to invade the CNS, probably the sensory and autonomic fibers of the trigeminal nerve. The effects of immunization with homologous (BHV-5) and heterologous (BHV-1) strains in prevention of neurological disease by BHV-5 were investigated. Five out of 10 rabbits (50%) immunized with BHV-5 showed mild and transient neurological signs and one died upon challenge. Interestingly, the degree of protection against BHV-5 challenge was higher in rabbits immunized with BHV-1: only two rabbits showed transiente neurological signs and subsequently recovered. Thus, prevention of neurological disease by BHV-5 in rabbits may be achieved by immunization with either BHV-5 or BHV-1, likely reflecting the extensive serological cross-reactivity between these viruses. Further studies in rabbits may help in understanding the pathogenesis and immune response to BHV-5 infection.

Coelhos são susceptíveis à infecção pelo herpesvírus bovino tipo 5 (BHV-5) e freqüentemente desenvolvem enfermidade neurológica aguda fatal após inoculação intranasal. A cinética da invasão do sistema nervoso central (SNC) de coelhos pelo BHV-5 foi estudada através de pesquisa de vírus em secções do SNC a diferentes intervalos pósinoculação. Após inoculação intranasal, o vírus foi inicialmente detectado no bulbo olfatório às 48h, seguido do córtex olfatório às 48/72h. Às 72/96h o vírus foi detectado também no gânglio trigêmeo, ponte e córtex cerebral. Dois experimentos foram realizados para avaliar a importância do sistema olfatório na invasão do SNC de coelhos pelo BHV-5. No primeiro experimento, coelhos foram inoculados com duas amostras do BHV-5 no saco conjuntival. Coelhos inoculados por essa via também desenvolveram a enfermidade neurológica, porém com menor freqüência com curso clínico tardio. No segundo experimento, doze coelhos foram submetidos à ablação cirúrgica do bulbo olfatório e posteriormente inoculados com o BHV-5 pela via intranasal. Onze de 12 coelhos controle (91,6%), não submetidos à cirurgia, desenvolveram a doença neurológica, contra quatro de 12 (33,3%) dos animais submetidos à remoção cirúrgica do bulbo olfatório. Esses resultados demonstram que o sistema olfatório constitui-se na principal via de acesso do BHV-5 ao encéfalo de coelhos após inoculação intranasal. No entanto, o desenvolvimento de infecção neurológica em coelhos inoculados pela via conjuntival e em coelhos sem o bulbo olfatório indica que o BHV-5 pode utilizar outras vias para invadir o SNC, provavelmente as. fibras sensoriais e autonômicas que compõe o nervo trigêmeo. Os efeitos da imunização com vírus homólogo (BHV-5) e heterólogo (BHV-1) na proteção à infecção neurológica foram investigados. Cinco entre 10 coelhos (50%) imunizados com o BHV-5 apresentaram sinais neurológicos discretos e transitórios e um morreu após o desafio com o BHV-5. Curiosamente, o grau de proteção foi superior nos coelhos imunizados com o BHV-1: apenas dois animais apresentaram sinais clínicos passageiros e recuperaram-se. Portanto, proteção da enfermidade neurológica pelo BHV-5 em coelhos pode ser obtida por imunização com o BHV-5 ou BHV-1, provavelmente devido à extensa reatividade sorológica cruzada entre esses vírus. Estudos adicionais em coelhos podem auxiliar no esclarecimento da patogênese e resposta imunológica a infecção pelo BHV-5.

In the sixties and seventies there was an extraordinary increase in export of horse meat products to Europe andjapah. This favored an increase in risk of Salmonella outspread through those products to human and animal consumer populations. Thus, from an exporting company dealing with horse meat located in northeastern Brazil (state of Pernambuco), 19,238 fragments of more external muscles, Salmonella was isolated from 666 samples colleted from 433 animals (horses and donkeys). The serotyping of 745 isolates showed 98 serovars pertaining to 14 serogroups, predominantly classified into subspecies 1 (98.9%). S. Anatum, S. Carrau, S. Saintpaul, S. Agona, and S. Typhimurium were the most frequent serovars isolated. Preliminary data indicate that the primary causes for the presence of Salmonella in the meats probably was contact with feces from slaughtered animals, as well a.s possible contamination of environments, in view of the absence of human carriers researched in part of the personnel.

Nas décadas de 60 e 70, houve um extraordinário incremento da exportação de produtos cárneos de equídeos dos países da América do Sul para a Europa e Japão. Este acontecimento favoreceu o aumento de risco da veiculação de Salmonella através desses produtos, para as populações humana e animal, consumidoras. Assim, num estabelecimento industrial e exportador de carne de eqüídeos localizado no nordeste do Brasil (Pernambuco), foram analisados bacteriologicamente, 19.238 fragmentos de músculos mais externos, que revelaram 666 exames positivos referentes a 433 animais (eqüinos e asininos) e resultando no isolamento de 745 cepas de Salmonella. Na amostragem foram·caracterizados do ponto de vista antigénico 98 sorovares, predominantemente classificados na subespécie 1 (98,9%) e tendo como os mais freqüentes S. Anatum, S. Carrau, S. Saintpaul, S. Agona e S. Typhimurium. Pelas análises efetuadas admite-se que as causas primordiais da presença de Salmonella nas carnes, provavelmente decorreu do contato com os excretas dos animais abatidos, bem como pela possível contaminação ambiental resultante, tendo em vista a ausência de portadores humanos, pesquisados numa parcela do pessoal.

Feline immunodeficiencyvirus (FIV) infection of domestic cats is one of the most promising animal models for the infection by the human immunodeficiency virus (HIV) which causes acquired immunodeficiency syndrome (AIDS). lnfected cats may develop a disease similar to that observed in AIDS patients, with increased susceptibility to opportunistic infections. Ln this study we used the polymerase chain reaction (PCR) to detect proviral DNA of feline immunodeficiency virus on the blood and tissue samples from cats with a clinical diagnosis of immunodeficiency. The PCR primers were used to amplify the gag gene, which is conserved among different isolates. From 40 samples analyzed, 15 were positive and 4 of them were submitted to hybridization to confirm the specificity of the amplified fragments. These results confirm the presence of FIV in domestic cats in Rio Grande do Sul, Brazil.

A infecção de gatos domésticos pelo Vírus da Imunodeficiência Felina (FIV) é um dos modelos mais promissores para o estudo da infecção pelo vírus da imunodeficiência humana (HIV) que causa a Síndrome de Imunodeficiência Adquirida (AIDS). O FIV causa, em gatos, uma enfermidade similar àquela observada em pacientes com AIDS, sobretudo no que diz respeito ao aumento da susceptibilidade a infecções oportunistas. No presente estudo, utilizou-se a Reação em Cadeia da Polimerase (PCR), com o objetivo de detectar o provírus do FIV em gatos com sinais clínicos de imunodeficiência. O fragmento de DNA escolhido como alvo para amplificação situa-se no gene gag do lentivírus felino, o qual é conservado entre as diferentes amostras do vírus. O DNA utilizado foi extraído a partir de amostras de sangue e de tecidos de animais com suspeita clínica de imunodeficiência. Das 40 amostras analisadas, 15 foram positivas, das quais 4 foram submetidas à hibridização, confirmando a especificidade dos fragmentos amplificados. Esses resultados demonstram a presença do FIV na população de gatos domésticos do Rio Grande do Sul, Brasil.

The occurrence of the infection with Bovine Leukosis Virus (BLV) was examined in agar-gel immunodiffusion (AGID) test and with indirect enzyme-linked immunosorbent assay (ELISA), in the State of Pará, Brazil. The blood sera were collected from different breeds including Nelore, Piemontes, Si mental, Holstein Frisian, Indubrasil, Girolanda, Sim brasil and their crossbreedings. The majority of the animals were adults. The overall occurrence of infections was 49.8% (359/721) and 26.0% (174/668) for ELISA and AGID test, respectively. All animal groups examined showed infection in ELISA, however in the AGID test two groups were sera negative.

A ocorrência da infecção pelo Vírus da Leucose Enzoótica dos Bovinos (BLV) no Estado do Pará, foi estudada através do método de imunodifusão em ágar-gel (AGID) e por um ensaio imunoenzimatico (ELISA) indireto, paralelamente. Os exames foram realizados com amostras de soros sanguíneos oriundos de bovinos de diferentes raças sendo a maioria deles adultos. A prevalência observada foi de 49,8% (359/721) no ELISA e 26,0% (174/668) no AGID. Todos os 14 grupos dos animais estudados pelo ELISA indireto, mostraram a existência da infeção, enquanto que pelo método da AGID, dois grupos de animais foram negativos.
