#701 - Poisoning by Palicourea grandiflora (Rubiaceae) of cattle in Rondonia

Abstract in English:

The toxicity of Palicourea grandiflora (H.B.K.) Standl. for cattle was demonstrated experimentally. The fresh leaves of P. grandiflora were given orally to ten bovines in amounts that varied from 0.5 to 12.2 g of the plant per kg of body weight. One to 2 g/kg ofthe plant were necessary to cause death. The first symptoms of poisoning were observed between 6 and 25 hours after the administration of the plant material. ln the three experiments in which large amounts (5 g/kg and more) of the fresh plant were given, two animals were found dead in the moming and the third died after being disturbed. Three additional animals which received between 1.0 and 2.5 g/kg of the fresh plant also died; apparently healthy for approximately 16 to 25 hours, death was caused or precipitated by only 2 to 10 minutes of exercise. The symptoms of poisoning were: an initial period during which the animal refused to move; lying down or falling to the ground in an upright position, then onto its side; opistotonus, peddling movements with the legs, bellowing; and death. The clinical symptoms, after the animal fell down, lasted from 3 to 8 minutes. Post-mortem exarninations were negative. Histopathological exarninations showed the most characteristic lesion to be hydropic vacuolar degeneration of the epithelial lining of the distal convoluted kidney tubules, seen in four of the six animals which died after ingestion of the fresh plant. It was concluded that this shrub is a cause of "sudden death" in cattle observed in Rondônia. Four months after harvest, the air-dried plant kept at room temperature retained one fourth, and after 6 months, one eighth of the toxicity of fresh material. The effect of the plant was cumulative when given in daily doses corresponding to one fifth and one tenth of the lethal dose, but not when given in smaller doses.

Através de experimentos em bovinos foi demonstrada a toxidez de Palicourea grandiflora (H.B.K.) Standl., da família Rubiaceae. As folhas recém-coletadas de P. grandiflora foram administradas por via oral a dez bovinos em quantidades que variaram de 0,25 a 12,2 g/kg de peso dos animais. A dose letal foi de 1 a 2 g/kg. Os primeiros sintomas de intoxicação foram observados a partir de 6 horas 3 7 min. até 24 horas 45 min. após a ingestão da planta. Nos três experimentos em que grandes quantidades da planta (a partir de 5 g/kg) foram administradas, dois bovinos amanheceram mortos sem terem sido tocados e o terceiro morreu após ter sido tocado somente para levantar-se. Nos três outros experimentos que também terminaram com a morte dos bovinos, os animais aparentemente estavam sadios quando, 16 horas 30 min. a 24 horas 30 min. após a ingestão da planta, foram movimentados, e sua morte foi provocada ou precipitada pelo exercício de 2 a 10 minutos. Os sintomas da intoxicação consistiram em que o animal, após um período em que simplesmente não queria ou não podia mais andar, se deitava, ou caía, inicialmente em decúbito estemal, e logo após em decúbito lateral; o animal então apresentava espasmo opistotônico, fazia alguns movimentos de pedalagem, berrava algumas vezes, e morria. A evolução do quadro clínico, desde a queda do animal, foi superaguda, variando entre 3 a 8 minutos. Os achados de necropsia foram negativos. Os exames histopatológicos revelaram como lesão mais característica degeneração hidrópico-vacuolar das células epiteliais dos túbulos uriníferos contornados distais em 4 dos 6 bovinos que morreram em virtude da ingestão da planta. Concluiu-se que este arbusto é causa de "mortes súbitas" em bovinos no Território de Rondônia. A planta dessecada retinha, após 4 meses de conservação em temperatura de ambiente, um quarto e, após 6 meses, um oitavo da toxicidade da planta fresca recém-coletada. A planta possui poder acumulativo quando dada em doses diárias de 1/5 e 1/10 da dose letal, porém não em doses menores.


#702 - Studies on the congenital transmission of avian lymphoid leukosis virus

Abstract in English:

White Leghorn hens from two commercial lines were identified as shedders or non-shedders of the lymphoid leukosis virus (LL) using the micro-complement fixation test. This test detected the group-specific antigen (gs-ag) of the leukosis/sarcoma group of viruses when used directly on the albumen of unincubated fresh eggs. It was found that 21.4% of line A hens and 17.2% of line B hens eliminated gs-ag into their eggs. Sequential studies of the elimination of the gs-ag, demonstrated that infected hens can be either intermittent or consistent shedders of this antigen in their eggs. Electron microscopy studies demonstrated the presence of type-C viral particles in the pâncreas and in the magnum of the oviduct of hens that had consistently shed the gs-ag in the egg albumen. The indirect immunofluorescence test detected gs-ag in the cytoplasm of peripheral blood leucocytes and was used to detect infected roosters that were utilized as semen donors. Artificial insemination of non-shedder hens with semen obtained from infected rooters resulted in the shedding of gs-ag into the eggs of one of the hens. Moreover, there was seroconversion in 11 of the 26 hens from both lines, inseminated with semen from the infected roosters. In was concluded that LL virus infection could be introduced, through insemination, to non-infected hens using sêmen from infected roosters.

A microprova de fixação do complemento permitiu a identificação de galinhas Leghorn brancas das linhagens A e B infectadas com o vírus da leucose linfóide aviária (LL). Esta prova detectou o antígeno específico (ag-gs) do vírus do grupo leucose/sarcoma aviário quando foi utilizada diretamente na albumina de ovos frescos não incubados. Encontrou-se que 21,4% das galinhas da linhagem A e 17,2% das galinhas da linhagem B do plantel geral eliminavam ag-gs nos seus ovos. Foi demonstrado também que as galinhas infectadas podem eliminar ag-gs nos ovos, constantemente ou intermitentemente. Estudos realizados com o microscópio eletrônico demonstraram a presença de partículas virais do tipo C no pâncreas e na porção do magnum do oviduto de galinhas que tinham eliminado ag-gs do vírus da LL cons-tantemente na albumina dos ovos. A técnica de imunofluorescência indireta detectou ag-gs no citoplasma de leucócitos de aves portadoras e foi utilizada para identificar galos infectados a serem usados como doadores de sêmen. A inseminação de galinhas não eliminadoras de ag-gs do vírus da LL com sêmen de galos portadores resultou na eliminação de ag-gs nos ovos de uma das galinhas inseminadas. Mais ainda, houve uma conversão sorológica, em 11 das 26 galinhas das linhagens A e B após serem inseminadas com sêmen de galos portadores, demonstrando-se assim a participação do galo como introdutor da infecção via sêmen.
