Prof. Dr. Orlando Marques de Paiva 87, Cidade Universitária, São Paulo, SP 05508-900, Brazil. E-mail: ceambrosio@usp.br Thirty diaphragms of sheep of Santa Inês breed were studied regarding their origin, division and arrangement of the right and left phrenic nerves (Fde), and the participation of other nerves in the innervation of the diaphragm. By fixing and dissecting pieces, it was found that phrenic nerves (F) frequently come from the ventral branches of the 5th (C5) and 6th (C6) cervical spinal nerves (Ec), at right (46.67%) and at left (43.33%). The F often form a lumbocostal trunk, sternal branches at right (40.00%) and lumbar, costal and esternal branches at left (36.68%). The lumbar branches of F innervate frequently at left (96.67%) the homolateral pillar of the diaphragma, and at right (50.00%) they give fillets to Vena cava caudalis. The costal branches of the F innervate at left (90.00%) and at right (76.66%) the dorsal and ventral regions of the pars costalis. The sternal branches of the F innervate at right (100.00%) and at left (83.33%) the pars sternalis and the ventral region of the pars costalis at the same side. The intercostal nerves (VII to XII pairs, 63.33%) contribute to innervate the diaphragm of Santa Inês sheep.

#282 - Comparison of two commercial kits and two extraction methods for fecal glucocorticoid analysis in ocelots (Leopardus pardalis) submitted to ACTH challenge, p.329-334

Abstract in English:

ABSTRACT.- Dias E.A., Nichi M. & Guimarães M.A.B.V. 2008. Comparison of two commercial kits and two extraction methods for fecal glucocorticoid analysis in ocelots (Leopardus pardalis) submitted to ACTH challenge. Pesquisa Veterinária Brasileira 28(7):329-334. Departamento de Reprodução Animal, Faculdade de Medicina Veterinária e Zootecnia, Universidade de São Paulo, Av. Prof. Dr. Orlando Marques de Paiva 87, Cidade Universitária, São Paulo, SP 05508-270, Brazil. E-mail: eantdias@usp.br The ocelot (Leopardus pardalis) is included in list of wild felid species protected by CITES and is part of conservation strategies that necessarily involve the use of assisted reproduction techniques, which requires practical and minimally invasive techniques of high reproducibility that permit the study of animal reproductive physiology. The objective of this study was to compare and validate two commercial assays: ImmuChem Double Antibody Corticosterone 125I RIA from ICN Biomedicals, Costa Mesa, CA, USA; and Coat-a-Count Cortisol 125I RIA from DPC, Los Angeles, CA, USA, for assessment of fecal glucocorticoid metabolites in ocelots submitted to ACTH (adrenocorticotropic hormone) challenge. Fecal samples were collected from five ocelots kept at the Brazilian Center of Neotropical Felines, Associação Mata Ciliar, São Paulo, Brazil, and one of the animals was chosen as a negative control. The experiment was conducted over a period of 9 days. On day 0, a total dose of 100 IU ACTH was administered intramuscularly. Immediately after collection the samples were stored at -20C in labeled plastic bags. The hormone metabolites were subsequently extracted and assayed using the two commercial kits. Previously it was performed a trial with the DPC kit to check the best extraction method for hormones metabolites. Data were analyzed with the SAS program for Windows V8 and reported as means ± SEM. The Schwarzenberger extraction method was slightly better when compared with the Wasser extraction method (103,334.56 ± 19,010.37ng/g of wet feces and 59,223.61 ± 12,725.36ng/g of wet feces respectively; P=0,0657). The ICN kit detected an increase in glucocorticoid metabolite concentrations in a more reliable manner. Metabolite concentrations (ng/g wet feces) on day 0 and day 1 were 66,956.28 ± 36,786.93 and 92,991.19 ± 28,555.63 for the DPC kit, and 205,483.32 ± 83,811.32 and 814,578.75 ± 292,150.47 for the ICN kit, respectively. The limit of detection for the ICN kit was 7.7 ng/mL for 100% B/Bo (25ng/mL for 88%B/Bo) and for the DPC kit it was 0.2ug/dL for 90.95% B/Bo (1ug/dL for 81.27% B/Bo). In conclusion it was confirmed that the Schwarzenberger extraction method and the ICN kit are superior for extracting and measuring fecal glucocorticoid metabolites in ocelot fecal samples.

Departamento de Epidemiologia Experimental Aplicada às Zoonoses, Faculdade de Medicina Veterinária e Zootecnia, Universidade de São Paulo, Prof. Dr. Orlando de Marques de Paiva 87, Cidade Universitária, São Paulo, SP 05508-270, Brazil. E-mail: animalexotico@terra.com.br The usual surgical technique for accessing the coelomic cavity of chelonians through the horny shields and bone plates of plastron by using a Dremel machine (plastron osteotomy) is considered slow, traumatic, and very painful for the patient. In this paper a technique of coelioscopic-assisted prefemoral oophorectomy is described in red-eared slider. The time spent on accessing the coelomic cavity, removal of the follicles and ovaries, and anesthetic recovery was markedly decreased (three hours less) in comparison with the traditional technique. The rapid return of the patient to normal activity demonstrates an important reduction in pain at post-surgical recovery. The extra cost resulting of the use of endoscopy and a specialized veterinarian service is clearly justified by the positive results described above.

#285 - Celiac artery in New Zealand rabbit: Anatomical study of its origin and arrangement for experimental research and surgical practice, p.237-240

Abstract in English:

ABSTRACT.- Abidu-Figueiredo M., Xavier-Silva B., Cardinot T.M., Babinski M.A. & Chagas M.A. 2008. Celiac artery in New Zealand rabbit: Anatomical study of its origin and arrangement for experimental research and surgical practice. Pesquisa Veterinária Brasileira 28(5):237-240. Departamento de Anatomia Animal, Instituto de Biologia, Universidade Federal Rural do Rio de Janeiro, Seropédica, RJ 23890-000, Brazil. E-mail: marceloabidu@gmail.com Rabbits have been used as an experimental model in many diseases and for the study of toxicology, pharmacology and surgery in many universities. However, some aspects of their macro anatomy need a more detailed description, especially the abdominal and pelvic arterial vascular system, which has a huge variability in distribution and trajectory. Thirty cadaveric adult New Zealand rabbits, 13 male and 17 female, with an average weight and rostrum-sacral length of 2.5 kg and 40cm, respectively, were used. The thoracic aorta was cannulated and the vascular system was filled with stained latex S-65. The celiac artery and its proximal branches were dissected and lengthened in order to evidence origin and proximal ramifications. The celiac artery emerged between the 12th and 13th thoracic vertebra in 11 (36.7%) rabbits; at the level of the 13th thoracic vertebra in 6 (20%) rabbits; between the 13th thoracic vertebra and the 1st lumbar vertebra in 12 (40%) rabbits; and at the level of the 1st lumbar vertebra in only one (3.3%) rabbit. The mean length of the celiac artery was 0.5cm. The celiac artery first branch was the lienal artery, the second branch was the left gastric artery and the hepatic artery arose from the left gastric artery in all the dissected rabbits. No relation was observed between the celiac artery length and the rostrum-sacral length in rabbits. The number of left gastric and lienal artery branches and the distribution of celiac artery origin are not gender dependent.

Pesquisa Veterinária Brasileira 28(5):249-252. Departamento de Morfofisiologia Veterinária, Centro de Ciências Agrárias, Universidade Federal do Piauí, Teresina, PI 64049-550, Brazil. E-mail: carvalhomam@uol.com.br Twenty pairs of agouti (Dasyprocta prymnolopha Wagler, 1831) kidneys were studied to describe the arterial anatomical-surgical segments. The renal arteries were injected with stained acetate vinyl, followed by procedures of acid corrosion in order to obtain vascular casts. It was found that the renal artery is always single and bifurcated into ventral and dorsal sectorial arteries. The sectorial arteries reached the kidneys (100% of the cases) through the hilus. These vessels gave origin to segmental branches responsible for kidney irrigation. At the right kidney, the ventral sectorial arteries gave origin to 3 (60% of the cases), 4 (35%) and 5 (5%) segmental branches; the dorsal sectorial arteries gave origin to 3 (30%), 4 (45%), 5 (20%) and 6 (5%) segmental arteries separated by a vascular sector. At the left kidney, the ventral sectorial arteries originated 2 (10%), 3 (55%) or 4 (35%) segmental branches; the dorsal sectorial arteries gave origin to 3 (25%), 4 (50%) and 5 (25%) segmental branches. Based on the arterial distribution of agouti kidneys, independent sections and arterial segments were found, so that it is possible to accomplish partial kidney resection surgery.

Universidade de São Paulo, Cidade Universitária, Av. Prof. Dr. Orlando Marques de Paiva 87, São Paulo, SP 05508-900, Brazil. E-mail: ceambrosio@usp.br The study aimed to characterize gross and microscopic features of the yolk sac in paca (Agouti paca) and agouti (Dasyprocta aguti) in early gestation. Fragments of the yolk sac of 3 paca and 3 agouti fetuses at early gestation were taken and processed for histological and ultrastructural analyses. Gross features of the vitelline placenta in both species showed its insertion over the main placenta surface and projections to the embryos/fetuses. Microscopically, the vitelline placenta was constituted by endoderm epithelium and mesenchyme, in which vitelline vessels are abundant. The ultrastructure of the samples showed that the visceral yolk sac of the paca was formed by endodermic cells with nuclei in the median region, and that the visceral yolk sac of the agouti was formed by nuclei arranged apically; other characteristic was the large number of mitochondrias, eletrodense vesicles with microvilosities We conclude that (1) the vitelline placenta of the two species presents insertion in the surface of the main placenta; (2) the vitelline placenta of paca rests on the Reichert’s membrane, whereas the agouti vitelline placenta does not have this membrane; (3) the chorion and allantoic are fusioned; and (4) the chorioallantoic placenta and the yolk sac in both species are reversed and vascularized.

E-mail: watanabe@icb.usp.br The three-dimensional characteristics of the intracellular components of acinar and ductal cells were revealed using the osmium-DMSO-osmium method. The samples were macerated in diluted osmium after fractured in DMSO solution. The stacks of the rough endoplasmic reticulum are revealed intermingling by several mitochondria. The lamellae of the rough endoplasmic reticulum are located around the nuclei at basal portion and these structures are shown in three-dimensional HRSEM images.

Prof. Dr. Orlando Marques de Paiva 87, São Paulo, SP 05508-270, Brazil. E-mail: juvet@usp.br Herbst corpuscles of the palatine mucosa of ostrich were studied by light microscopy. The corpuscles are composed of an outer core, inner core and central nerve terminal. The outer core presents numerous lamellae, while the inner core shows compact structure of cytoplasm sheets. The corpuscles are elongate or oval in shape and are surrounded by bundles of collagen fibers. Each lamella is composed of a dense network of thick fibrils. The terminal axons are located along the axis and form a bulb terminal. The fibers of external core stained by Picrosirius and examined by polarized light microscopy revealed to be green in color like type I collagen fibers, and at the periphery is a large amount of collagen type III. The corpuscles are surrounded by flat cells and dense collagen fibers at the periphery.

ABSTRACT.- Orsi A.M., Domeniconi R.F., Simões K., Stefanini M.A. & Baraldi-Artoni S.M. 2007. [Seasonal variability in epididymal duct of the domestic quail: morphologic features.] Variabilidade sazonal no ducto epididimário de codorna doméstica: observações morfológicas. Pesquisa Veterinária Brasileira 27(12):495-500. Departamento de Anatomia, Universidade Estadual Paulista, Cx. Postal 510, Botucatu, SP 18618-000, Brazil. E-mail: amorsi@ibb.unesp.br Small but expressive variability was noted on the epididymidis duct (ED) of domestic quail along the year, with more evidence in autumn of the quiescent phase of the annual testis cycle in this species. Spring features of ED had a general similar pattern in summer and winter. They were characterized by enlargement of epididymis tubule, storage of spermatozoa into the luminal compartment and presence of mitochondria, ER lamellae, several variable vesicles, and lysosomes localized mainly on the apical cytoplasm of principal cells (P) of the epididymal epithelium. These P cells features indicated a process of endocytosis and perhaps protein secretion. Autumn quiescence was marked by a convolute pattern of the epididymis tubule, lacking of spermatozoa and small amount of exfoliate heterogeneous material inside the luminal compartment at light microscopy. Ultrastructural degenerative features mainly apical cytoplasmic debris were seen in the supranuclear cytoplasm of lining P cells.
